This dissertation describes the optimization and characterization of self-assembled monolayer (SAM) multilayer SERS substrates for improving performance of different types of substrates. SERS substrates derived from metal film on nanostructures were modified with multiple metal films interspaced with SAM dielectric spacers to achieve multilayered SERS substrates. The fundamental concept of this substrate geometry exploited the cumulative effect of the multiple electromagnetic fields and various properties of SAMs to optimize SERS enhancement of multilayer SERS substrates to about 20-fold compared to conventional substrates.

Book excerpt: Surface enhanced Raman spectroscopy (SERS) is a highly sensitive technique for quantifying trace amounts of analyte adsorbed at a roughened metal surface. We have shown that novel gold nanostructured films, simply fabricated using gold nanoparticles and latex microspheres, can be used as highly sensitive SERS substrates. The films are templated by layers of colloidal crystals and display long ranged ordered regions, and porosity on two length scales that results in high surface area. By integrating these substrates into a flow chamber we have obtained consistent reproducible data and demonstrated the quantitative detection of sodium cyanide in the concentration range from 5 to 500 ppb, with detection significantly enhanced by lowering the pH after cyanide adsorption. We have found that the optimum size of the latex microspheres is approximately equal to the wavelength of the laser. Finally, the SERS activity of the gold substrates coated with silver nanoparticles was investigated using methyl phosphonic acid (MPA). Detection of MPA was dependent on the time between substrate coating and the SERS measurement. These studies show that novel materials formed by simple wet chemistry techniques can be used efficiently in practical devices for the detection of chemical agents.

An interesting example was provided by pyromellitic diimide (PMDI), a model compound for polyimides, and silver substrates. Normal Raman spectra of PMDI were characterized by strong bands near 1772, 1370, 760, and 655 1/cm and medium intensity bands near 1630 and 1205 1/cm were weak in SERS spectra obtained from films of PMDI spin-coated onto silver island films while those near 1370 and 1205 1/cm and a new band near 685 1/cm were strongest. Differences between the normal Raman and SERS spectra were related to orientation effects indicating that PMDI was absorbed onto silver with a vertical conformation in which the rings were perpendicular to the surface and one imide group was in contact with the surface. This is evident by the number of SERS publications in the last decade, from 580 in 2005 to almost 2000 in 2014. There are now companies worldwide that are dedicated to manufacturing SERS substrate, sensors, and other SERS-based products. The miniaturization of Raman instrumentation from bulky and expensive bench-top designs to inexpensive handheld spectrometers has allowed SERS to be employed as an in situ detection technique. Some examples of the industrial success of Raman and SERS are rapid raw material identification (RMID), Process Analytical Technology (PAT) and drug screening in pharma and biotech industries, continuous surveillance of food, water, and environmental safety, and the diagnosis and continuous monitoring of diseases like cancer, diabetes, etc. This progress in translating Raman and SERS techniques into commercial use and commercial products is possible because of the close collaboration between scientists and engineers. However, the weak signals of Raman spectroscopy is an inherent limitation of the spontaneous Raman spectroscopy technique. Therefore, several modifications have been made, including but not limited to, surface-enhanced Raman spectroscopy (SERS), spatially offset Raman spectroscopy (SORS), transmission Raman spectroscopy (TRS), coherent anti-Stokes Raman spectroscopy (CARS), and tip-enhanced Raman spectroscopy (TERS). Spontaneous Raman spectroscopy has become a one-of-a kind, portable, and affordable technology for in-situ detection and continuous monitoring in resource-limited-settings. The primary focus of this textbook is to try to cover the fundamentals in SERS methods of preparing substrates and sensors, and efficient spectral analysis approaches like chemometrics for diverse applications, listed in the paragraph above. However, with a goal to give a true picture to the audience when assessing the scope of spontaneous Raman and Surface-enhanced Raman for specific applications, we have briefly introduced industrially successful examples of other types of Raman spectroscopy techniques, such as SORS, TRS, and CARS.
