Book excerpt: A link between inflammation and cancer has been established many years ago, yet it is only recently that the potential significance of this connection has become apparent. Although several examples of chronic inflammatory conditions, often induced by persistent irritation and/or infection, developing into cancer have been known for some time, there has been a notable resistance to contemplate the possibility that this association may apply in a causative way to other cancers. Examples for such progression from chronic inflammation to cancer are colon carcinoma developing with increased frequency in patients with ulcerative colitis, and the increased incidence of bladder cancer in patients suffering from chronic Schistosoma infection. Inflammation and cancer have been recognized to be linked in another context for many years, i.e., with regards to pathologies resembling chronic lacerations or 'wounds that do not heal.' More recently, the immunology of wound healing has given us clues as to the mechanistic link between inflammation and cancer, in as much as wounds and chronic inflammation turn off local cell-mediated immune responses and switch on growth factor release as well the growth of new blood vessels - angiogenesis. Both of these are features of most types of tumours, which suggest that tumours may require an immunologically shielded milieu and a growth factor-rich environment.

Book excerpt: The prevention of colorectal cancer is a subject of increasing medical importance, and there have been a number of promising recent developments. This book examines in detail important aspects of the current status and future prospects for chemoprevention of colorectal tumors. Research into the mechanisms that lead from early colorectal adenoma to colorectal cancer is discussed. The role and modes of action of available drugs such as celecoxib and sulindac are described, and recent data from aspirin trials are analyzed. In addition, the possible impact of nutritional agents with anti-inflammatory properties is considered, and strategies applicable in those with a high level of genetic risk are evaluated. An important feature of the book is its holistic perspective, making it relevant for gastroenterologists, internists, general practitioners, oncologists, colorectal and gastroenterological surgeons, and public health practitioners.

Available in PDF, EPUB and Kindle. Book excerpt: Cytokines, a broad class of proteins, have a role in fighting infection and in mounting an immune response; in mediating cell differentiation and embryogenesis; in promoting cell growth; and in inhibiting growth or causing cell death. Recent advances in cell culture, monoclonal antibodies and recombinant DNA technology have made it possible to isolate these proteins and to obtain sufficient quantities to conduct clinical studies as well as laboratory experiments. Thus, the foundations have been laid for a new form of cancer therapy and for understanding how abnormalities in the cytokine network could contribute to the process of malignancy. Up-to-date and comprehensive, this book brings together current research and discusses it in the context of its present and potential clinical application. The book is intended for cancer researchers, clinical oncologists, and those involved in the pharmaceutical and biological product industries.

It offers valuable insights into their diagnostic, prognostic, and therapeutic implications, making it a valuable resource for researchers, clinicians, and students interested in the field of cancer immunology and therapy. This book illustrates the importance and significance of the cytokine and chemokine signaling networks in tumor development and progression. It describes the complex networks mediated by cytokine and chemokine receptors promoting tumor cell proliferation, site-directed metastasis, and activation of angiogenic switch in tumor cells. The books also shed light on the heterogeneity of cytokines and chemokine in solid malignancies and their impact on tumor progression and therapeutic outcomes. The chapters provide current information about the types of cytokine-chemokine interactions in promoting cancer stem cell-like characteristics, epithelial to mesenchymal transition, and modulation of the tumor microenvironment. The significance of the complex interactions in cancer biology in the light of therapeutic resistance is also highlighted. The chapters also describe recent advancements in the therapeutic potential of targeting the pro-tumor cytokine and chemokine networks and limiting tumor cell metastasis. Finally, the book also provides a comprehensive yet representative description of a large number of challenges associated with targeting these vital chemokine-cytokine networks. Given its content, the book provides valuable information for researchers in the field of cancer biology and molecular medicine.

Book excerpt: Over the last decade, cytokine research has emerged as one of the most exciting and critical fields for providing fundamental knowledge of normal and abnormal human development. Today, it is apparent that cytokines orchestrate growth from the early embryonic stage to maturity and are responsible for the normal function of virtually every organ system. Furthermore, virtually all disease states have been associated, at least in part, with cytokine aberrations. In this volume, the editors have brought together internationally known experts in the field of interleukin research to provide a comprehensive review of the biology of the interleukins and their role in both health and illness, while maintaining a balance between the basic science and clinical aspects. Cytokines: Interleukins and their Receptors should be of interest to a wide variety of researchers including clinical hematologists, oncologists, immunologists, in addition to medical and PhD students and researchers with an interest in cytokines.

Available in PDF, EPUB and Kindle. Book excerpt: Genetic alterations in cancer, in addition to being the fundamental drivers of tumorigenesis, can give rise to a variety of metabolic adaptations that allow cancer cells to survive and proliferate in diverse tumor microenvironments. This metabolic flexibility is different from normal cellular metabolic processes and leads to heterogeneity in cancer metabolism within the same cancer type or even within the same tumor. In this book, we delve into the complexity and diversity of cancer metabolism, and highlight how understanding the heterogeneity of cancer metabolism is fundamental to the development of effective metabolism-based therapeutic strategies. Deciphering how cancer cells utilize various nutrient resources will enable clinicians and researchers to pair specific chemotherapeutic agents with patients who are most likely to respond with positive outcomes, allowing for more cost-effective and personalized cancer therapeutic strategies.

Available in PDF, EPUB and Kindle. Book excerpt: At the time of the first edition of Principles of Cancer Biotherapy in 1987, this book represented the first comprehensive textbook on biological therapy. In 1991, when the second edition was published, there was still some doubt on the part of many oncologists and cancer researchers as to the therapeutic value of these new approaches. By 2003 and the fourth edition, it was generally agreed that biopharmaceuticals were producing major opportunities for new cancer therapies. Cancer biotherapy has now truly matured into the fourth modality of cancer treatment. This fifth revised edition describes the tremendous progress that has been made in recent years using biologicals in cancer treatment. This book summarizes an evolving science and a rapidly changing medical practice in biotherapy. In this new millennium, it is now possible to envision a much more diversified system of cancer research and treatment that will afford greater opportunities for a patient’s personalized cancer treatment. This was first envisioned in the 1987 initial edition of this textbook and is now a “new” and popular approach to cancer treatment. Some forms of cancer biotherapy use the strategy of tumor stabilization and control through continued biological therapy, akin to the use of insulin in the treatment of diabetes. This textbook illustrates new methods of thinking and new strategies for control of cancer. It is always difficult to move from past dogma to future opportunity, but this fifth edition of Principles of Cancer Biotherapy illustrates why it is so important to the patients for researchers and clinicians to explore and quickly apply these new opportunities in cancer biotherapy.
