Available in PDF, EPUB and Kindle. Book excerpt: Isocyanide based multicomponent reactions, including the Ugi four-component and Passerini three-component reactions, offer chemists a powerful tool for quickly constructing complex molecules. A particular form of the Ugi reaction, in which two of the four reacting components are introduced into a single molecule, is useful for synthesizing cyclic products. Using a newly developed convertible isocyanide, known as indole isocyanide, in the Ugi reaction allowed for the mild synthesis of various pyroglutamic acids. This effort led first to a racemic total synthesis of the natural product dysibetaine. A project to achieve the stereoselective synthesis of the same natural product led to the discovery of a unique Ugi reaction. Usually a carboxylic acid is one of the four reacting components in this reaction, however in this unusual case an ester acts as the carboxylic acid component. In addition to the natural product syntheses, a detailed explanation of the scope and mechanism of the new convertible indole isocyanide is presented. Finally, we reveal a new stereoselective, Brønsted-acid catalyzed Passerini-type reaction.

Book excerpt: Recent Advances in Applications of Name Reactions in Multicomponent Reactions is an ideal reference for researchers and postgraduate students studying organic chemistry, as well as synthetic organic chemists working on the development of novel methodologies for the synthesis of various heterocyclic systems, especially drug design and discovery, in both academia and industry. The book reviews recent applications of name reactions in multicomponents for the synthesis of heterocycles and examines recent advances in applications of significant name reactions, such as Ugi and Passirini, Click, Knoevenagel, Michael, Diels-Alder, Aldol, Mannich, Heck, Huisgen, and Suzuki in MCRs. These reactions can be used in the synthesis of a wide variety of novel heterocycles with different sizes and heteroatoms, as well as in the total synthesis of natural products in order to decrease the number of synthetic steps. Since chiral inductions are necessary for most of these sequential name reactions, their asymmetric catalyzed reactions are also described. - Includes the synthesis of many heterocycles, which is ideal for synthetic organic chemists engaged in the synthesis of heterocyclic systems - Covers the recent advances of asymmetric synthesis of a wide range of heterocycles in satisfactory enantioselectivities (ees) or distereoselectivities (des) - Reviews the synthesis of a wide variety of interesting heterocycles by using a combination of different and versatile name reactions via MCRs

Book excerpt: While very useful for studying syntheses of molecular diversity, multi-component reactions also offer rapid access to a variety of complex molecules that are relevant for biological applications. Multi-component Reactions in Molecular Diversity analyzes these reactions, whether they are realized by organometallic, ionic or even radical processes. It highlights popular methods based on monotype reactions (cascade, tandem, domino) and their efficiency and academic industrial domain are illustrated. This book also investigates the most efficient ways to prepare complex molecules. Multi-component reactions are in tune with the concepts of atom and steps economy, which are of prior importance in all the reported processes ? from the laboratory to the pilot scale. The essential criteria for green chemistry are also examined in the book in detail.

Book excerpt: With the explosion of combinatorial solid-phase methods, access to information has become one of the main barriers facing a synthetic chemist who is contemplating a combinatorial approach to a medicinal chemistry problem. The Combinatorial Index is an answer to that problem. This compendium of methods from the primary literature provides quick and convenient access to reliable synthetic transformations as well as information on linkers and analytical methods. Each synthetic procedure is preceded by a section entitled"Points of Interest,"which highlights the strengths and weaknesses of the various studies. The index also covers the use of solution-based synthesis for the generation of molecular diversity. - Organized for rapid retrieval of published information on classes of synthetic transformations, linkers, and analytical methods - Serves as a laboratory manual for bench chemists - Includes a chapter on linkers to assist in choice of linking strategy - Discusses strengths and limitations of the various methods - Contains a structural index showing functional group transformations in solid-phase synthesis
