Available in PDF, EPUB and Kindle. Book excerpt: The extracellular matrix (ECM) is an acellular three-dimensional network composed of proteins, glycoproteins, proteoglycans and exopolysaccharides. It primarily serves as a structural component in the tissues and organs of plants and animals, or forms biofilms in which bacterial cells are embedded. ECMs are highly dynamic structures that undergo continuous remodeling, and disruptions are frequently the result of pathological processes associated with severe diseases such as arteriosclerosis, neurodegenerative illness or cancer. In turn, bacterial biofilms are a source of concern for human health, as they are associated with resistance to antibiotics. Although exopolysaccharides are crucial for ECM formation and function, they have received considerably little attention to date. The respective chapters of this book comprehensively address such issues, and provide reviews on the structural, biochemical, molecular and biophysical properties of exopolysaccharides. These components are abundantly produced by virtually all taxa including bacteria, algae, plants, fungi, invertebrates and vertebrates. They include long unbranched homopolymers (cellulose, chitin/chitosan), linear copolymers (alginate, agarose), peptoglycans such as murein, heteropolymers like a variety of glycosaminoglycans (hyaluronan, dermatan, keratin, heparin, Pel), and branched heteropolymers such as pectin and hemicellulose. A separate chapter is dedicated to modern industrial and biomedical applications of exopolysaccharides and polysaccharide-based biocomposites. Their unique chemical, physical and mechanical properties have attracted considerable interest, inspired basic and applied research, and have already been harnessed to form structural biocomposite hybrids for tailor-made applications in regenerative medicine, bioengineering and biosensor design. Given its scope, this book provides a substantial source of basic and applied information for a wide range of scientists, as well as valuable textbook for graduate and advanced undergraduate students.

Available in PDF, EPUB and Kindle. Book excerpt: This book provides a state-of-the-art compendium on the role of proteoglycans and glycosaminoglycans during development and in cancer. It also suggests directions for novel therapeutic and biotechnological applications in stem cell biology. Proteoglycans and glycosaminoglycans, as part of the extracellular matrix, are multifunctional modulators of growth factor, cytokine, integrin and morphogen signaling, which determine both self-renewal, senescence and/or differentiation of stem cells during development. Since proteoglycans modulate cell adhesion and migration they are important organizers of the extracellular matrix within the proper stem cell niche. A malfunctioning of proteoglycans and glycosaminoglycans contributes to the cancer stem cell phenotype, which is linked to therapeutic resistance and recurrence in malignant disease. This book is essential reading for anyone interested in the extracellular matrix and its role in development. The series Biology of Extracellular Matrix is published in collaboration with the American Society for Matrix Biology.

Available in PDF, EPUB and Kindle. Book excerpt: Proteoglycans are some of the most elaborate macromolecules of mammalian and lower organisms. The covalent attachment of at least five types of glycosami- glycan side chains to more than forty individual protein cores makes these molecules quite complex and endows them with a multitude of biological functions. Proteoglycan Protocols offers a comprehensive and up-to-date collection of prepa- tive and analytical methods for the in-depth analysis of proteoglycans. Featuring st- by-step detailed protocols, this book will enable both novice and experienced researchers to isolate intact proteoglycans from tissues and cultured cells, to establish the composition of their carbohydrate moieties, to generate strategies for prokaryotic and eukaryotic expression, to utilize methods for the suppression of specific proteoglycan gene expression and for the detection of mutant cells and degradation products, and to study specific interactions between proteoglycans and extracellular matrix proteins as well as growth factors and their receptors. The readers will find concise, yet comprehensive techniques carefully drafted by leading experts in the field. Each chapter commences with a general Introduction, followed by a detailed Materials section, and an easy-to-follow Methods section. An asset of each chapter is the extensive notation that includes troubleshooting tips and practical considerations that are often lacking in formal methodology papers. The reader will find this section most valuable because it is clearly provided by experienced scientists who have first-hand knowledge of the techniques they outline. In addition, most of the chapters are well illustrated with examples of typical data generated with each method.

Book excerpt: This volume is a reference handbook focusing on diseases like Marfan syndrome, Ehlers-Danlos syndrome, Loeys-Dietz syndrome and other heritable soft connective tissue diseases. The book presents detailed information for both basic scientists and for clinicians seeing patients. It is also a stepping stone for new investigations and studies that goes beyond the facts about the composition and biochemistry of the connective tissue and extracellular matrix, as the authors connect individual components to specific aspects of various soft tissue disorders and to the actual or potential treatment of them. Progress in Heritable Soft Connective Tissue Diseases features very prominent physicians and scientists as contributors who bring their most recent discoveries to the benefit of readers. Their expertise will help clinicians with proper diagnosis of sometimes elusive and uncommon heritable diseases of soft connective tissues. This book also offers an update on the pathophysiology of these diseases, including an emphasis on unifying aspects such as connections between embryonic development of the different types of connective tissues and systems, and the role of TGF-beta in development and physiology of soft tissues. This new set of data explains, at least in part, why many of these disorders are interconnected, though the primary pathophysiological events, such as gene mutations, may be different for each disorder.

Available in PDF, EPUB and Kindle. Book excerpt: In the ten-year interval since the first edition of this volume went to press, our knowledge of extracellular matrix (ECM) function and structure has enor mously increased. Extracellular matrix and cell-matrix interaction are now routine topics in the meetings and annual reviews sponsored by cell biology societies. Research in molecular biology has so advanced the number of known matrix molecules and the topic of gene structure and regulation that we won dered how best to incorporate the new material. For example, we deliberated over the inclusion of chapters on molecular genetics. We decided that with judicious editing we could present the recent findings in molecular biology within the same cell biology framework that was used for the first edition, using three broad headings: what is extracellular matrix, how is it made, and what does it do for cells? Maintaining control over the review of literature on the subject of ECM was not always an easy task, but we felt it was essential to production of a highly readable volume, one compact enough to serve the the student as an introduction and the investigator as a quick update on graduate the important recent discoveries.
