All DNA samples assayed, heated or unheated, and fractionated or unfractionated, showed some degree of DNA-primer activity. The chromatographic profiles of heated DNA were different from those of unheated DNA. Phenol and p-aminosalicylate, used in the preparation of DNA, did not alter the patterns of DNA obtained by Ecteola chromatography. Sephadex chromatography was found to be a rapid and effective method to deionize DNA solutions. DNA-primer activity was increased by an ammonium hydroxide gradient. At 4 degrees, treatment of DNA by phenol and p-aminosalicylate, Ecteola column chromatography with a salt gradient, Sephadex column chromatography with deionized distilled water to desalt DNA, and lyophilization did not alter DNA-primer activity. The results indicate that DNA-primer activity, per se, is not specifically associated with one particular fraction of DNA. (Author).

The remaining chapters cover a range of chromatographic procedures based on the interaction of a specific ligand with its target protein or other macromolecule. Some chapters cover non-specific interactions using peptides, inhibitors, and antibodies as the affinity ligand while others focus on specific groups of molecules : oligosaccharides and glycosylated proteins, nucleotide-binding proteins, proteins binding free and chelated metal ions, and DNA binding proteins.

The borderline between population genetics and phylogenetics is vague and difficult to assess, and a combination of both types of tools is best when it is not clear with which area one is dealing. Furthermore, it is not now appropriate to use just one type of marker in any kind of study; most markers have the potential to misinform under certain conditions, so it is always wise to incorporate at least two different types of assessments into any project. This volume is designed to facilitate this sort of multiple approach and provides comparative data on most currently available methods so that researchers can more intelligently select those appropriate to their area of interest, regardless of whether it is in the realm of breeding or evolutionary biology.
