Available in PDF, EPUB and Kindle. Book excerpt: This book presents a new emerging concept of "Integrative Structural Biology". It covers current trends of the molecular and cellular structural biology, providing new methods to observe, validate, and keep the structural models of the large cellular machines with recent scientific results. Structures of very large macromolecular machines in cells are being determined by combining observations from complementary experimental methods. Thus, this volume presents the each methods such as X-ray crystallography, NMR spectroscopy, 3DEM, small-angle scattering (SAS), FRET, crosslinking, and enables the readers to understand the hybrid methods. This book discusses how those integrative models should be represented, validated and archived. A unique highlight of this book is discussion of the data validation and archive, which are big problems in this filed along with the progress of this field. The researchers in biology will be interested in this book as a guide book for learning the current structure biology, but also those in structure biology may use this book as a comprehensive reference to cover broad topics.

Available in PDF, EPUB and Kindle. Book excerpt: This volume is a collection of the contributions presented at the 42nd Erice Crystallographic Course whose main objective was to train the younger generation on advanced methods and techniques for examining structural and dynamic aspects of biological macromolecules. The papers review the techniques used to study protein assemblies and their dynamics, including X-ray diffraction and scattering, electron cryo-electron microscopy, electro nanospray mass spectrometry, NMR, protein docking and molecular dynamics. A key theme throughout the book is the dependence of modern structural science on multiple experimental and computational techniques, and it is the development of these techniques and their integration that will take us forward in the future.

Book excerpt: This volume explores strategies and detailed protocols for the preparation of macromolecular complexes and their characterization in view of structural analysis. The chapters in this book are separated into three parts: Part One focuses on sample preparation, and covers strategies for recombinant expression of multiprotein complexes in prokaryotic and eukaryotic hosts, for genome engineering using the CRISPR/Cas9 system and for production of specific binders such as reformatted antibodies and artificial binding proteins. Part Two looks at the biophysical methods that can provide useful indicators for sample optimization, and often complement structural information obtained with core technologies for structure determination—x-ray crystallography and cryo-electron microscopy—by quantitative solution data. Part Three discusses the characterization of multiprotein complexes in a cellular environment using the latest technologies and in vivo approaches. Written in the highly successful Methods in Molecular Biology series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Cutting-edge and authoritative, Multiprotein Complexes: Methods and Protocols is a valuable resource for structural and molecular biologists who need to prepare multi-components for their applications, and for other scientists working on macromolecular assemblies from other angles that need to know the latest approaches that the field has to offer.

Book Synopsis Macromolecules by : Finn Wold

Book excerpt: In this book we discuss the status of the structure- function analysis of biological macromolecules and macromolecular complexes. The ultimate goal of the analysis must be to explain all the functional properties of the molecules in question in terms of their completely defined three-dimensional structure, and the analysis thus contains three separate components: the determination of structure, the determination and quantitation of function, and final correlation of this information into the structure-function model. The first component, the structural analysis, is reviewed only briefly, and this book therefore leans heavily on Barker's and Van Holde's books in this series for proper background and documentation for this component. The second component, the analysis of functional properties, is given broader consideration (Chapters I, 2, 5, and 9), but the main emphasis has been the step-by-step development of the structure-function models. It is hoped that this approach will clearly illustrate the typical progression of scientific model building from the first clear definition of the problem and the statement of the hypothesis through ever-increasing refinements of experimental tests toward the final answer. It is also hoped that the statements of philosophy, principles, and scientific method that are the bases for this approach are of broad enough validity to survive even after its models have become obsolete. With this approach, it is essential to inform the reader in unequivocal terms that this book is not a summary of final conclusions and complete stories which can be submitted to memory. Each system discussed should be considered very critically, and the models should be evaluated in terms of the available evidence. The only "facts" are the experimental data; the interpretation of this data into models is only convincing to the extent that it makes logical sense to the individual examining it. Since both space and common sense prohibits a continuous reiteration of this statement throughout the book, be prepared to encounter some models and hypotheses which are based on sound experimental evidence as well as some which have no experimental basis at all. In neither case are they "facts," but in either case they represent ideas which can be subjected to further experimental tests. If the book helps to sharpen this critical evaluation of both ideas and the experimental test of the hypotheses, one of its major purposes has been fulfilled.

Book excerpt: The volume presents a survey of the research by Kurt Wüthrich and his associates during the period 1965 to 1994. A selection of reprints of original papers on the use of NMR spectroscopy in structural biology is supplemented with an introduction, which outlines the foundations and the historical development of the use of NMR spectroscopy for the determination of three-dimensional structures of biological macromolecules in solution. The original papers are presented in groups highlighting protein structure determination by NMR, studies of dynamic properties and hydration of biological macromolecules, and practical applications of the NMR methodology in fields such as enzymology, transcriptional regulation, immunosuppression and protein folding.
