Book excerpt: Drug chemistry sections of forensic crime laboratories are tasked with analyzing a variety of unknown materials, a subset of which are derived from plants. In comparison to the analysis of small-molecule drugs, these plant matrices are incredibly complex and present unique challenges. When traditional chromatographic approaches are used to interrogate these materials in forensic investigations, large amounts of time, resources, and funds are directed towards the extensive sample preparation steps required. Exacerbating the issue is that the matrix characteristics often result in instrument downtime to accomplish the maintenance required to address ingredients adhering to columns or clogging syringes, which are common problems when testing Cannabis-derived samples. Furthermore, the increased legalization and decriminalization of marijuana in the U.S. has contributed to the rise in popularity of Cannabis products, which will require new testing methods to analyze the myriad of sample types available as the industry continues to evolve. Therefore, the unique capabilities of direct analysis in real time – high-resolution mass spectrometry (DART-HRMS) were investigated for the analysis of plant materials, including Cannabis, as an approach to circumventing many of the sample preparation and other difficulties often encountered. A DART-HRMS triage approach, which requires no sample pretreatment steps, has been developed for the detection of Cannabis-related molecules in a wide variety of sample types, including edibles, beverages, concentrates, topicals, vaporizers, marijuana/hemp flower, and pre-rolls. Additional applications of DART-HRMS for the analysis of Cannabis materials include the following: (1) differentiation of cannabinoids; (2) quantification of cannabinoids in edibles extracts; and (3) distinguishing between hemp and marijuana varieties of C. sativa through statistical analysis of mass spectral data. DART-HRMS was also proven effective for the analysis of plant-based legal-highs, specifically those identified as “plants of concern” by the United Nations Office on Drugs and Crime. Ambient ionization mass spectrometry (AIMS) approaches were developed for the rapid detection and validated quantification of psychoactive small-molecule constituents in complex plant matrices. The methods described would greatly improve forensic science and criminal justice practices in the U.S. for the analysis of complex plant materials, both Cannabis-derived and plant-based legal-highs.

This dissertation describes the development and implementation of rapid and novel MS based methods for the analysis of targeted analytes such as drugs and metabolites from complex biological matrices in two main fields of application, drug development and phytometabolomics. We explored the potential of quantitative mass spectrometry imaging (MSI) by the ambient MS technique, desorption electrospray ionization-mass spectrometry (DESI-MS). The discovery of quantitation MSI methods were explored and evaluated based on its analytical figures of merit (precision, accuracy, linearity, and so forth). Triple threat methods covering the identification, quantification and mapping the spatial distribution of analytes from biological tissue sections opens new avenues in MSI. The development of targeted qualitative and quantitative ESI-MS, DESI-MS and multistage mass spectrometry, DESI-MSn, methods for plant metabolomics were also investigated. DESI-MS and DESI-MSn methods coupled offline to normal phase high performance thin layer chromatography (HPTLC) and C18 functionalized reverse phase (RPTLC) were created to identify and separate bioactive secondary metabolites from natural products such as seeds, roots, twigs, and leaves. The implementation of these rapid, robust and high-throughput methods allowed for the discovery of novel phytochemicals in the plant species under investigation.

The development of soft ionization methods such as electrospray ionization (ESI) and matrix-assisted laser desorption ionization (MALDI) have made these applications possible. However, the requirement for extensive sample preparation has limited analyses that require complex sampling. These limitations in the analysis process have led to the expansion of ambient ionization mass spectrometry. This dissertation describes the development and optimization of novel ambient ionization methods that rely on porous/cellulose-based substrates such as thread and paper. Chapter 2 depicts a thread spray ionization method with forensic serology applications. Thread spray ionization enables direct analysis of trace levels of blood on textiles without extensive sample preparation reducing backlog seen in many criminal laboratories. This dissertation describes the successful implementation of thread spray as a rapid presumptive method of blood-stained textiles through the identification of heme in whole blood. Thread spray ionization is also a suitable confirmatory method through the direct differentiation of human, canine, and horse blood by determining the species’ specific phospholipid lipid profiles. Chapter 3 outlines thread spray ionization as a rapid in situ method for detection of agrochemicals in complex matrices such as surface water and foodstuff. Determination of pesticide residues in a wide variety of matrices is an ongoing challenge due to small quantities of analytes and substantial amounts of interfering substances that can be coextracted with the analytes. In this chapter, we detail the use of a nano-thread spray mass spectrometry approach that utilizes a nano spray like emitter, as opposed to the traditional thread spray ionization apparatus, to enhance on-thread separation of agrochemicals and environmental matrices. This analysis was used for the detection of glyphosate (GLYP) and its metabolite, aminomethylphosphonic acid (AMPA), in surface water. Nano thread spray ionization was then used for the detection of the pesticides diphenylamine (DPA) and thiabendazole on the surface and cores of post-harvest dipped fruits. Chapter 4 describes the development and optimization of a 3-dimensional (3D) wax-printed microfluidic paper-based analytical device, μPAD, for multiplexed disease detection. The 3D wax-printed μPAD is designed for on-line sample splitting, reagent storage, and MS analysis via stacking different layers of two-dimensional (2D) wax-printed paper substrates together. Assembled devices’ transport efficiency were evaluated using Bovine Serum Albumin (BSA-FITC), where μPADs efficiently transported BSA-FITC through devices. We believe the proof-of-concept 3D wax-printed μPAD described in this chapter will be capable of direct multiplex analysis for paper-based immunoassay in resource-limited settings.

Book excerpt: Ambient mass spectrometry (MS) is a new and growing sub-field in MS which has opened new research avenues, particularly for applications relating to the analysis of solid samples. Results on the implementation and application of ambient MS techniques including: desorption electrospray ionization (DESI) and direct analysis in real time (DART) indicated that these techniques could serve as complementary tools for the rapid qualitative screening of pharmaceuticals, allowing up to two orders of magnitude improvement in throughput compared to traditional methods such as liquid chromatography MS. The selectivity of DESI could be enhanced by performing the experiment in the reactive mode. In this mode, complexation reactions between reagents added to the spray solvent and analytes on the sample surface resulted in analyte stabilization, inhibiting fragmentation. They also resulted in a concomitant enhancement in the analyte surface activity, facilitating their evaporation from secondary droplets culminating in an improvement in sensitivity. Also for drug tablets analysis, the analyte signal dependency on DESI geometrical set-up variables could be mitigated following the careful and controlled addition of an isotopically labeled internal standard (IS) to the sample or by spraying samples with a pair of reagents with different affinities for the analyte. Either of these approaches resulted in an analyte-to-IS signal ratio (in the former) or an analyte complex ratio (in the later), which was largely independent of DESI experimental variables allowing quantitative analysis using this technique. DESI MS was also observed to be a very powerful tool for determining the 2-D distribution of various pharmaceutically important compounds on tablet and tissue surfaces. The ability to map the distribution of molecules of interest by DESI MS has very great implications in drug tablet quality control and in determining the role of chemical signals presented on tissue surfaces. DESI was observed to be limited to ionizing molecules of medium to high polarities without much limitation in terms of mass range, whereas DART was better suited for the analysis of molecules within a broader range of polarities, but within a more limited mass range (up to 800 Da approximately). These limitations were circumvented by implementing a novel multimode ambient ion source, desorption electrospray/metastable-induced ionization (DEMI), which combines various aspects of DESI and DART. Initial experiments with the DEMI ion source demonstrated its ability to enable the simultaneous analysis of molecules within a broader range of polarities and masses than DESI and DART alone.

This dissertation serves to outline recent advancements made towards the improvements of ambient ionization MS technologies as well as the application of these techniques towards disease indications that have yet to be explored. Chapter 2 provides data and discussion about how relative humidity can contribute to variability and poor data quality during desorption electrospray ionization (DESI) MS experiments, providing essential information about how atmospheric conditions can contribute to the robustness of the technique. Chapters 3 and 4 present methods for integration of new separation and fragmentation techniques with ambient ionization methods in order to increase the amount of information that can be gathered from a biological specimen. Finally, Chapter 5 presents results from the utilization of DESI-MS imaging towards understanding endometriosis, a chronic gynecological condition that has limited diagnosis and treatment options for patients. The ideal approach would involve a singular, portable device for rapid, on-site analysis of broad chemical classes residing diverse environments. The pursuit of high throughput via on-site analysis has led to the merging of ambient ionization methods, which allow for rapid screening of samples in their native state, and portable mass spectrometric (MS) systems with ambient sampling capability. Numerous ambient ionization methods have been reported, varying in terms of complexity and rigor of operation, and both paper spray ionization - mass spectrometry (PSI-MS) and filter cone spray ionization - mass spectrometry (FCSI-MS) stand out in terms of broad applicability. In this work, a streamlined approach for the investigation of bulk, trace, and surface-swabbed chemicals was examined using a combination of PSI and FCSI sources. Rapid screening of N-nitrosamine (NAMs) contamination served as an efficacy test for this combined approach, as this class of carcinogenic and genotoxic compounds has been found recently in a myriad of matrices of interest to forensics, pharmaceutical quality control, and environmental science.While PSI-MS and FCSI-MS were shown capable to rapidly process a wide range of sample types, several performance and user-based issues were observed with the ion sources utilized, leading to the utilization of 3D printing-based prototyping as a way to quickly design and test innovative solutions. This led to the generation of a new ambient ionization technique, 3D-printed cone spray ionization (3D-PCSI), as means of overcoming known issues with bulk sample screening, and its application towards profiling chemical warfare agent (CWA) simulants in soil was demonstrated. Both liquid chromatography-mass spectrometry (LC-MS) and mass spectrometry imaging (MSI) techniques are useful for large-scale detection and identification of small molecules, peptides, and proteins in samples, with MSI additionally being able to determine the spatial distribution of molecules within a tissue section. In this thesis, LC-MS and MSI approaches were applied to study symbiotic relationships. In the plant Medicago truncatula, a symbiotic relationship with rhizobia bacteria in specialized organs, root nodules, of the plant results in the reduction of atmospheric nitrogen into ammonia (termed biological nitrogen fixation). MSI methods were developed to investigate the small molecule and peptide content in the root nodules. Furthermore, a developed MSI method was applied to study salt stress, which negatively affects development of the symbiotic relationship in the root nodules. The other relationship explored was the complex interactions between the gut microbiome and its host. To investigate this complicated relationship, multiomics studies, which combine two or more single omics studies, such as metabolomics and proteomics, were utilized to achieve a more comprehensive analysis of the gut microbiome. Developed multiomic approaches were applied to study the response of the gut microbiome to pathogenic infection. Overall, this work develops a multifaceted approach to investigate symbiotic relationships, resulting in new method development, improved MSI sample preparation protocols, and increased knowledge about molecular players involved in the symbiosis.

Book excerpt: Portable mass spectrometry (MS) has become an area of interest due to the ability to conduct chemical analyses in native environments. These instruments have the potential to increase efficiency and analytical throughput via real-time experimentation, minimizing the need for sample transportation. One drive towards the development of fieldable mass spectrometers is the potential for coupling to novel ambient ionization methods (or "ambient MS"), which can generate spectral data at ambient conditions from unprepared or minimally prepared samples of interest. Since the first introduction of the desorption electrospray ionization (DESI) technique in 2004, a myriad of novel techniques have been reported and applied, such as paper spray ionization (PSI), direct analysis in real time (DART), filter cone spray ionization (FCSI), etc. These methods allow for diverse sample types to be processed, opening the door to a variety of impactful applications.Forensics and homeland security are fields that can benefit from portable instrumentation, creating quick analytical capabilities during a criminal investigation or public safety events. Though it has been observed that portable MS systems can identify samples such as drugs of abuse, chemical warfare agents (CWAs), and explosives in a range of sample matrices, other factors/criteria must be considered before implementing into standard procedural methods of evidence collection. A factor to consider, however, is the instrumentals performance during exposure to common environmental conditions. The effect of temperature, relative humidity and windspeed were tested on the FLIR Systems Griffin Atmospheric Inlet-Mass Spectrometer (AI-MS) 1.2, observing target detection, signal intensity and signal duration while implementing PSI-MS, a newer ambient MS method that employs disposable paper substrates for sample collection and processing. Trends were interpreted to determine optimal conditions for high instrument performance, as well as potential conditions that yield a negative impact on sample processing. Other criteria are of note, such as the recommendations provided by the Scientific Working Group for the Analysis of Seized Drugs (SWGDRUG) for generating instrumental methods for forensic evidence confirmation; these recommendations center on the need for a multi-method analytical scheme for producing a definitive identification with prosecutorial ramifications. Towards this, a platform for combining surface enhanced Raman spectroscopy (SERS) and PSI-MS was developed to characterize the processing of authentic illicit drug analytes. A validation study was then performed, testing the overall reliability and accuracy of the dual methodology through a user blinded error rate study. Further, the potential admissibility of forensic drug evidentiary data produced from portable MS systems was examined, using the constraints of the Daubert or Frye standards for forensic evidence testimony as a guide. Overall, the efforts contained in this thesis seek to demonstrate the potential of portable mass spectrometry towards forensic/homeland security investigations.
