Book excerpt: Nearly 97% of the human genome is the non-coding DNA, which varies from one species to another, and changes in these sequences are frequently noticed to manifest clinical and circumstantial malfunction. Numerous non-protein-coding genes are recently found to encode microRNAs, which are responsible for RNA-mediated gene silencing through RNA interference (RNAi)-like pathways. MicroRNAs (miRNAs), small single-stranded 17–25 nucleotide RNAs capable of interfering with intracellular messenger RNAs (mRNAs) that contain either complete or partial complementarity, are useful for the design of new therapies against cancer polymorphism and viral mutation. Currently over 1000 native miRNA species found in vertebrates and many more new miRNA homologs continue to be identified; however, most of their functions remain to be determined. In this book, many new perspectives of the miRNA research are reviewed and discussed, including their roles in stem cell maintenance, embryonic development, tissue differentiation, adult physiology, disease pathology, cancer research, viral infection, genetic engineering in plants, and utility in cosmetic applications. These new findings may not only provide significant insight into the various mechanisms of miRNAs but also offer a great opportunity in developing new therapeutic interventions.

Available in PDF, EPUB and Kindle. Book excerpt: MicroRNAs (miRNAs), endogenous noncoding regulatory mRNAs of around 22-nucleotides long, have rapidly emerged as one of the key governors of the gene expression regulatory program in cells of varying species, with ever-increasing implications in the control of the fundamental biological processes and in the pathogenesis of adult humans. The exciting findings in this field have inspired us with a premise and a promise that miRNAs will ultimately be taken to the heart for therapy of human disease. While miRNAs have been considered potential therapeutic targets for disease treatment, it remains obscured what strategies we can use to achieve the goal. In the past years, we have witnessed a rapid evolving of many creative, innovative, inventive strategies and methodologies pertinent to miRNA research and applications. These technologies have convincingly demonstrated their efficacy and reliability in producing gain-of-function or loss-of-function of miRNAs through targeting miRNA expression/biogenesis/function, providing new tools for elucidating miRNA functions and opening up a new avenue for the development of new agents targeting miRNAs for therapeutic aims. The present book provides comprehensive descriptions of these technologies and their applications to miRNA research and to new drug design for miRNA-related diseases. It starts with an overview of up-to-date knowledge of miRNA biology and the potential of miRNAs as therapeutic targets for human disease, followed by an introduction of the new concept of miRNA interference (miRNAi) and the perspectives of miRNAi technologies in general terms. In the following, each chapter introduces one of the miRNAi technologies with detailed descriptions of state-of-the-art design, procedures, principles and applications to basic research, R and D and clinical therapy.

Available in PDF, EPUB and Kindle. Book excerpt: MicroRNA (miRNA) refers to a small single-stranded non-coding RNA molecule. It can be found in animals, plants and some viruses. miRNAs function in post-transcriptional regulation of gene expression and RNA silencing. They work through base-pairing along with complementary sequences in mRNA molecules. Several mammalian cell types contain a high concentration of miRNAs as circulating in extracellular fluid, which are released into body fluids such as blood and cerebrospinal fluid, and could be used as biomarkers in a variety of disorders. MiRNAs play a role in gene regulation and can also produce histone modifications and DNA methylation at promoter sites, which can affect the target gene expression. MiRNAs are important in modulating complex enzymatic cascades, such as the hemostatic blood coagulation system. The topics included in this book on microRNAs are of utmost significance and bound to provide incredible insights to the readers. Researchers and students engaged in the study of this molecule will be assisted by it.

Available in PDF, EPUB and Kindle. Book excerpt: miRNAs are a class of endogenous, small non-protein coding RNA molecules (~ 22 nucleotides) which are novel post-transcriptional regulators of gene expression. Since we have hundreds of miRNAs, the major challenge is now to understand their specific biological function. In fact the experimental evidence suggests that signaling pathways could be ideal candidates for miRNA-mediated regulation. Several studies suggest that miRNAs affect the responsiveness of cells to signaling molecules such as WNT, Notch, TGF-β and EGFR. Altered expression of particular miRNAs has been implicated in the onset and development of cancer and could be used as potential biomarkers for the disease. Recently, many studies have found miRNAs have crucial regulatory roles in Cancer stem cells (CSCs) a kind of tumor initiating cells (TICs) and dormancy. Findings also suggest that DNA methylation may be important in regulating the expression of many miRNAs in several cancer initiating cells. Several miRNAs are known to either upregulated or downregulated in CSCs when compared to non-cancerous cells from the same tissues. CSCs are a small subpopulation of cells identified in a variety of tumors and involve in self-renewal, differentiation, chemoresistance and tumorigenesis. The volume will give a comprehensive account of important advancements in the area of miRNAs and cancer.

Book excerpt: Written with biologists, biochemists and other molecular scientists in mind, this volume meets the long-felt need for a textbook dedicated to the topic and recreates the excitement surrounding the scientific revolution sparked by the discovery of RNA interference in 1998. Students and instructors alike will profit from the author's exclusive first-hand knowledge, drawing on his breakthrough discoveries at the Tuschl lab at Rockefeller University. Gunter Meister abandons the traditionalist treatment of nucleic acids found in most biochemistry and molecular biology texts, adopting instead a modern approach in both concept and scope. The text is divided into three parts, on mRNA, non-coding RNA, and RNomics, and the author addresses the traditional roles of RNA in the transmission and regulation of genetic information, as well as the recently discovered functions of small RNA species in pathogen defense, cell differentiation and higher-level genomic regulation. All set to become the standard for teaching molecular science to biologists and biochemists.

Book excerpt: microRNAs (miRNAs) are small non-coding RNAs that regulate various biological phenomena, such as development and homeostasis. The dysregulation of miRNA leads to disease progression, particularly of cancer. In Circulating MicroRNAs: Methods and Protocols, expert researchers in the field detail recent advances in the isolation, purification and analysis of circulating miRNAs from a variety of sources for research. The book is divided into three main topics. The first section involves the study of secretory miRNAs in cell-cell communication, and the second, the study of circulating miRNAs in body fluids. The last describes the novel techniques used to study circulating miRNAs. Written in the highly successful Methods in Molecular BiologyTM series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and key tips on troubleshooting and avoiding known pitfalls. Authoritative and practical, Circulating MicroRNAs: Methods and Protocols seeks to aid scientists in dealing with the recent advances of RNAi technology from the bench to the bedside.
