Available in PDF, EPUB and Kindle. Book excerpt: A better "casting" could not be conceived. The authors of this book are gold smiths on the subject. I have followed their work since their "entry" into cyto genetics and I have a high esteem for them. I consider it an honour to be asked to write the preface of their opus. Paul Popescu, Directeur de Recherche at INRA, has also played a promi nent part in the development of animal cytogenetics, especially in domestic animals. He is able to tell you the cost of a translocation in a pig breeding farm or a cow population: a fortune! P. Popescu has played a great part in gene mapping of these species using "in situ DNA hybridisation". His contributions are recognised world-wide. His laboratory receives many visitors every year and it serves as a reference for domestic animal cytogenetics. Helene Hayes, Charge de Recherche at INRA, has collaborated with P. POPESCU in the elaboration of the "at hand" techniques and in many other discoveries which are listed in her bibliography. She showed the fascinating correspondence between bovine and human chromosomes and the com pared gene maps of domestic bovidae.

Book excerpt: Decades before the recent advances in molecular biology and the knowledge of the complete nucleotide sequence of several genomes, cytogenetic analysis provided the first information concerning the genome organisation. The exploration of molecular biology techniques in the cytogenetic area represents a powerful tool for advancement in the construction of physical chromosome maps of the genomes. The most important advances in cytogenetics comes from the physical anchorage of genetic linkage maps in the chromosomes through the hybridisation of DNA markers onto chromosomes. This book presents and discusses current research in the study of animal genomes under the focus of cytogenetics.

Available in PDF, EPUB and Kindle. Book excerpt: The history of science is mostly written retrospec tively, a generation or two after the actual events being discussed. Science historians are now analyzing and evaluating the origins of evolutionary and genetical theory in the nineteenth century and a sort of "Darwin industry" seems to have grown up. A history of mammalian cytogenetics by one of the main participants is, hence, a very welcome change, since it has a vividness, an immediacy and a personal flavor which these scholarly tomes and the official biog raphies of scientists mostly lack. The life of the author, Chinese-born, T. C. Hsu, has been a romantic and color ful one, and he is himself a unique personality, so that his book is a very unusual blend of reminiscences, history of his special field (which has transformed human genetics) and wise comments on the mistakes made along the way. The best qualities of a very fine Chinese mind have contributed to Dr. Hsu's career, including this book. Those qualities (which seem to me especially Chinese) include a kind of transparent honesty, a very direct em pirical approach to problems and superb technical ability.

Available in PDF, EPUB and Kindle. Book excerpt: Chromosomes, as the genetic vehicles, provide the basic material for a large proportion of genetic investigations, from the construction of gene maps and models of chromosome organization, to the inves tigation of gene function and dysfunction. The study of chromosomes has developed in parallel with other aspects of molecular genetics, beginning with the first preparations of chromosomes from animal cells, through the development of banding techniques, which permitted the unequivocal identification of each chromosome in a karyotype, to the present analytical methods of molecular cytogenetics. Although some of these techniques have been in use for many years, and can be learned relatively easily, most published scientific reports—as a result of pressure on space from editors, and the response to that pressure by authors—contain little in the way of technical detail, and thus are rarely adequate for a researcher hoping to find all the necessary information to embark on a method from scratch. A new user needs not only a detailed description of the methods, but also some help with problem solving and sorting out the difficulties en countered in handling any biological system. This was the require ment to which the series Methods in Molecular Biology is addressed, and Chromosome Analysis Protocols forms a part of this series.

Available in PDF, EPUB and Kindle. Book excerpt: Cytogenetics is the study of chromosome morphology, structure, pathology, function, and behavior. The field has evolved to embrace molecular cytogenetic changes, now termed cytogenomics. Cytogeneticists utilize an assortment of procedures to investigate the full complement of chromosomes and/or a targeted region within a specific chromosome in metaphase or interphase. Tools include routine analysis of G-banded chromosomes, specialized stains that address specific chromosomal structures, and molecular probes, such as fluorescence in situ hybridization (FISH) and chromosome microarray analysis, which employ a variety of methods to highlight a region as small as a single, specific genetic sequence under investigation. The AGT Cytogenetics Laboratory Manual, Fourth Edition offers a comprehensive description of the diagnostic tests offered by the clinical laboratory and explains the science behind them. One of the most valuable assets is its rich compilation of laboratory-tested protocols currently being used in leading laboratories, along with practical advice for nearly every area of interest to cytogeneticists. In addition to covering essential topics that have been the backbone of cytogenetics for over 60 years, such as the basic components of a cell, use of a microscope, human tissue processing for cytogenetic analysis (prenatal, constitutional, and neoplastic), laboratory safety, and the mechanisms behind chromosome rearrangement and aneuploidy, this edition introduces new and expanded chapters by experts in the field. Some of these new topics include a unique collection of chromosome heteromorphisms; clinical examples of genomic imprinting; an example-driven overview of chromosomal microarray; mathematics specifically geared for the cytogeneticist; usage of ISCN’s cytogenetic language to describe chromosome changes; tips for laboratory management; examples of laboratory information systems; a collection of internet and library resources; and a special chapter on animal chromosomes for the research and zoo cytogeneticist. The range of topics is thus broad yet comprehensive, offering the student a resource that teaches the procedures performed in the cytogenetics laboratory environment, and the laboratory professional with a peer-reviewed reference that explores the basis of each of these procedures. This makes it a useful resource for researchers, clinicians, and lab professionals, as well as students in a university or medical school setting.

Available in PDF, EPUB and Kindle. Book excerpt: The present Special Issue refers to the last 30 years of contributions to the application of cytogenetics to domestic animals, as clearly reported in the five-paper review. In addition, eight original contributions regarding the cytogenetics of several animal species are included. In each contribution, several thematic areas and various cytogenetic techniques are proposed to better demonstrate the potentiality of this biological technology in the genetic improvement of livestock. Special attention was paid to the presence of chromosomal abnormalities in domestic animals and their effects on reproduction, suggesting the routine use of karyotype analyses on reproducers to drastically reduce the effects on reproduction and the economic losses encountered by breeders. In addition to classical cytogenetic analyses such as chromosome banding, the use of molecular cytogenetics, especially the FISH mapping technique, with both specific molecular markers (generally BAC clones) and chromosome painting probes were emphasized in all papers to better study the following: (a) Chromosomal abnormalities; (b) The chromosomal evolution of species; (c) Physical gene mapping in light of genomic sequences in the most important domestic species. Finally, a specific review is dedicated to molecular cytogenetics, in which examples of applied molecular techniques, including the FISH technique, comparative genomic hybridization array, and PCR-based method, are reported.

Book excerpt: This special topic issue on Veterinary Cytogenetics is dedicated to the Swedish scientist Ingemar Gustavsson, one of the pioneers of domestic animal cytogenetics, whose ground-breaking research has shaped this discipline for over 40 years. During his long and productive career he focused on chromosomes and their fundamental role in animal physiology, fertility, health and production in the context of agriculture and veterinary medicine, and he made important contributions to our understanding of mitosis, meiosis, chromosome integrity, and the transmission of chromosomal variation in domestic animals. This collection of reviews and original articles illustrates how the field of veterinary cytogenetics evolved from the study of the structure and number of chromosomes to modern molecular cytogenetics with its refined technology for research, diagnostic, prognostic or reproductive purposes. The goal of this publication is not to present an exhaustive volume covering each area of veterinary cytogenetics, but to give examples of specific subjects tackled by research groups working mainly in Europe and North America.
