Book excerpt: The regulation of cholesterol homeostatsis is of considerable interest because of the host of studies that show a clear positive relationship between plasma cholesterol levels and the risk of coronary heart disease. It has become apparent that plasma cholesterol levels are affected by a number of life style factors including the amount and type of fat in the diet. However, the mechanisms whereby dietary fat affects plasma cholesterol remain unclear. In this study we have investigated the effects of the levels and types of dietary fat on the cholesterol homeostasis. We used the FIB strain of hamster as a model as this strain has been shown to be sensitive to dietary fat induced atherosclerosis. We propose that this sensitivity reflects differences in the regulation of lipid metabolism between the F1B and the parent strain of hamsters. -- Cholesterol homeostasis in the liver is tightly regulated by several sterol-sensitive regulatory proteins and receptors, including LDL receptor, HMG-CoA reductase, ACAT, CYP7, and SREBPs. It is postulated that the ER cholesterol is the cholesterol regulatory pool plays a major role in the regulation of these enzymes of the whole cell. -- We examined the effect of different dietary fats on the activity or expression of these proteins and observed that the activities of these enzymes are not directly regulated by the fats and cholesterol in the diet; instead, they appear more responsive to changes in the lipid environment of the microsome in the F1B hamster liver. Both the HMG-CoA reductase and the ACAT activity are positively correlated with the level of n-3 fatty acids in the microsome, and ACAT activity also depend on the content of microsomal cholesterol. -- We suggested that the levels of n-3 fatty acids and cholesterol in the diets may change the lipid composition microsomal membranes. This might alter membrane fluidity or the distribution of the key enzymes and regulatory proteins in the membrane of the ER. We suggest that the formation of microdomains (rafts) may sequester much of the cholesterol into pools that are separate from the cholesterol regulatory pool in the ER. However, further studies on the lipid composition of these microdomains are needed for clarify our hypothesis.

This book was released on 2004 with total page 394 pages. Available in PDF, EPUB and Kindle. Book excerpt: Cardiovascular disease (CVD) is the leading cause of death and disability in the United States. Its genesis can be attributed, in part, to dietary and lifestyle patterns. Fat in the diet is comprised of different classes of fatty acids: saturated (SFA), monounsaturated (MUFA), polyunsaturated (PUFA), trans, each having characteristic effects on both high density lipoprotein (HDL) and non-HDL [(nHDL) total - HDL cholesterol] cholesterol concentrations. Investigated first was the time course and response of Charles River (CR) and Bio Breeders (F1B) Golden-Syrian hamsters to dietary SFA (butter) on serum lipoproteins and aortic cholesteryl ester in. After 12 weeks, CR hamsters had significantly lower serum total and nHDL cholesterol concentrations, but higher aortic cholesteryl ester deposition than the F1B hamsters (P
Book Synopsis Similar Effects of Dietary Oleic and Palmitic Acid on Plasma Lipids and in Vivo Lipoprotein Metabolism in Normocholesterolemic Hamsters Fed Purified Diets with Low Cholesterol ( 40 Mg/1000 Kcal) But Different Quantities of Fat by : Pramod Khosla

This book was released on 1996 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: Dietary myristic (0 - 0.50% en) and linoleic acid (5.2 5.9% en) were relatively constant across all diets, which contained a background level of cholesterol ( 40 mg/1000 kcal), and were formulated so than protein, vitamins and minerals were constant per calorie. All animals were fed a fixed amount of calories for 6-8 week periods. Plasma lipids (n=15-20 per group) and lipoprotein cholesterol concentrations were determined following sequential ultracentrifugation, from individual animals (n=5-6 per group).

This book was released on 2007 with total page 230 pages. Available in PDF, EPUB and Kindle. Book excerpt: In study 1 the main changes were seen in the lipoprotein subfraction distribution. Carbohydrate restricted diet (CRD) decreased the levels of the smaller LDL particles (P < 0.001). In study 2 the CRD lowered the levels of hepatic total and esterified cholesterol, as well as ACAT activity compared to HChol (P < 0.05). In contrast dietary cholesterol downregulated HMG-CoA reductase mRNA levels (P < 0.05).
