Isolates of the PVY species are highly variable at biological, serological and molecular levels. Epidemiological studies have highlighted the emergence of distinct potato PVY variants able to induce necroses on potato tubers. Due to the lack of efficient resistance to PVY isolates inducing necrotic symptoms in cultivated varieties and the plant-to-plant transmission of isolates through the daughter tubers, PVY has become the most economically important virus for the potato industry. The review offers an overview of several decades of research on PVY but also focuses on the latest data obtained by expert on PVY worldwide on the biological characteristics of PVY, interactions between aphids-hosts, its evolution and management. Identified knowledge gaps to understand further PVY biology will be discussed.

Book excerpt: In North America, management of viruses in seed potatoes has primarily been done through seed certification. In last decade, the effectiveness of certification has been challenged by the reemergence of Potato virus Y (PVY). The potyvirus is transmitted by aphids in a non-persistent manner. Three major factors have led to its reemergence: i) the occurrence of PVY strains that show mild symptoms and cause tuber necrotic ringspot disease; ii) the release and popularity of potato cultivars that do not show symptoms of infection; and iii) the limited availability of resistant cultivars. These factors have resulted in higher incidence of PVY in seed potatoes and rejection of seed lots in recent years, which has an economic impact on the seed potato industry. Additionally, seed certification programs have seen an increase in the cost of PVY management. The research presented here describes the development and evaluation of tools that can improve current PVY management. I addressed the need for a more cost-efficient and flexible assay to detect potato viruses through the optimization of a chemiluminescent dot-blot immunoassay. I also evaluated the usefulness of using minitubers as seed to reduce the risk of PVY in seed propagation. I found that, compared to plants grown from conventional seed, minituber-grown plants produced lower yields, emerged and matured later, but were not more susceptible to PVY. Finally, I collaborated with breeders at the University of Wisconsin-Madison to address the need for PVY resistant commercial potato cultivars in two projects. The first project involved the application of marker-assisted selection to introduce PVY resistance genes Ryadg and Rysto into Wisconsin cultivars. The results of screening populations derived from crosses with resistant parents provided evidence of the usefulness of molecular markers for selecting PVY resistant plants and suggest that PVY resistance is not linked to undesirable traits. The second project focused on identifying and evaluating markers linked to PVY resistance gene Rychc. The results suggest that the markers developed are useful for selecting resistant plants, as well as provide evidence of the presence of a lethal factor associated with PVY susceptibility that could aid in identifying resistant individuals.
