Book excerpt: This book describes every manner in which collagen is involved in normal anddisease-altered states of the various organs and systems. In the first part of the book the biochemical aspects of collagens are reviewed, including their structure, heterogeneity, syntheses, and degradation. The main part focuses on the involvement of collagen in bone, cartilage, tendon, muscle, heart, vessels, lungs, liver, skin, eye, ear, teeth, periodontal tissues, kidneys and reproductive, hemopoietic, and nervous systems. The influence of radiation and nutrition on collagen, the role of collagen in neoplasms, the hormonal control of its metabolism, immunobiology and the pharmacology of collagen are also described. The most important feature of the book is the comprehensive review of the medical aspects of collagen, from those known in detail to those only hypothesized including hereditary disorders affecting collagen and so-called collagen diseases. Each chapter reviewsknown or possible mechanisms of collagen involvement and changes in indices of collagen which can be measured in clinical practice to monitor these phenomena. The fact that collagen is involved into the pathophysiology of almost all organs and body systems means that physicians in almost all branches of medicine will find this book of great interest.

This comprehensive work also includes areas of the medical field in which the basic aspects could be applied. It explains that both cells and matrix are altered in disease states because of the strong interactions established between cells and the extracellular matrix. The aim of this book is to close the existing gap between basic scientists and clinical investigators. This reference is an absolute must for all biological chemists, clinical investigators, and pathologists. Students of these professions will find this reading both informative and useful as well.

Book excerpt: connective tissues are essential for the physical functioning of the animals's body. The condition of the various connective tissues is governed by biochemical factors, anabolism and catabolism, that are controlled by specific enzymes. Any change outside the normal range of metabolism, for instance induced by immunological reactions, may induce a pathological disturbance. The result can be acute or chronic inflammation, or loss of normal function, expressed in loosening, dilatation, breaking, wear, stiffness, shrinking, scars, stenosis, and cirrhosis or any other kind of fibrosis. A first step toward improving our understanding of the feedback mecha nism that maintains the biological status and texture of a given connective tissue is to combine what is known about synthesis and enzymatic degradation of the components of fibers and ground substance. Common pathological phenomena like chronic inflammation of immune reactions can be either the result of the cause of disturbances in the sensitive balance of connective tissue metabolism. Nowadays con nective tissues are less and less regarded as brady trophic tissue but rather as a stimulating and many-sided problem of research. Before we can understand the pathogenesis of the connective tissue diseases that result in the destructive processes mentioned above, basic research will be necessary. This research will be furthered by a constant exchange of information and the results of· observations. To promote this exchange of information between scientists, symposia on connective tissue research are organized at regular intervals.

This book reviews the data concerning the primary structure of collagen and explores the physical features of elastin. Organized into six chapters, this volume begins with an overview of the amino acid sequence of collagen. This text then examines the relationship of the structure and function of elastic tissues and reviews the usefulness of immunological methods when applied to structural studies. Other chapters consider the substantial progress in the elucidation of the antigen determinants of the collagen molecule. This book discusses as well the immunological properties of collagen that may have considerable bearing on the causes of some connective tissue diseases. The final chapter deals with the extensive investigations into glycoproteins in various connective tissues of various species at various ages. This book is a valuable resource for biochemists and physicians.

Available in PDF, EPUB and Kindle. Book excerpt: This book aims at providing insights into the collagen superfamily and the remarkable diversity of collagen function within the extracellular matrix. Additionally, the mechanisms underlying collagen-related diseases such as dystrophic epidermolysis bullosa, osteogenesis imperfecta, as well as collagen-related myopathies and neurological disorders are discussed. Collagens are the most abundant extracellular matrix proteins in organisms. Their primary function is to provide structural support and strength to cells and to maintain biomechanical integrity of tissues. However, collagens can no longer be considered just as structural proteins. They can act as extracellular modulators of signaling events and serve critical regulatory roles in various cell functions during embryonic development and adult homeostasis. Furthermore, collagens are associated with a broad spectrum of heritability-related diseases known as “collagenopathies” that affect a multitude of organs and tissues including sensorial organs. The book is a useful introduction to the field for junior scientists, interested in extracellular matrix research. It is also an interesting read for advanced scientists and clinicians working on collagens and collagenopathies, giving them a broader view of the field beyond their area of specialization.

Book excerpt: This volume is a reference handbook focusing on diseases like Marfan syndrome, Ehlers-Danlos syndrome, Loeys-Dietz syndrome and other heritable soft connective tissue diseases. The book presents detailed information for both basic scientists and for clinicians seeing patients. It is also a stepping stone for new investigations and studies that goes beyond the facts about the composition and biochemistry of the connective tissue and extracellular matrix, as the authors connect individual components to specific aspects of various soft tissue disorders and to the actual or potential treatment of them. Progress in Heritable Soft Connective Tissue Diseases features very prominent physicians and scientists as contributors who bring their most recent discoveries to the benefit of readers. Their expertise will help clinicians with proper diagnosis of sometimes elusive and uncommon heritable diseases of soft connective tissues. This book also offers an update on the pathophysiology of these diseases, including an emphasis on unifying aspects such as connections between embryonic development of the different types of connective tissues and systems, and the role of TGF-beta in development and physiology of soft tissues. This new set of data explains, at least in part, why many of these disorders are interconnected, though the primary pathophysiological events, such as gene mutations, may be different for each disorder.

Available in PDF, EPUB and Kindle. Book excerpt: International Review of Connective Tissue Research, Volume 8, provides an overview of the state of knowledge in connective tissue research. The details of the structure of collagen and its biosynthesis are now well established, and attention has focused on the finer details of the biochemical mechanisms involved. The knowledge achieved so far is also being applied to studies of various physiological, developmental, and pathological mechanisms involving the connective tissues. These trends are reflected in the subject covered by this volume. The enzymes involved in the posttranslation modifications of collagen are being intensively investigated. The book deals with three of these: lysyl oxidase, the enzyme involved in the first stage of cross-link formation; and the two glycosyl transferases involved in the glycosylation of hydroxylysome residues. New collagen types are reviewed together with recent work on collagen messenger RNA. Both of these topics are important to the understanding of the control of collagen biosynthesis. The involvement of connective tissues in physiological and pathological processes is discussed in three areas. These are interaction of collagen with platelets in hemostasis; the biosynthesis of connective tissue components by smooth muscle cells; and biochemical research on the intervertebral disc, the disease of which is behind the majority of back pain problems.

Available in PDF, EPUB and Kindle. Book excerpt: In the ten-year interval since the first edition of this volume went to press, our knowledge of extracellular matrix (ECM) function and structure has enor mously increased. Extracellular matrix and cell-matrix interaction are now routine topics in the meetings and annual reviews sponsored by cell biology societies. Research in molecular biology has so advanced the number of known matrix molecules and the topic of gene structure and regulation that we won dered how best to incorporate the new material. For example, we deliberated over the inclusion of chapters on molecular genetics. We decided that with judicious editing we could present the recent findings in molecular biology within the same cell biology framework that was used for the first edition, using three broad headings: what is extracellular matrix, how is it made, and what does it do for cells? Maintaining control over the review of literature on the subject of ECM was not always an easy task, but we felt it was essential to production of a highly readable volume, one compact enough to serve the the student as an introduction and the investigator as a quick update on graduate the important recent discoveries.
