Book Synopsis Development of In-Tether Carbon Chiral Center-Induced Helical Peptide by : Kuan Hu
Download or read book Development of In-Tether Carbon Chiral Center-Induced Helical Peptide written by Kuan Hu and published by Springer Nature. This book was released on 2021-01-26 with total page 113 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book focuses on the development of stapled peptides, a novel molecular modality used to regulate aberrant intracellular protein–protein interactions (PPIs). The author designs and presents a novel helical peptide stabilization methodology by constructing a chiral cross-linker moiety, namely “chiral center induced peptide helicity (CIH)”. The book demonstrates that a precisely positioned carbon chiral center on tether can decisively determine the secondary structure of a peptide, and that the R-configured peptide is helical, while the S-configured peptide is non-helical. Further, it reports that helicity-enhanced R isomer peptides displayed significantly enhanced cell permeability and target binding affinity, as well as tumor inhibition efficiency, in comparison to S isomer peptides. The book will not only advance readers’ understanding of the basic principle of stapled peptides, but also accelerate the clinical transformation of stapled peptide drugs.

Available in PDF, EPUB and Kindle. Book excerpt: A successful seientific career requires constant effort on the part of the seientist. If such a career is to be achieved by seientists in developing countries where the faeilities are not easily available, even more dedication is required. Dr. K. M. Sivanandaiah, having completed a successful career in the field of peptide chemistry, is such a seientist. Being weIl aware of the limited research facilities available in India, we are extraordinarily appreciative of Professor Sivanandaiah's dedication to the advancement of science. As a Professor of Chemistry and, until his retirement in 1992, the Head of the Department of Studies in Chemistry at Bangalore University, India, he not only devoted much of his time to his students, teaching basic organic chemistry, but was also able to contribute to the field of peptide chemistry. After completing 25 years of service at Central College, Bangalore, where he was admired as an outstanding teacher, Professor Sivanandaiah is still active in research. He is now Professor Emeritus and continues to contribute to the field of peptides. As former students of Professor Sivanandaiah, we felt that publishing a book containing articles related to the design, synthesis, conformation, and biological activity of peptides and written by eminent scientists in the field of peptide research would be a fitting tribute to his role in the field of bioorganic chemistry.

Book Synopsis Inhibitors of Protein–Protein Interactions by : Ali Tavassoli
Download or read book Inhibitors of Protein–Protein Interactions written by Ali Tavassoli and published by Royal Society of Chemistry. This book was released on 2020-12-07 with total page 357 pages. Available in PDF, EPUB and Kindle. Book excerpt: Protein-protein interactions (PPI) are at the heart of the majority of cellular processes, and are frequently dysregulated or usurped in disease. Given this central role, the inhibition of PPIs has been of significant interest as a means of treating a wide variety of diseases. However, there are inherent challenges in developing molecules capable of disrupting the relatively featureless and large interfacial areas involved. Despite this, there have been a number of successes in this field in recent years using both traditional drug discovery approaches and innovative, interdisciplinary strategies using novel chemical scaffolds. This book comprehensively covers the various aspects of PPI inhibition, encompassing small molecules, peptidomimetics, cyclic peptides, stapled peptides and macrocycles. Illustrated throughout with successful case studies, this book provides a holistic, cutting-edge view of the subject area and is ideal for chemical biologists and medicinal chemists interested in developing PPI inhibitors.
Book Synopsis Bifunctional Helical Peptide Catalysts for Enzyme-like Reactivity and Selectivity and Selective Stapling of Natural Amino Acid Residues with Hydrophilic Squaric Acid Derivatives by : Michael James Kinghorn
Download or read book Bifunctional Helical Peptide Catalysts for Enzyme-like Reactivity and Selectivity and Selective Stapling of Natural Amino Acid Residues with Hydrophilic Squaric Acid Derivatives written by Michael James Kinghorn and published by . This book was released on 2019 with total page 154 pages. Available in PDF, EPUB and Kindle. Book excerpt: Studies were also conducted into the stabilization of peptide secondary structure via covalent linking of nucleophilic amino acid side chains with squaric acid residues. Under mild conditions, stapling of nitrogen, sulfur and oxygen residues can readily be achieved in either organic or aqueous media. Squaric acid staples display pH selectivity for specific side chains and selective removal of diester staples (diserine staple) is demonstrated with methylamine. This new method for peptide stapling is shown to dramatically increase the proteolytic stability of eIF4E cancer inhibitor proteins, which typically are prone to quick degradation. Tyrosidine and RGD peptide analogues were synthesized and cyclized on resin in order to provide a new pathway to macrocyclization of antibacterial and integrin binding cyclic peptides.
Book Synopsis Peptide Materials by : Carlos Aleman
Download or read book Peptide Materials written by Carlos Aleman and published by John Wiley & Sons. This book was released on 2013-03-29 with total page 479 pages. Available in PDF, EPUB and Kindle. Book excerpt: Peptides are the building blocks of the natural world; with varied sequences and structures, they enrich materials producing more complex shapes, scaffolds and chemical properties with tailorable functionality. Essentially based on self-assembly and self-organization and mimicking the strategies that occur in Nature, peptide materials have been developed to accomplish certain functions such as the creation of specific secondary structures (a- or 310-helices, b-turns, b-sheets, coiled coils) or biocompatible surfaces with predetermined properties. They also play a key role in the generation of hybrid materials e.g. as peptide-inorganic biomineralized systems and peptide/polymer conjugates, producing smart materials for imaging, bioelectronics, biosensing and molecular recognition applications. Organized into four sections, the book covers the fundamentals of peptide materials, peptide nanostructures, peptide conjugates and hybrid nanomaterials, and applications with chapters including: Properties of peptide scaffolds in solution and on solid substrates Nanostructures, peptide assembly, and peptide nanostructure design Soft spherical structures obtained from amphiphilic peptides and peptide-polymer hybrids Functionalization of carbon nanotubes with peptides Adsorption of peptides on metal and oxide surfaces Peptide applications including tissue engineering, molecular switches, peptide drugs and drug delivery Peptide Materials: From Nanostructures to Applications gives a truly interdisciplinary review, and should appeal to graduate students and researchers in the fields of materials science, nanotechnology, biomedicine and engineering as well as researchers in biomaterials and bio-inspired smart materials.

Topics covered include the chemistry of unnatural amino acids, peptide- and scaffold-based peptidomimetics, amino acid-side chain isosteres, backbone isosteres, dipeptide isosteres, beta-turn peptidomimetics, proline-mimetics as turn inducers, cyclic scaffolds, amino acid surrogates, and scaffolds for combinatorial chemistry of peptidomimetics. Case studies in the hit-to-lead process, such as the development of integrin ligands and thrombin inhibitors, illustrate the successful application of peptidomimetics in drug discovery.
