There is an introduction to electrochemistry followed by considerations to make when studying electron transfer at the nanoscale using voltammetric and amperometric measurements. Chapter 2 discusses three types of new nanoelectrodes that have been developed. These nanoelectrodes are different from previous nanoelectrodes in their materials and fabrication methods. Chapter 3 introduces the reader to electrode arrays and their properties. A new way of fabricating microelectrode arrays with high density and a nearly 100% success rate in terms of electrode activity is presented. In Chapter 4 a new electrochemical imaging technique developed in our lab is introduced. This new technique requires bipolar electrode arrays, new variants of which are fabricated using several methods which are discussed. Chapter 5 moves away from Faradaic current measurements toward ionic current measurements of nanoparticles. Emulsified oil droplets are monitored with ionic current measurements and fluorescence imaging to reveal their interactions at a nanopore. In Chapter 6 a new method for fabricating nanopores is outlined along with substantial experiments and finite element simulations to describe their properties. The ability of this new nanopore to measure nanoparticles is illustrated through resistive-pulse sensing. These new nanopores are compared with another type of nanopore using both experiment and simulation.

Book excerpt: This dissertation presents the construction and application of micro and nanoscale electrodes for electroanalytical analysis. The studies presented herein encompass two main areas: electrochemical catalysis, and studies of the dynamics of single cell exocytosis. The first portion of this dissertation engages the use of Pt nanoelectrodes to study the stability and electrocatalytic properties of materials. A single nanoparticle electrode (SNPE) was fabricated by immobilizing a single Au nanoparticle on a Pt disk nanoelectrode via an amine-terminated silane cross linker. In this manner we were able to effectively study the electrochemistry and electrocatalytic activity of single Au nanoparticles and found that the electrocatalytic activity is dependent on nanoparticle size. This study can further the understanding of the structure-function relationship in nanoparticle based electrocatalysis. Further work was conducted to probe the stability of Pt nanoelectrodes under conditions of potential cycling. Pt based catalysts are known to deteriorate under such conditions due to losses in electrochemical surface area and Pt dissolution. By using Pt disk nanoelectrodes we were able to study Pt dissolution via steady-state voltammetry. We observed an enhanced dissolution rate and higher charge density on nanoelectrodes than that previously found on macro scale electrodes. The goal of the second portion of this dissertation is to develop new analytical methods to study the dynamics of exocytosis from single cells. The secretion of neurotransmitters plays a key role in neuronal communication, and our studies highlight how bipolar electrochemistry can be employed to enhance detection of neurotransmitters from single cells. First, we developed a theory to quantitatively characterize the voltammetric behavior of bipolar carbon fiber microelectrodes and secondly applied those principles to single cell detection. We showed that by simply adding an additional redox mediator to the back-fill solution of a carbon fiber microelectrode, there is a significant enhancement in detection. Additionally we used solid state nanopores to detect individual phospholipid vesicles in solution. Vesicles are key cellular components that play essential biological roles especially in neurotransmission. This work represents preliminary studies in detection and size determination from vesicles isolated from individual cells.

In this sense, "New electrochemical strategies for synthesising micro- and nano-structures" proposes, generalizes and discusses the development of novel electrochemical methods, strategies or media to prepare different kinds of micro- and nanostructured materials. The leitmotiv could be focused on the possibility of preparing micro-patterned (Cu), nanoparticulated (Co, CoNi, CoPt), composited (CoPt-Ni), mesoporous (CoNi, CoPt, Pt, CoNi@Pt or CoNi@Au) or nanopowdered (CoPt) deposits with various shape (films, nanorods or nanowires) by using the proposed strategies (Electrochemical nano-Fabrication using Chemistry and Engineering (EnFACE) technology or classical/ionic liquid microemulsions as a soft electrodeposition templates) to obtain micro/nanomaterials for specific magnetic, catalytic or biomedical applications. The present thesis is organized in eight chapters, in which several published scientific papers in international journals with our original results have been included in order to discuss and analyze our work. The first one introduces the electrodeposition (Chapter 1) - history, theoretical fundaments, advantages and disadvantages and introduce the state-of-the-art of electrochemical micro- and nano-fabrication -, which is fundamental in order to understand our work. The aims and the experimental details of our research are presented and detailed in chapter 2 and chapter 3, respectively. It is important emphasize that "New electrochemical strategies for synthesising micro- and nano-structures" could be grouped into three main areas, according the aims introduced above: (a) The optimization of EnFACE technology as an alternative method to classical masked-electrodeposition to prepare metallic microstructures on large conductive substrates, with the final objective of proposing this technology as a scalable, economic and environmentally-friendly synthesis approach to microfabrication; this could be relevant in the electronics and sensors industries. This work is presented in chapter 4; (b) Proposing, discussing and analyzing the use of different types of classical microemulsions as new electrochemical media for synthesising nanostructures -- that is, a new shape-controlled electrodeposition approach based on the use of soft-template systems. In this sense, the possibility of obtaining different types of Co-based structures with varied geometries and shapes as a function of the microemulsions conditions has been demonstrated. However, the low or moderate deposition rates and efficiencies leading us to explore and propose the use of alternative (water-in-ionic liquid) microemulsions as soft-template electrochemical media with improved conductivity and low environmental impact for shape-controlled electrodeposition. These electrodeposition media were tested to prepare magnetic nanostructured materials (nanoparticulated CoPt magnetic films or CoPt-Ni composites) with controlled size, composition and magnetism. Based on our results, the use of water -in-ionic liquid (W/IL) microemulsions have been revealed to be a versatile, green, simple and inexpensive approach to 1) synthesizing nanoparticles with controlled size and composition, and therefore magnetic behavior, by controlling the droplet size and aqueous solution concentration respectively, or 2) developing layers of composites using a one-step procedure, in which simultaneous electrodeposition from aqueous droplets and continuous ionic liquid components could take place. This work is presented in chapter 5; (c) Proposing, generalizing and discussing new electrochemical strategies (electrodeposition in water-in-ionic liquid, bicontinuous and ionic liquid-in-water microemulsions) for synthesising mesoporous nanomaterials ((Pt, CoPt, CoNi, CoNi @Au or CoNi@Pt) for catalysis (the electrooxidation of methanol or ethanol in order to propose competitive electrocatalysts with poor- Pt content for fuel cell reactions) or drug delivery with enhanced therapeutic effect as a consequence of their magnetic stimulation. This results are presented in chapter 6. Lastly, to complete the thesis, the main conclusions obtained are collected in chapter 7, while a summary of the thesis in Catalan language is given in chapter 8.
