Three-dimensional Particle Tracking Velocimetry (3D-PTV) is a powerful and comprehensive measurement technique that provides instantaneous 3D volumetric velocity information and Lagrangian description of particle trajectories. This work is based on the 3D-PTV technique that was developed at the Institute of Geodesy and Photogrammetric at the Swiss Federal Institute of Technology, Zurich. Theoretical principles of the technique were studied based on geometric imaging and computer vision. A complete 3D-PTV system was designed, assembled, synchronized, and tested. A study of the accuracy of the 3D-PTV system in terms of the influence of each component was performed. 2D-PIV (Particle Image Velocimetry) measurements were done on the same experimental setup used in the 3D-PTV system. Results showed good quantitative agreement between the velocity information obtained with the two systems, which validates the results of the 3D-PTV measurement system. The developed 3D PTV system was applied to the study of mixing in a stirred tank showing that this technique has great potential for the study of low Reynolds number flows where the time-resolved trajectory of particles is important. Further improvement on the configurations and data analysis of the system can provide higher accuracy, higher spatial and temporal resolution, and less complexity, which will lead to an extension on its application.

Book excerpt: Microfluidics is the study of fluid flow at very small scales (micro -- one millionth of a meter) and is prevalent in many areas of science and engineering. Typical applications include lab-on-a-chip devices, microfluidic fuel cells, and DNA separation technologies. Many of these microfluidic devices rely on micron-resolution velocimetry measurements to improve microchannel design and characterize existing devices. Methods such as micro particle imaging velocimetry (microPIV) and micro particle tracking velocimetry (microPTV) are mature and established methods for characterization of steady 2D flow fields. Increasingly complex microdevices require techniques that measure unsteady and/or three dimensional velocity fields. This dissertation presents a method for three-dimensional velocimetry of unsteady microflows based on spinning disk confocal microscopy and depth scanning of a microvolume. High-speed 2D unsteady velocity fields are resolved by acquiring images of particle motion using a high-speed CMOS camera and confocal microscope. The confocal microscope spatially filters out of focus light using a rotating disk of pinholes placed in the imaging path, improving the ability of the system to resolve unsteady microPIV measurements by improving the image and correlation signal to noise ratio. For 3D3C measurements, a piezo-actuated objective positioner quickly scans the depth of the microvolume and collects 2D image slices, which are stacked into 3D images. Super resolution microPIV interrogates these 3D images using microPIV as a predictor field for tracking individual particles with microPTV. The 3D3C diagnostic is demonstrated by measuring a pressure driven flow in a three-dimensional expanding microchannel. The experimental velocimetry data acquired at 30 Hz with instantaneous spatial resolution of 4.5 by 4.5 by 4.5 microns agrees well with a computational model of the flow field. The technique allows for isosurface visualization of time resolved 3D3C particle motion and high spatial resolution velocity measurements without requiring a calibration step or reconstruction algorithms. Several applications are investigated, including 3D quantitative fluorescence imaging of isotachophoresis plugs advecting through a microchannel and the dynamics of reaction induced colloidal crystal deposition.

First, errors in velocity estimation can be reduced by improving the accuracy of the particle centre estimation by maximizing the spatial resolution of the image, with the constraint that it must contain the mixing layer. As the extent of the mixing layer is a function of its free stream velocity, channel width, and depth, the horizontal projection of mixing layers with 31 different combinations of flow parameters (width, depth, and free stream velocities) are studied to develop a model to predict the mixing layer extent as a function of the flow parameters. Secondly, an enhanced particle linking algorithm is proposed to avoid establishing incorrect particle trajectories representing atypical particle displacements. The algorithm uses an ellipsoid search region that varies in space and whose size is a function of the velocity at the position of the particle. The ellipsoid dimension is defined based on the ellipsoid confidence interval containing 99.75% of the velocity at the position of the particle. The velocity field at the surface of the mixing layer is measured using the PTV technique, and the mean, the root mean square, and the gradients of the velocity are analysed to understand the horizontal flow dynamics of the developing region of the shallow mixing layer. In addition, the vertical variation of the velocity is studied, using 3-D PTV, to characterize the vertical nonuniformity of the velocity of the developing mixing layer, and its consequence for particle dispersion"--

Of the various techniques currently used to visualize flow fields, particle image velocimetry (PIV) offers great advantages because of the high resolution quantitative results it provides. The feasibility of using synthetic aperture (SA) imaging to conduct 3D Particle Image Velocimetry (PIV) has been previously demonstrated. This thesis presents the development of a technique that extends SA imaging to 3D Particle Tracking Velocimetry (PTV), adding the ability to conduct Lagrangian studies on flow fields over time. A new method has been developed to accurately locate seeding particles in volumes reconstructed using SA imaging, based on a proposed thresholding and average intensity based scheme, which is able to locate particles in densely seeded flows. In addition, a new and much faster method for reconstructing volumes, based on a homography fit (HF) refocusing method, is proposed which facilitates rapid processing of large amounts of data recorded using high speed cameras for example. The capability of using SA imaging to conduct PTV is demonstrated by tracking located particles using the relaxation based algorithm. The developed technique provides accurate and high resolution PTV results at a much higher computation speed compared to other state of the art techniques. If engineered further,the presented technique has the potential to become the method of choice to conduct velocimetry.

Book excerpt: While most research on particle tracking velocimetry (PTV) is devoted either to 2D flows or to small scale 3D flows, this paper describes a complete 3D PTV algorithm and some applications to indoor airflow velocity measurement. A particle detection procedure specially adapted to the physical characteristics of neutrally buoyant helium filled soap bubbles is described. In particular, overlarge particle images are removed. Particle center are calculated by a weigh averaging method. Two temporal tracking schemes are presented and compared. The first one is based on fast normalized cross-correlation with Lagrangian extrapolation in image space to solve ambiguities. The second uses polynomial regression to find an estimated position and applies a quality criterion based on a minimization of changes in particle acceleration; to increase the measurement area and the number of trajectories, the correspondence problem is addressed by a new procedure involving fundamental matrices using both three and two 3D calibrated cameras. 3D triangulation is done by a least squares method. Some guidelines are given in terms of camera and light positioning for 3D PTV in large volumes with various wall colors. Applications of the algorithm include Lagrangian tracking: (i) in a light-gray walled 3.1mx3.1mx2.5m high test-room; (ii) inside a black walled 5.5mx3.7mx2.4m high test-room; (iii) over a heat source and inside; (iv) in experimental aircraft cabin; (v) in a 100cmx 100cmx100cm tank filled with an aqueous solution using continuous light and 10μm-large hollow particles. Results show that the algorithm is capable of tracking more than 1400 tracers in volumes up to 3mx3mx1.2m high.

Some existing mixed reality headsets include synchronized front-facing cameras that, among other things, can be used to track naturally occurring tracer particles (such as dust or snowflakes) to estimate particle velocity field in real time. The current work presents a 3D particle tracking velocimetry (PTV) method for use with MR systems, which combines various monocular cues to match particles between corresponding stereo images. Binocular disparity is used to estimate particle distance from an observer. Individual particles are tracked through time and used to construct the vector field of a scene. A digital display of velocity vectors can be broadcasted into a user's surrounding environment with the MR headset to be used as a flow visualization tool. The mixed reality particle tracking velocimetry (MR-PTV) approach was optimized to perform in natural conditions where particle size, particle color, and lighting are non-uniform. The approach was first tested using synthetic particle image data obtained by discrete element method simulations then experimentally validated for particles transported by a flume flow using the Microsoft HoloLens 2 MR headset. Uniform flow and flow around a body were considered experimentally. Experimental velocity measurements are compared to computational fluid dynamics results. The resulting MR-PTV system can be used for a variety of industry, scientific and recreational purposes for field-based measurement of particle velocities in real time.

Book excerpt: This dissertation describes a novel, low cost strategy of using particle streak (track) images for accurate micro-channel velocity field mapping. It is shown that 2-dimensional, 2-component fields can be efficiently obtained using the spatial variation of particle track lengths in micro-channels. The velocity field is a critical performance feature of many microfluidic devices. Since it is often the case that un-modeled micro-scale physics frustrates principled design methodologies, particle based velocity field estimation is an essential design and validation tool. Current technologies that achieve this goal use particle constellation correlation strategies and rely heavily on costly, high-speed imaging hardware. The proposed image/ video processing based method achieves comparable accuracy for fraction of the cost. In the context of micro-channel velocimetry, the usability of particle streaks has been poorly studied so far. Their use has remained restricted mostly to bulk flow measurements and occasional ad-hoc uses in microfluidics. A second look at the usability of particle streak lengths in this work reveals that they can be efficiently used, after approximately 15 years from their first use for micro-channel velocimetry. Particle tracks in steady, smooth microfluidic flows is mathematically modeled and a framework for using experimentally observed particle track lengths for local velocity field estimation is introduced here, followed by algorithm implementation and quantitative verification. Further, experimental considerations and image processing techniques that can facilitate the proposed methods are also discussed in this dissertation. Unavailability of benchmarked particle track image data motivated the implementation of a simulation framework with the capability to generate exposure time controlled particle track image sequence for velocity vector fields. This dissertation also describes this work and shows that arbitrary velocity fields designed in computational fluid dynamics software tools can be used to obtain such images. Apart from aiding gold-standard data generation, such images would find use for quick microfluidic flow field visualization and help improve device designs.
