Book excerpt: Three experiments were carried out in calves to investigate the effect of changes in dietary PUFA supply on; the fatty acid composition of primary lymphoid organs and tissues, ex-vivo lymphocyte proliferation response, eosinophil and mucosal mast cell numbers in the gut and on immune response to nematode parasites Ostertagia ostertagi and Cooperia oncophora infection. In experiment 1, two groups of Jersey bull calves were fed on 25 g/day of either an n-3 enriched fish oil (FO) (n-3 group) or a binary mixture of palm/rapeseed oil (normal group) for a period of 21 days (n=10) or 42 days (n=12). In experiment 2, calves (n=40) were allocated at birth to 7 treatment groups, which received for 21 days oils supplements similar to experiment 1 in either whole milk or milk replacer. While in experiment 3, calves (n=24) were allocated to two dietary treatment groups (n=12) and fed oil supplements similar to experiment 1. Within each dietary treatment group, calves (n=8) were infected for 8 weeks (three times/week) with 2000 stage 3 larvae of Ostertagia ostertagi and Cooperia oncophora. The remaining calves (n=4) were uninfected controls. In experiments 1 and 3 calves were fed on 500 g/day of a standard milk replacer hay and calf weaner pellets (1.5 X maintenance of metabolizable protein (MP) and metabolizable energy (ME) requirements). 30-mg a-tocopherol acetate/kg PUFA was fed all the calves. In exp. 1 lymphocytes, plasma and tissue samples were collected for fatty acid analysis. In exp. 2 tissue samples were collected for fatty acid analysis, lymphocyte stimulation responses and determination of mucosal mast cells and eosinophils numbers. While in exp. 3 faeces samples were collected twice per week for faecal egg counts (FEC) and blood for plasma collection was sampled every week. At slaughter, tissue sample for fatty acid analysis were collected, while abomasal, duodenum and terminal ileum samples were collected for MMC and eosinophil counts. The gut contents from abomasum and intestine were washed and collected for worm counts. Book excerpt: This dissertation includes studies on the effect of dietary CLA on the growth, fat accumulation and fatty acid status of chicken, and chicken meat quality as influenced by irradiation. Results showed that dietary CLA did not have significant effects on the growth rate and feed efficiency in chicken. And high levels of dietary CLA slightly reduced the whole body fat content. High ratio of dietary CLA can incorporate into chicken meat and egg yolk. Dietary CLA reduced the concentration of monounsaturated fatty acids. The concentration of polyunsaturated fatty acids, including arachidonic acid, linolenic acid and linolenic acid, also reduced as the dietary CLA level increased. However, when the dietary level of linolenic acid in diet was high, dietary CLA stimulated the synthesis of DHA and EPA, which might directly relate to the biological effects of CLA. High level of dietary CLA influenced the quality of meat, which was slightly harder and drier compared to the control meat. Dietary CLA significantly improved the oxidative stability of chicken meat. The reason for the improved oxidative and color stability of meat patties during storage should be due to the reduced unsaturated fatty acid content in chicken muscles, which improved lipid and color stability and reduced volatile production in both irradiated and nonirradiated meat during storage. Irradiation greatly increased the volatile production and induced a metal-like off-odor in chicken rolls, and dietary CLA had synergistic effect on this metal-like off-odor. Irradiation also increased the redness of chicken rolls. Consumers had a preference for the color of irradiated chicken rolls, while their reactions to the flavor of irradiated chicken rolls were quite negative.

Book excerpt: This study was conducted to test the hypothesis that enriching developing adipose tissue in long chain omega-3 long chain polyunsaturated fatty acids (n-3 LCPUFA) reduces adiposity compared to enrichment in n-6 LCUFA. Sixty Cobb 500 broiler breeders were fed a diet containing either 5% fish oil (FO) or corn oil (CO). After four weeks, fertilized eggs were collected and hatched, and all chicks were fed the same corn oil-based broiler starter diet. At days 7 and 14 post-hatch, chicks in each group were weighed, euthanized and blood, liver, breast, and subcutaneous and abdominal fat samples harvested. Gas or liquid chromatography and mass spectrometry were used to determine the effects of diet on fatty acid profiles of tissue triglyceride, free fatty acid, phospholipid pools and protein expression. RNA sequencing and PCR was used to determine the effect on gene expression. Mixed model ANOVA and least square means were used to identify effects of maternal diet and age*diet interactions on chick phenotypes (SAS v9.4, Cary NC). Differentially expressed genes were determined using the DESeq2 R package. Hen dietary fat source did not significantly affect performance. Flax seed is a rich source of [alpha]-linolenic acid and can be used to increase n-3 FA in poultry meat. Previous studies have shown that flax seed in combination with carbohydrase enzymes in broiler diets enhances long chain (LC) n-3 FA in hepatic tissue. Three experiments were conducted to evaluate the effect of flax seed and carbohydrase enzyme addition on production performance, total tract lipid digestibility, hepatic fatty acid composition, liver lipid class distribution and lipid metabolism in broiler chickens. In Experiment 1, broiler chicks were placed in individual cages and fed isocaloric, isonitrogenous corn-soy diets: Control, Flax (10% flax), and Flax+E (Flax+0.05% enzyme). The diets were fed for a period of 42 days. Total tract digestibility of lipids and FA were assessed from day 19 to 21 of growth.
