Available in PDF, EPUB and Kindle. Book excerpt: Genetics and Biotechnology of Bacilli, Volume 2 is a collection of papers from the "Fourth International Conference on Bacilli" held in California on June 21-24, 1987. One paper reviews the results of cloning and characterization of genes for secreted enzyme and of genes that control the expression of secreted enzymes in relation with other prokaryotic regulatory systems. Other papers tackle the regulation of gene expression during sporulation, the sigma factors, bacterial toxins, and antibiotic resistance genes. One paper reports that three genes responsible for the code for peptides found in BT cuboidal crystals have been successfully cloned. The three codes have different toxic characteristics in relation to tobacco hornworm larvae or mosquito larvae. Other papers examine replication. Such examination pertains to the two levels of control on the chromosome involved in DNA replication, or to the possible functional importance that several membranes associated DNA subcomplexes can have in Bacillus subtilis, where one of these appear to control initiation. Other papers discus secretion and extracellular enzymes, as well as, the different genetic systems and methods. This collection can prove beneficial for biochemists, micro-biologists, cellular researchers, and academicians involved in the study of cellular biology, microchemistry, or toxicology.

Available in PDF, EPUB and Kindle. Book excerpt: This work describes the current knowledge of biochemical mechanisms regulating initiation of DNA replication in Escherichia coli, which focuses on the control of activity of the DnaA protein. Examples of direct linkages between DNA replication and other cellular processes are provided. In addition, similarities of the mechanisms of regulation of DNA replication operating in prokaryotic and eukaryotic cells are identified, and implications for understanding more complex processes, like carcinogenesis are suggested. Studies of recent years provided evidence that regulation of DNA replication in bacteria is more complex than previously anticipated. Multiple layers of control seem to ensure coordination of this process with the increase of cellular mass and the division cycle. Metabolic processes and membrane composition may serve as points where integration of genome replication with growth conditions occurs. It is also likely that coupling of DNA synthesis with cellular metabolism may involve interactions of replication proteins with other macromolecular complexes, responsible for various cellular processes. Thus, the exact set of factors participating in triggering the replication initiation may differ depending on growth conditions. Therefore, understanding the regulation of DNA duplication requires placing this process in the context of the current knowledge on bacterial metabolism, as well as cellular and chromosomal structure. Moreover, in both Escherichia coli and eukaryotic cells, replication initiator proteins were shown to play other roles in addition to driving the assembly of replication complexes, which constitutes another, yet not sufficiently understood, layer of coordinating DNA replication with the cell cycle.

Book excerpt: This second of two volumes on Plant Genome Diversity provides, in 20 chapters, insights into the structural evolution of plant genomes with all its variations. Starting with an outline of plant phylogeny and its reconstruction, the second part of the volume describes the architecture and dynamics of the plant cell nucleus, the third examines the evolution and diversity of the karyotype in various lineages, including angiosperms, gymnosperms and monilophytes. The fourth part presents the mechanisms of polyploidization and its biological consequences and significance for land plant evolution. The fifth part deals with genome size evolution and its biological significance. Together with Volume I, this comprehensive book on the plant genome is intended for students and professionals in all fields of plant science, offering as it does a convenient entry into a burgeoning literature in a fast-moving field.

Available in PDF, EPUB and Kindle. Book excerpt: Intracellular checkpoint controls constitute a network of signal transd- tion pathways that protect cells from external stresses and internal errors. Ext- nal stresses can be generated by the continuous assault of DNA-damaging agents, such as environmental mutagens, ultraviolet (UV) light, ionizing radiation, or the reactive oxygen species that can arise during normal cellular metabolism. In response to any of these assaults on the integrity of the genome, the activation of the network of checkpoint control pathways can lead to diverse cellular responses, such as cell cycle arrest, DNA repair, or elimination of the cell by cell death (apoptosis) if the damage cannot be repaired. Moreover, internal errors can occur during the highly orchestrated replication of the cellular genome and its distribution into daughter cells. Here, the temporal order of these cell cycle events must be strictly enforced—for example, to ensure that DNA replication is c- plete and occurs only once before cell division, or to monitor mitotic spindle assembly, and to prevent exit from mitosis until chromosome segregation has been completed. Thus, well functioning checkpoint mechanisms are central to the maintenance of genomic integrity and the basic viability of cells and, the- fore, are essential for proper development and survival. The importance of proper functioning of checkpoints becomes plainly obvious under conditions in which this control network malfunctions and fails. Depending on the severity and timing, failure of this machinery can lead to embryonic lethality, genetic diseases, and cancer.

Available in PDF, EPUB and Kindle. Book excerpt: Now in its second year, Progress in Cell Cycle Research was conceived to serve as an up to date introduction to various aspects of the cell division cycle. Although an annual review in any field of scientific investigation can never be as current as desired, especially in the cell cycle field, we hope that this volume will be helpful to students, to recent graduates considering a de1liation in subject and to investigators at the fringe of the cell cycle field wishing to bridge frontiers. An instructive approach to many subjects in biology is often to make comparisons between evolutionary distant organisms. If one is willing to accept that yeast represent a model primitive eukaryote, then it is possible to make some interesting comparisons of cell cycle control mechanisms between mammals and our little unicellular cousins. By and large unicellular organisms have no need for intracellular communication. With the exception of the mating phenomenon in S. cerevisiae and perhaps some nutritional sensing mechanisms, cellular division of yeast proceeds with complete disregard for neighbourly communication. Multicellular organisms on the other hand, depend entirely on intracellular communication to maintain structural integrity. Consequently, elaborate networks have evolved to either prevent or promote appropriate cell division in multicellular organisms. Yet, as described in chapter two the rudimentary mechanisms for fine tuning the cell division cycle in higher eukaryotes are already apparent in yeast.
