Book excerpt: This dissertation described preparation, characterization and intracellular localization of peptide-modified gold nanoparticles as multifunctional nuclear targeting vectors. The vectors have the potential advantages for intracellular diagnostics and therapeutic delivery. Gold nanoparticles were used in this study as carriers to combine multiple-functional cell-translocating peptides and antisense oligonucleotides. Two proteins, bovine serum albumin (BSA) and streptavidin (SA), have been systematically studied for crosslinking peptides or oligonucleotides with gold nanoparticles. Protocols for modifying gold nanoparticles with BSA-peptide conjugates are described within. The resulting constructs were characterized using a number of techniques including UV-visible absorbance spectroscopy and fluorescence spectroscopy in order to quantify peptide-BSA binding isotherms, exchange rates, critical flocculation concentrations, and the composition of mixed peptide-BSA monolayers on gold nanoparticles. The abilities of BSA-peptide/gold complexes to target the nucleus of several cell lines (HeLa, 3T3-NIH and HepG2) were explored. Four peptide/nanoparticle complexes were investigated, particles modified with the adenovirus nuclear localization signal (NLS), the adenovirus receptor-mediated endocytosis (RME) peptide, the adenovirus fiber containing the RME and NLS and the NLS for SV 40. The translocation of the gold nanoparticles was identified by video-enhanced color differential interference contrast (VEC-DIC) microscopy. The strong affinity between SA and biotin was employed to conjugate SA with biotinylated peptide and oligonucleotide. Systematical studies have been done to characterize the complexes, including SA adsorption on nanoparticles, biotinylated DNA adsorption on SA-coated nanoparticles, stabilities in solution, exchange rate in cell growth media and the composition of mixed SA-biotin peptide and SA-biotin DNA monolayers on gold nanoparticles. Synthesized materials were characterized using an orthogonal approach that included dynamic light scattering (DLS), UV-Vis absorbance (UV-Vis), transmission electron microscopy (TEM), atomic force microscope (AFM), and thermogravimetric analysis (TGA). Very importantly, the end-product in this study demonstrated excellent colloidal stability in multiple aqueous environments including over a wide pH and temperature range of physiological relevance. The product also exhibited long-term storage stability under ambient conditions. Furthermore, the cytotoxicity of Au-PEI@NIST was investigated using the CHO K1 cell line and exhibited dose proportional toxicity, which suggested potential for use as a positive control in nanotoxicology studies. The results and stringent testing regimen point toward an effective and sustainable approach that can serve as a clarified starting point for the development of more complex conjugates for nanomedicine area.

Book excerpt: Biocompatible gold nanoparticles have gained considerable attention in recent years for potential applications in nanomedicine due to their interesting size dependent chemical, electronic and optical properties. In particular, the prospective use of gold nanoparticles as contrast enhancement agents in X-ray Computed Tomography (CT) and Photo Acoustic Tomography for early diagnosis of specific tumors is being extensively researched. Additionally, gold nanoparticles show promise in enhancing the effectiveness of various targeted cancer treatments such as radiotherapy and photothermal therapy. For these applications, biocompatible gold nanoparticles labeled with specific tumor targeting biomolecules are needed for site specific delivery. In the present project, gold nanoparticles stabilized and labeled with carbohydrate (starch) and glycoprotein (gum arabic) have been generated, characterized and tested for in vitro and in vivo stability. They are found to localize in specific tissues in the animal models. Additionally, gold nanoparticles labeled with a cancer seeking peptide, bombesin, exhibited excellent binding affinity towards prostate and breast cancer cells. The degree of contrast enhancement in cancer imaging or effectiveness of cancer treatments is limited by the number of nanoparticles that can be localized at the target tumor/cancer site. One way to augment the localization of nanoparticles at the target tissue is to utilize gold nanochains that hold more number of nanoparticles. Therefore, we developed biocompatible gold nanochains formed by self assembly of nanoparticles on gum arabic and they were shown to be in vitro stable. The change of optical properties of gold nanoparticles upon slight modification of the surrounding environment is the basis for the development of biosensors. Therefore, Surface Enhanced Raman Scattering (SERS), a spectroscopic method where the Raman scattering signal, which is sensitive to the molecular structure, is enhanced in the presence of gold nanoparticles has emerged as a powerful tool for the detection of specific molecules. Consequently, there is need for nanostructures that give maximum SERS signal. In the present project, gold nanoparticles set in agarose gel have been demonstrated to be excellent SERS substrates compared to commercially available gold nanoparticles for DNA nucleosides.

Book excerpt: Metal nanoparticles (NPs) are valued in fields including biomedical imaging, chemical sensing, catalysis, and fundamental physics. Their utility often stems from their unique optical and electronic properties, but their broad applicability is due to their robust surface chemistry that allows a wide variety of ligands to be bound to them to tune stability, solubility, and reactivity. However, there has been little work focusing on linking these two aspects of nanoparticles, despite their importance. The lack of work, in part, stems from the significant challenges arising from the inherent dispersity within nanoparticle populations and complicated characterization techniques in which sample measurement can be difficult or provide limited information. The focus of this thesis is to refine the understanding of the structure-property relationship in gold nanosystems by improving the characterization of gold surfaces and by controlling gold nanoparticle properties through surface chemistry. This thesis reports the characterization of the gold-thiol surface, where thermal treatment of gold nanoparticles (AuNPs) volatilized gold and sulfur, which was collected and subsequently quantified by inductively coupled plasma mass spectrometry and atomic emission spectroscopy. It was found that the mass loss in these samples ranged from 0-30\% gold by mass, rather than purely from the thiol ligands, and that the level of gold desorption is related to the Au--S bond strength.This study calls into question the usefulness of thermogravimetric analysis as a characterization technique of gold nanosytems. Additionally, the influence of surface chemistry on the electronic structure of gold nanosystems has been probed using magnetic spectroscopies. First, electron spin resonance has been used to examine the perturbation to the superatomic structure of Au$_{25}$(SR)$_#x18;$ nanoclusters (NCs) by a series of alkanethiolate ligands in hexane and tetrahydrofuran solvents. It has been found that the nanocluster electronic is sensitive to surface chemistry, with the $g_x$ component of the electronic \textit{g}-factor shifting linearly towards the free electron \textit{g}-value, $g_e$, and the perturbation is large, given the minute changes in ligand identity. The changes are dissimilar to those seen in gold nanoparticles with similar surface chemistry, indicating that use of AuNCs as model systems for larger, metallic AuNPs should be reevaluated. Finally, Evans Method nuclear magnetic resonance spectroscopy has been used to probe the density of states (DOS) of gold nanoparticles protected by aromatic thiolate ligands. Aromatic ligands are a common tool for tuning the electronic structure of inorganic complexes, and this thesis reports their capability to alter the electronic structure of gold nanoparticles. The DOS of these nanoparticles increases with increasing electron withdrawing character of the aromatic ligands, and it has been found that trends in controlling the gold nanoparticle's DOS found with aromatic thiols cannot necessarily be compared to those found with aliphatic thiols. This work examines not only how surface chemistry influences the properties of nanosystems, but also the usefulness of model systems and naive assumptions about characterization techniques.

Laser desorption/ionization mass spectrometry (LDI-MS) has been used to monitor AuNPs in complex biological samples but there are still many ways in which AuNPs can be used with MS. In this dissertation, the use of AuNPs to assist in analyte ionization has been investigated. Their ability to enhance signal from biomolecules based on their surface chemistry, size, aggregation and method of deposition has been studied. The first use of an inkjet printer to create surfaces from which analytes can be sampled is discussed and revealed that the aggregation of the NPs is crucial to analyte enhancement. In addition, the investigation of the use of both AuNPs and a small amount of matrix that is typically used in matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) in order to detect previously difficult to ionize ligands in biological samples is reported and revealed the existence of a synergistic relationship between the two components. The method by which AuNPs dry and form a coffee ring is also taken advantage of to easily locate and enhance signal from larger biomolecules due to the concentration of the analyte, AuNP and matrix to the formed ring. Finally, desorption efficiencies and energies required for these ionization processes are all investigated and compared in order to better understand the mechanism of ionization.

An electrochemical technique Chronocoulometry (CC) quantified the surface coverage of ssDNA on gold using a redox label. The issues that affected the hybridization efficiency of DNA on the gold surface were analyzed and parameters for optimum hybridization were established by the same techniques used to detect ssDNA on the surface. Though gold is a well-known surface, the thiol chemistry, which is the basis of attaching DNA on the surface, is not robust and reproducible due to the desorption of probes from the surface during hybridization, which involves elevating the temperature. Hybridization yield on gold surfaces could not be determined accurately as the desorption of probe from the surface was a competing phenomenon during hybridization event. As a result, new substrates were analyzed and indium-doped tin oxide (ITO) was found to be a very useful substrate because of the ease with which it can be modified with COOH, Phosphates, Phosphonates and silanes. In addition, the physical properties of ITO make it an attractive candidate to use in creating bioarrays. It is highly conductive and tansparent in the visible region and reflective in the IR. Compared to the thiol chemistry on gold the phosphates are much stronger and resistant to elevated temperatures used for hybridization. ITO electrodes can sustain relatively high potentials required to oxidize gold nanoparticles used as labels to detect hybridization. All these properties can be utilized to characterize ssDNA modified ITO surfaces and surface DNA hybridization either by optical or electrical methods. 12-Phosphonododecanoic acid (12-PDA) formed a well-ordered monolayer on the ITO.
