Dans un premier temps, de nouvelles méthodologies RMN 2D en milieu cristal liquide chiral ont été développées afin de simplifier l’analyse spectrale des spectres proton et carbone-13 permettant de mesurer simplement des couplages dipolaires et de visualiser la différenciation énantiomérique. Ensuite, nous avons étudié l’effet de la température, du champ magnétique, du degré de polymérisation et de la concentration en polymère sur la différenciation énantiomérique. Nous avons montré que ces paramètres modulent différemment celle-ci et qu’ils sont importants dans le cadre d’une optimisation de la méthode. Puis, nous avons décrit une méthode empirique de détermination de la configuration absolue par RMN du deutérium en abondance naturelle. Nous avons montré, dans le cas de dérivés d’époxydes chiraux, qu’à partir de l’étude d’une série de composés homologues de référence, nous avons une très bonne corrélation entre la forme tridimensionnelle du composé et son orientation dans la phase chirale anisotrope. Ceci nous a permis de déterminer la configuration absolue inconnue d’un composé chiral. Enfin, cette même série de dérivés d’époxydes a été étudiée par RMN du proton et du carbone-13. En calculant l’orientation des composés, nous étions en mesure de déterminer la configuration absolue d’un composé chiral inconnu nous amenant à la même conclusion que l’étude par RMN du deutérium. De plus, en généralisant ce calcul à l’ensemble de la molécule, nous avons montré qu’il est possible de déterminer la configuration relative des protons diastéréotopes du méthylène.

Nous avons d'abord montré que les expériences comprenant des impulsions sélectives permettaient d'extraire des informations des spectres proton et carbone-13 qui étaient inaccessibles sur les spectres monodimensionnels standards. Nous avons ensuite utilisé d'autres techniques pour mesurer un maximum de couplages dipolaires pour préciser de manière non-ambiguë les configurations relatives de centres stéréogènes dans des molécules rigides. Nous avons ainsi montré que pour la DHEA, un stéroïde, une seule géométrie relative des substituants était compatible avec l'ensemble des couplages dipolaires mesurés et conduisait à un écart type entre couplages dipolaires calculés et expérimentaux correct. Cette méthode est promise à un grand avenir car elle est beaucoup moins ambiguë que les méthodes classiques utilisées en milieu isotrope. Enfin, nous avons développé cette méthode dans le cas des molécules flexibles où le problème se complique car il existe une corrélation totale entre conformation moléculaire et ordre orientationnel. Le nombre de paramètres à itérer explose rapidement, alors que le nombre de mesurables reste constant, et donc un traitement général est impossible. En prenant pour exemple le 1,2-dibromopropane, nous avons montré que le problème est gérable, à condition d'utiliser un modèle paramétrique pour calculer la valeur moyenne des paramètres d'ordre pour chaque conformation. Les paramètres à itérer, sont alors beaucoup moins nombreux et le problème peut être traité. Ainsi, nous avons montré qu'avec quatre indices de liaison et la géométrie des différents conformères obtenue par calcul ab initio, nous pouvions obtenir un excellent accord entre les couplages dipolaires calculés et expérimentaux, et déterminer sans ambiguïté la configuration relative des protons méthyléniques par rapport à la configuration du centre chiral.

Book excerpt: La spectroscopie RMN 2D du deutérium en abondance naturelle (DAN) en utilisant les cristaux liquides chiraux comme solvant de RMN est une méthodologie efficace pour deux raisons: i) l'interaction quadrupolaire (interaction RMN sensible à l'ordre orientationnel des molécules et spécifique aux noyaux de spin I>1/2) n'est plus nulle en moyenne comme dans les liquides; ii) la chiralité du milieu permet de discriminer spectralement des énantiomères (molécule chirale) ou des directions énantiotopes (molécule prochirale). L'objectif principal de ce travail de Thèse était d'explorer le potentiel analytique de cette technique pour analyser le fractionnement isotopique naturel deutérium dans le cas d'ester d'acides gras saturés (AGS) et (poly)insaturés (AGI). Le but ultime de ce travail est de fournir de nouvelles informations (de nature stéréochimique) pour améliorer la compréhension de certains mécanismes enzymatiques impliqués au cours de leur biosynthèse. Dans ce contexte, nous avons montré que l'utilisation de solutions organiques orientées de polypeptides (mésophases chirales) utilisant des co-solvants polaires comme le DMF ou la pyridine (Py) fournissait les meilleurs résultats en termes d'(énantio)discrimination spectrale (nombre de sites et amplitude) sur la base des déplacements chimiques et des éclatements quadrupolaires du deutérium. Ainsi, la mésophase chirale (PBLG/Py) a été utilisée avec succès pour analyser les AGI comme le linoléate de méthyle (C18), pour lequel nous avons pu mesurer l'excès énantio-isotopomèrique (eei) au niveau de chaque site méthylène de la molécule. Pour la première fois, d'importantes informations relatives à la stéréochimie des mécanismes enzymatiques, inaccessibles par la méthode conventionnelle SNIF-NMR®, ont été déterminées. La même méthodologie impliquant la mésophase achirale (PBG/Py) et chirale (PBLG/Py) s'est révélée également être un excellent outil pour l'analyse des SAFA sous leur forme libre (C14 à C18) ou sous leur forme de triglycérides (3*C4 et 3*C14). L'attribution des doublets 2H détectés sur les spectres RMN 2D anisotropes des SAFA a été confirmée par l'élaboration d'un modèle théorique capable de prédire le comportement orientationnel de ces molécules flexibles. Comme exemple illustratif, la méthode a été appliquée pour étudier les mécanismes enzymatiques convertissant le linoléate en vernoléate dans deux plantes différentes. Enfin, pour améliorer la compréhension des interactions «soluté-polypeptide» impliquées dans les mécanismes d'orientation et d'énantio-discrimination, nous avons étudié par RMN 2D DAN, l'évolution des paramètres d'ordre (matrice de Saupe) de deux molécules prochirales, rigides et apolaires dissoutes dans une mésophase chirale préparée en mélangeant deux polypeptides de même stéréochimie (L), mais dont la nature chimique des chaînes latérales est différente, en fonction de leur proportion massique respective.

Les mésophases utilisées sont des solutions d'homopolypeptides de synthèse dans un solvant organique hélicogène dans lequel le polymère adopte une conformation en hélice alpha.Les interactions énantiodiscriminantes sont-elles dues aux carbones asymétriques du polypeptide, à la chiralité macromoléculaire de la conformation en hélice ou aux deux effets ? Pour tenter de répondre à cette question, nous avons étudié des poly-(L-aspartates) ayant la propriété de passer d'une conformation en hélice a droite à une conformation en hélice gauche avec l'augmentation de la température. Cette étude a montré que l'hélice semble jouer un rôle important dans le phénomène d'énantiodiscrimination.Dans une seconde partie, le système glucopone®/n-hexanol/trizma a permis de visualiser les énantiomères de molécules hydrosolubles par RMN du deutérium découplé des protons et d'en estimer l'excès énantiomérique.Enfin, nous avons proposé une stratégie de dosage des amines par RMN dans les cristaux liquides chiraux. La RMN du carbone-13 permet visualiser des énantiomères soit par découplage des protons, en utilisant une expérience à deux dimensions : la HETSERF. La RMN du deutérium se révèle plus sensible à une différence d'ordre. La deutération de la molécule en cours de synthèse n'étant pas toujours réalisable facilement, nous avons mis au point une technique de dérivation des amines primaires et secondaires avec un agent achiral deutérié.

Book excerpt: Les cristaux liquides (CL) sont des milieux anisotropes et fluides, particulièrement intéressants comme solvants pour la résonance magnétique nucléaire (RMN). Ils permettent notamment de déterminer la structure de solutés et d'étudier leur comportement orientationnel. En particulier, dans un CL chiral (CLC), les interactions énantiosélectives soluté/solvant imposent deux énantiomères ou deux éléments énantiotopes à s'orienter différemment l'un de l'autre, contrairement au comportement observé dans un CL achiral. Ainsi, sur la base d'arguments de la Théorie des Groupes, il est récemment apparu que l'orientation des solutés non plans de symétrie Cs, C2v, D2d ou S4 dans un CL nématique achiral et dans un CLC nématique est différente, ce qui mène dans leur cas à des spectres de RMN distincts. L'objectif principal de cette Thèse est de valider expérimentalement ce concept, tout en illustrant le potentiel analytique de la RMN en milieu CLC dans le domaine de la stéréochimie organique. Pour cela, diverses structures rigides ou flexibles, polaires ou apolaires, de symétrie Cs, C2v, et D2d sont étudiées par RMN du 13C et du 2H notamment, dans un CLC nématique polypeptidique faiblement orienté pour solvant. Dans chaque exemple, les différences d'orientation des groupements, directions ou faces énantiotopes sont mises en évidence puis analysées en terme de paramètres d'ordre orientationnel. En particulier, l'inéquivalence dans un environnement chiral des directions énantiotopes de molécules C2v prochirales sans atome tétraédrique prostéréogène est présentée. En marge de ces résultats, nous montrons l'intérêt analytique de la RMN du 2H en abondance naturelle pour mesurer la pureté énantiomérique de molécules chirales ou analyser les rapports isotopiques 1H/2H de solutés prochiraux. L'ensemble des travaux démontre que la RMN en milieu orienté chiral et/ou achiral est un outil d'analyse performant, capable de fournir des informations de stéréochimie indispensables aux chimistes.

He also taught political economy successively from 1815 until his death in three Parisian establishments: the Athénée, the Conservatory of Arts and Trades, and the Collège de France. The texts in which Say exposes his approach to political economy have not been available in the English language until now except for the fourth edition of the ‘Preliminary Discourse’ which serves as an introduction to the Treatise. This book presents a translation which renders his works accessible to the English speaking world. For the first time, English readers will be able to become directly immersed in Say’s principal texts, where he develops his conception of political economy. Jean-Baptiste Say and Political Economy proposes a translation of a selection of eleven of Say’s texts. The first three are versions of the ‘Preliminary Discourse’ from the Treatise’s editions of 1803, 1814 and 1826 with the variations of the editions of 1817, 1819 and 1841. The following four texts are the opening discourses pronounced at the Conservatory in 1820 and 1828 and the Collège de France in 1831 and 1832. The eighth text is the ‘General Considerations’ which open the Complete Course in Practical Political Economy of 1828, with the variations of the 1840 re-edition. The final three texts are those Say devotes to ‘the progress of political economy’ in what is akin to a history of economic thought. This volume is of great importance to economic historians and people studying Jean-Baptiste Say, as well as those who are interested in economic theory and philosophy and political economy.
