Available in PDF, EPUB and Kindle. Book excerpt: Optical Coherence Tomography (OCT) is a non-invasive three-dimensional imaging technique. OCT synthesizes a cross-sectional image from a series of lateral adjacent depth scans, and with a two-dimensional scanning scheme, three-dimensional intensity image of sample can be constructed. Due to its non-invasive capability, OCT has been widely applied in ophthalmology, cardiology and dermatology; and in addition to three-dimensional intensity image construction, various functional OCT imaging techniques have been developed for clinical application. My research is focused on developing functional OCT systems for application in ophthalmology, including polarization-sensitive optical coherence tomography (PS-OCT) for retinal nerve fiber layer (RNFL) birefringence measurement and dual-wavelength photothermal optical coherence tomography (DWP-OCT) for microvasculature blood oxygen saturation (SO2) measurement. In the study, a single-mode-fiber based polarization-sensitive swept-source OCT (PS-SS-OCT) with polarization modulator, polarization-sensitive bulk-optics balanced detection module is constructed and polarization processing methods based on Stokes vectors are applied to determine birefringence. PS-OCT is able to provide human subject's RNFL thickness, phase retardation, and birefringence information. Degradation in the degree of polarization (DOP) along depth is investigated and its difference between four quadrants of RNFL (superior, temporal, inferior and nasal) indicates the structural property difference. DWP-OCT is a novel functional OCT system consisting of a phase-sensitive optical coherence tomography system (PhS-OCT) and two photothermal excitation lasers. PhS-OCT is based on a swept-source laser operating in the 1060 nm wavelength range; the two photothermal excitation lasers with wavelength 770 nm and 800 nm are intensity modulated at different frequencies. PhS-OCT probe beam and two photothermal excitation beams are combined and incident on the sample, optical pathlength (op) change on the sample introduced by two photothermal excitation beams are measured and used for blood SO2 estimation. A polarization microscope is proposed for future study. The polarization microscope is an imaging technique providing molecular structure and orientation based on probe light's polarization state information. The polarization microscope uses a wavelength tunable light source, and can achieve any incident polarization state by a retarder-rotator combination. Specimen's birefringence can be determined based on the changing of detected light amplitude.

Book excerpt: Because of its many advantages optical coherence tomography (OCT) has revolutionized the way in which retinal diseases are screened and managed and how treatments are monitored. In this volume the latest developments and findings are presented by experts in their respective fields. After a short introduction covering the available equipment and the basic techniques, the imaging features of various pathological findings in retinal diseases are presented. The topics cover the outer layers including new modalities for choroid imaging, out-layer diseases such as the various types of macular degeneration, retinal diseases such as diabetic retinopathy and vascular occlusion, and retina and vitreous interface pathologies. The final chapters are dedicated to the practicality of using OCT for the pre- and postsurgical evaluation of the posterior segment and for the differential diagnosis of vitreoretinal diseases as well as in the management of patients with retinal and neuro-ophthalmological diseases. Making the essentials of the recently held ESASO course on OCT available in one volume, this publication is a must-read for experienced as well as trainee ophthalmologists who need to use OCT in their daily practice.

Optical coherence tomography (OCT) is one of the most exciting inventions in biomedical optics. This dissertation represented my Ph.D. research work on ophthalmic OCT under the mentoring of Dr. Zhongping Chen at Beckman Laser Institute & medical clinic of University of California, Irvine (BLIMC-UCI). My other academic advisors included Dr. Bruce Tromberg, Dr. Anthony Durkin, Dr. Ron Kurtz, and Dr. Barry Kupperman. The in vivo spectral Doppler OCT imaging technology developed to image dynamic, pulsatile retinal blood vessels was highlighted in this dissertation. Some technical details of the ophthalmic OCT imaging system I developed at BLIMC-UCI were discussed. Some collaborated ophthalmic research work such as screening retina transplants and monitoring subsurface femtosecond laser photo-disruption on anterior segment of human eye for Glaucoma treatment were also presented. I wish the publication of this dissertation will not only promote scientific knowledge, but also stimulate interest of readers in developing more OCT applications.

Available in PDF, EPUB and Kindle. Book excerpt: Optical coherence tomography (OCT) is a non-invasive micrometer-resolution volumetric imaging modality that has been employed in diverse applications. In this thesis, we first describe a GPU accelerated program to perform FDOCT data processing and real time 3D volumetric rendering. The real time visualization of volumetric images provided by the GPU acceleration was essential to the rest of the work described in this thesis. Small animal models of retinal diseases serve as a vital component in vision research, and non-invasive in vivo imaging is becoming an increasingly important tool in the field. We describe the first adaptive optics optical coherence tomography (AOOCT) imaging system for high resolution mouse retinal imaging. Images of mouse retina acquired with AOOCT showed significant improvement in the brightness and contrast of capillaries and nerve fiber bundles. However, the accuracy of wavefront sensing limited the performance of AOOCT. A novel wavefront sensorless adaptive optics (WSAO) OCT system was developed to overcome the issues associated with conventional wavefront sensing. Combination of WSAO with OCT allows coherence gated, depth resolved aberration correction. Images of both pigmented and albino mouse retinas acquired using WSAO OCT system demonstrated superior image quality. The real time high resolution WSAO OCT was leveraged for a time course study of laser exposure in the retina.

Book excerpt: This book covers the results of the creation of methods for ophthalmologists support in OCT images automated analysis. These methods, like the application developed on their basis, are used during routine examinations carried out in hospital. The monograph comprises proposals of new and also of known algorithms, modified by authors, for image analysis and processing, presented on the basis of example of Matlab environment with Image Processing tools. The results are not only obtained fully automatically, but also repeatable, providing doctors with quantitative information on the degree of pathology occurring in the patient. In this case the anterior and posterior eye segment is analysed, e.g. the measurement of the filtration angle or individual layers thickness. To introduce the Readers to subtleties related to the implementation of selected fragments of algorithms, the notation of some of them in the Matlab environment has been given. The presented source code is shown only in the form of example of implementable selected algorithm. In no way we impose here the method of resolution on the Reader and we only provide the confirmation of a possibility of its practical implementation.
