Book excerpt: Amphiphilic molecules comprised of a hydrophilic and hydrophobic domain are able to self-assemble into a variety of higher order aggregates. These aggregate structures, and their diverse morphologies, have been utilized for the delivery of bioactive agents. Additionally, amphiphiles can be tailored to exhibit inherent bioactivity. This dissertation describes the design and synthesis of amphiphilic molecules that self-assemble into aggregate structures with defined physicochemical properties. Their formulation and biological activity for diverse biomedical and personal care applications are fully characterized. Amphiphilic macromolecules (AMs) conjugated to ligands known to activate the G-coupled protein receptor TGR5 were investigated as nanoparticle (NP) formulations for the reduction of inflammation in atherosclerotic macrophages. Macrophages propagate the atherosclerotic cascade by uncontrolled internalization of oxidized low-density lipoprotein (oxLDL) and subsequent secretion of inflammatory cytokines. AMs, based on an acylated sugar backbone conjugated to poly(ethylene glycol) (PEG), were synthesized containing a lithocholic acid (LCA) moiety, a known TGR5 agonist. Ligand-conjugated AMs were formulated into NPs to mitigate the lipid burden and inflammatory phenotype by competitively inhibiting oxLDL uptake through scavenger receptor (SR) interactions and activating the athero-protective receptor TGR5. Ligand-conjugated AM NPs significantly reduce oxLDL uptake compared to untreated controls and lower expression of inflammatory genes under direct control of TGR5. These studies demonstrate the potential of ligand-conjugated AM NPs to reduce the atherosclerotic phenotype in activated macrophages. Modifications were also made to AMs to enable their incorporation into distearoylphosphatidylcholine- (DSPC- ) based liposomes for delivery applications. Liposome use has aided in the bioavailability, solubility, and improved pharmacokinetic profiles of a wide variety of active ingredients for biomedical and personal care products. This work expands upon the AM design to generate two series of molecules that simultaneously stabilize liposome colloidal properties and can be utilized to fine-tune release profiles of encapsulated cargo. Two series of AMs were synthesized with variations in their hydrophobic domains. All AMs improve upon stability properties at storage and physiological temperatures compared to DSPC-based liposomes alone. The chemical features of AMs, particularly the degree of unsaturation in the hydrophobic domain, influence release of hydrophilic molecules from liposomes' interior. Molecular dynamics (MD) simulations reveal that AMs' chemical structures influence local lipid properties, leading to the experimentally observed results. Together, this data offers insight that can be applied to design AMs with desirable physicochemical properties for bioactive delivery. Small molecule cationic amphiphiles (CAms) were designed to combat the rapid rise in drug resistant bacteria. CAms were designed to target and compromise the structural integrity of bacteria membranes, leading to cell rupture and death. Discrete structural features of CAms were varied and structure-activity relationship studies were performed to guide the rational design of potent antimicrobials with desirable selectivity and cytocompatibility profiles. In particular, the effect of cationic conformational flexibility, hydrophobic domain flexibility, and hydrophobic domain architecture were evaluated. Their influence on antimicrobial efficacy in Gram-positive and Gram-negative bacteria was determined, and their safety profiles established by assessing their impact on mammalian cells. All CAms have potent activity against bacteria and hydrophobic domain rigidity and branched architecture contribute to specificity. The insights gained from this project will aid in the optimization of CAm structures. Together, these three primary projects build upon the design of biocompatible amphiphiles that enable the delivery of bioactive molecules. Thorough structure-activity relationship studies were performed in each chapter to identify and generate amphiphiles with desirable outcomes for the specific application.

Book excerpt: Supramolecular nanoassemblies capable of reducing non-specific interactions with biological macromolecules, such as proteins, are of great importance for various biological applications especially for targeted drug delivery therapeutics. Recently, zwitterionic materials have been shown to reduce non-specific interactions with biomolecules, owing both to their charge neutrality and their ability to form strong hydration layer around zwitterions via electrostatic interactions. This dissertation focuses on design, synthesis, thorough characterization, and applications of zwitterionic amphiphilic dendrimers and polymeric materials. Firstly, A new triazole-based zwitterionic moiety was conceived and incorporated as the hydrophilic functionality in facially amphiphilic dendrons. Self-assembly characteristics and the structural and functional characteristics of the zwitterionic dendrons were then evaluated by spectroscopic techniques and by comparison with the corresponding charge-neutral PEG and anionic carboxylate-based dendrons respectively. Surface charge measurements, temperature sensitivity and evaluation of interactions of these assemblies with proteins form the bases for these comparisons. Because of the charge neutral characteristics of zwitterionic moieties exposed to outer aqueous phase, these dendritic assemblies evade non-specific interactions with biological milieu while enhancing the responsiveness towards specific biological triggers, such as proteins. We further simplified this molecular design by developing synthetically accessible zwitterionic carboxybetaine based amphiphilic polymers. Zwitterionic amphiphilic polymers are synthesized in a simple one-pot reaction by treating reactive acrylate polymer with secondary amine and 2-bromoaceticacid. This zwitterionic polymer possesses hydrophilic glycinebetaine and hydrophobic decyl chains placed orthogonally and forms environment dependent self-assembled aggregates owing to its amphiphilicity. The zwitterionic assemblies stably encapsulate guest molecules and undergo pH-dependent zeta potential and size variations owing to the pH sensitivity of zwitterionic component. In a related project, we've developed degradable polycarbonate based amphiphilic polymers possessing hydrophilic peg and hydrophobic decyl components. This degradable polymers form self-assembled aggregates encapsulate hydrophobic guest molecules, undergo degradation upon changes in pH or in presence of enzymes (PLE, an esterase). Work is underway towards the design and synthesis of zwitterionic amphiphilic degradable polymers and their characterization. Over all the work presented in this thesis have implications in design and development of polymeric excipients for targeted drug delivery, controlled release of drugs, and stimuli-sensitive therapeutics.

In the first part of this work, the author describes the synthesis and characterization of two colloidal systems with dumbbell-shaped core-shell morphology. The core consists of poly(methyl methacrylate) and poly(styrene). As a special feature, the attached shell layer promotes the change of the particle size and the aspect ratio by simply changing the ionic strength or temperature. Depolarized dynamic light scattering (DDLS) was used to investigate how these stimuli affect the translational and the rotational diffusion in the highly diluted state. The hydrodynamics of the particles could be well described with stick-boundary conditions by using analytical expressions for a double sphere, prolate ellipsoid and cylinder or the shell model, respectively. Electron and scanning force microscopy were applied to image the particle morphology in real space. The second part is devoted to the hydrodynamics of monodisperse, submicron-sized colloidal clusters, which consist of one to four spherical building blocks. The author demonstrates that the shell model is an excellent tool to identify the rotational relaxations which are accessible by DDLS.

Self-assembled structures of purely peptidic amphiphiles exploit these properties to produce biocompatible, biodegradable, smart materials for drug administration. This thesis explores the design, synthesis, purification, characterization of purely peptidic amphiphiles, and the evaluation of potential applications. The first chapter provides a general introduction to the field of self-assembly and drug delivery as compared to nature’s delivery mechanisms. Further, the advantage of amino acid based molecules in producing smart materials for drug delivery applications is highlighted via biocompatibility and biodegradability considerations. Next, synthetic strategies and purification methods are discussed. Finally gramicidin A (gA) – a naturally occurring, short, hydrophobic, membrane-integrating peptide used to produce the amphiphilic peptides presented here – is introduced. Chapter two presents an initial approach to produce self-assembled structures by purely peptidic amphiphiles. The undecamer used features a repetitive L-tryptophan and D-leucine [LW-DL] motif representing the hydrophobic block, and an N-terminally attached hydrophilic (lysine or acetylated lysine) section. Besides solid-phase peptide synthesis and purification, the process that self-assembles micelles and spherical peptide particles, termed “peptide beads” was characterized as a function of temperature and solvent composition by means of electron paramagnetic resonance (EPR), dynamic and static light scattering, fluorimetry and electron microscopy. An equilibrium process between single peptide molecules, micelles and peptide beads is then presented. Chapter three examines the structure of self-assembled peptide beads of diameters between 200 to 1500 nm. The beads were analyzed by electron and atomic force microscopy (AFM), static and dynamic light-, and small angle X-ray scattering. The beads are seen to result from hierarchical organization of micellar-like subunits and confirm the concept of multicompartment micelles. An improved understanding of the beads’ capacity to embed hydrophobic and hydrophilic payloads and provide perspectives for drug delivery applications emerges. Chapter four presents a library of longer peptides, based on the full sequence of gA. The peptide design includes three parts: (a) a charged lysine part, (b) an acetylated lysine part and (c) a constant hydrophobic rod-like helix, based on gA. Stepwise replacement of free lysine (K) with acetylated lysine (X) generated the ten peptides Ac-X8-gA and KmX8-m-gA (m ranging from 0 to 8). With the change in the primary sequence, a change in secondary structure was observed. The transition reflected a change in the self-assembled structures from fibers to micelles. This demonstrates how even small point mutations influence the supramolecular outcome and serve as an important step to understanding and controlling self-assembly. In chapter five, the knowledge gained on gA-based peptides is applied to produce purely peptidic vesicles. The work here demonstrated that, to form such structures with short amphiphiles, additional stabilizing factors were necessary. Thus, we exploited different dimerization strategies to form stable peptide membranes and developed a general recipe to form purely peptidic vesicles. The vesicles demonstrated pH responsiveness as well as the capacity to embed hydrophilic and hydrophobic payloads in their structure. Chapter six presents the potential of self-assembled peptide beads in drug delivery applications. The hydrophobic and hydrophilic payload-filled peptide beads are shown to be internalized by human cells. Further, a method to increase embedding efficiency for RNA/DNA to 99% due to charge-driven complexation and embedding is presented. The internalization of the gene delivery vehicle into cells led to gene silencing through delivered siRNA and to antibiotic resistance, and siRNA production followed by gene silencing through a delivered plasmid. The delivery of co-embedded paclitaxel and doxorubicin was then probed and proven effective. The results also demonstrate that the new class of drug delivery material caused no measurable toxicity during the experiments. Therefore, the material is suggested as a biocompatible drug delivery vehicle for gene therapy and multi-drug delivery. In chapter 7, the self-assembly capacity of the peptide is used to template the dense packing of gold nanoparticles. The C-terminally cysteinated peptide Ac-X3-gT-C was used to coat gold nanoparticles and form gold core micelles. These micelles then aggregate to composite peptide-gold nanoparticles in which the individual gold nanoparticles remain separated from another. The dense packing of the gold nanoparticles offers opportunities for exceptional optical- and electronic properties as well as the use of composite material for a potential, triggered destruction of the peptide beads by the typical radiation absorption effect of gold nanoparticles. The latter could, in particular, be useful to control the release of embedded payloads. The final chapter summarizes and discusses the achievements of this work. Further, it gives an overview of ongoing work and an outlook for worthwhile research from the present point of view. This includes e.g. the development of drug delivery applications, the use of the presented peptidic self-assembly system as template material in nanosciences, as well as the use of the material to investigate cellular uptake pathways of nano-sized objects.
