Available in PDF, EPUB and Kindle. Book excerpt: This book focuses on the context dependency of cell signaling by showing how the endosomal system helps to structure and regulate signaling pathways. The location and concentration of signaling nodes regulate their activation cycles and engagement with distinct effector pathways. Whilst many cell signaling pathways are initiated from the cell surface, endocytosis provides an opportunity for modulating signaling networks’ output. In this book, first a series of reviews describe the endocytic and endosomal system and show how these subcellular platforms sort and regulate a wide range of signaling pathway components and phenotypic outputs. The book then reviews the latest scientific insights into how endocytic trafficking and subcellular location modulate a set of major pathways that are essential to normal cellular function and organisms’ development.

Book excerpt: Endosomes are membrane-bound compartments that transport internalized material from the plasma membrane to the lysosome and elsewhere. These compartments, often about 500 nm, but ranging in size, have the capability to sort molecules, routing some contents to the lysosomes for degradation, and recycling other materials back to the plasma membrane. The Golgi apparatus also provides molecules to the endosome, some of which are delivered to lysosomes and others are recycled back to the Golgi. Because of this ability to differentially deliver molecules, the endosome is viewed as a presorting structure. Endosomes are categorized by size, enzymatic content, morphology, and by other criteria such as the length of time it takes internalized material to reach them. Endosomes may provide platforms for cross talk between signaling systems, and this consideration has provided them elite status among cellular components that contribute to signaling. This volume provides descriptions of the range of methods used to analyze and evaluate these important compartments. The authors explain how these methods are able to provide important biological insights in the context of particular models

Book excerpt: This book reviews knowledge on the interconnection of signal transduction and endocytosis/intracellular trafficking. The chapters cover knowledge obtained by using different model systems. The first chapter deals with Receptor Tyrosin Kinases (RTKs) with emphasis on the Epidermal Growth Factor Receptor (EGF receptor) and the Platelet Derived Growth Factor Receptor (PDGF receptor). The second chapter deals with the RTK c-Met and with how this RTK becomes carcinogenic. The third chapter reviews recent understanding on the mechanisms of action of the numerous fibroblast growth factors and their receptors. In the fourth chapter we learn about the trafficking of and signalling from the Growth Hormone Receptor and how this receptor is controlled by ubiquitination. The fifth chapter is devoted to the Interleukin II receptor, essential for activation of T cells. Links between ubiquitination, signalling, endocytosis, and sorting are reviewed. The last chapter discusses current views on how monoubiquitination controls both signalling and trafficking and thereby the final outcome of receptor activation.

Available in PDF, EPUB and Kindle. Book excerpt: Endocytosis is a fundamental cellular process by means of which cells internalize extracellular and plasma membrane cargos for recycling or degradation. It is important for the establishment and maintenance of cell polarity, subcellular signaling and uptake of nutrients into specialized cells, but also for plant cell interactions with pathogenic and symbiotic microbes. Endocytosis starts by vesicle formation at the plasma membrane and progresses through early and late endosomal compartments. In these endosomes cargo is sorted and it is either recycled back to the plasma membrane, or degraded in the lytic vacuole. This book presents an overview of our current knowledge of endocytosis in plants with a main focus on the key molecules undergoing and regulating endocytosis. It also provides up to date methodological approaches as well as principles of protein, structural lipid, sugar and microbe internalization in plant cells. The individual chapters describe clathrin-mediated and fluid-phase endocytosis, as well as flotillin-mediated endocytosis and internalization of microbes. The book was written for a broad spectrum of readers including students, teachers and researchers.

Book excerpt: Endosomes are a heterogeneous population of endocytic vesicles and tubules that have captivated the interest of biologists for many years, partly due to their important cellular functions and partly due to their intriguing nature and dynamics. Endosomes represent a fascinating interconnected network of thousands of vesicles that transport various cargoes, mainly proteins and lipids, to distant cellular destinations. How endosomes function, what co-ordinates the molecular determinants at each step of their dynamic life cycle and what their biological and medical relevance is, are among the questions addressed in this book.

Available in PDF, EPUB and Kindle. Book excerpt: This book describes the Notch signaling pathway with a focus on molecular mechanisms. The Notch signaling pathway is a seemingly simple pathway that does not involve any second messenger. Upon ligand binding two consecutive proteolytic cleavages of the NOTCH receptor release the Notch intracellular domain from the membrane. The Notch intracellular domain migrates into the nucleus and activates gene expression. Recently, new technologies allowed us to better understand this pivotal signaling cascade and revealed new regulatory mechanisms. The different chapters cover many aspects of the Notch signaling focusing on the mechanisms governing the receptor/ligand interaction as well as on the downstream intracellular signaling events. Aspects of both canonical and non-canonical signaling are discussed and the function of Notch signaling in physiological and pathological contexts are elucidated. This book is not only intended for experts but it should also be a useful resource for young, sprouting scientists or interested scientists from other research areas, who may use this book as a stimulating starting point for further discoveries and developments.
