SERS spectra of the cured polymer were more simular to the SERS spectra of the polyamic acid than to the normal Raman spectrum of the polymide, indicating that curing of the polymer was inhibited by interaction of the polyamic acid with the substrate. Thus, the strongest band in the normal RAMAN spectrum of the polymide was near 1406 cm-1 and was assigned to the CNC axial stretching mode.

The mechanisms are associated with the large electric fields that can be induced at the surfaces of metal particles having small radii of curvature and with formation of charge transfer complexes between the adsorbed molecules and the substrate. Enhancement due to the charge transfer mechanism is obtained only for molecules immediately adjacent to the surface but enhancement due to the electromagnetic mechanism may extend several monolayers away from the surface. Since normal Raman scattering by polymers is weak and scattering by molecules adjacent to the substrate is strongly enhanced, SERS can be used for nondestructive characterization of interfaces between polymer films and metals. Book excerpt: Surface-enhanced Raman scattering was observed from thin films formed by 1,4- and 1,3-dinitrobenzene spin-coated onto silver island films from dilute solutions. During laser irradiation of 1,4-DNB films on silver substrates at relatively high laser powers, new bands appeared near 1152, 1350, 1412, and 1460/cm and bands near 1118, 1358, and 1372/cm decreased in intensity. A band near 1598/cm shifted to near 1605/ cm. The bands near 1152 and 1412/ cm were assigned to stretching modes of Phi-N and -N=N- bonds of azo compounds, respectively, indicating that polymerization of 1,4-DNB had occurred. The extent of polymerization was greatest when the surface coverage of adsorbed 1,4-DNB was greatest and when the SERS experiments were carried out in a nitrogen atmosphere, indicating that reducing conditions were required for the polymerization. No polymerization was observed for 1,3-DNB under similar circumstances. The difference was attributed to orientation of the adsorbed species. Thus, it was concluded that 1,4-DNB was adsorbed with a horizontal configuration in which the aromatic rings were parallel to the substrate surface and both nitro groups were in contact with the surface while 1,3-DNB was adsorbed with a vertical configuration in which the rings were perpendicular to the surface and only one nitro group was in contact with the surface.

Book excerpt: Although surface-enhanced Raman spectroscopy (SERS) has been a valuable tool for chemical detection, the understanding of the contributions of the chemical enhancement effect in nucleic acid-silver composites remains limited. Nucleic acid-silver composites have unique optical properties such as fluorescence and enhanced Raman generation, and thus need to be further analyzed and characterized. In this dissertation, the static polarizability chemical effect is used to determine the preferential binding sites of nucleic acids to silver nanoparticles. The binding configuration dictates the optical properties, so it is imperative to determine which atoms are binding to the silver structures. Time-dependent density functional theory simulations are used to calculate the Raman frequencies of several systems and are then quantitatively compared to experimental measurements. The approximate composition of the binding sites for each nucleic acid to silver films is determined using the simulated Raman correlation spectroscopy process. In the second part of the dissertation, the charge-transfer effect in nucleic acid-silver composites is harnessed in order to selectively enhance or reduce the SERS signals via wavelength selection. Depending on the molecular properties of the nucleic acids, the optimal excitation wavelength shifts from visible to near-infrared. The properties can be taken advantage of to better control the systems under study. The charge-transfer effect is further understood by utilizing a thin film of aluminum oxide to prevent electron transfer from occurring, and thus enabling the calculation of the order of magnitude of the charge-transfer effect. The characterization of nucleic acid-silver composites is imperative to fully understanding the unique and exciting properties that these systems provide. While SERS has been limited in terms of DNA detection or sequencing, the restrictions have been caused by insufficient information. This dissertation offers additional information on the systems and explains the discrepancies seen in previous measurements of nucleic acid-silver composites.

Book excerpt: Surface-enhanced Raman scattering (SERS) has been used successfully as an ultrasensitive technique for the direct analysis of dye-labelled oligonucleotides using aggregated silver nanoparticles. Key to this technique are the SERS probe and SERS substrate. Initial studies were made on the detection of phosphorothioate modified TAMRA-labelled oligonucleotides whereby a non-bridging oxygen atom in the phosphodiester backbone is replaced by sulfur. TAMRA-labelled phosphorothioate oligonucleotides with the same base pair sequence but different number of modified phosphate unites were compared to unmodified TAMRA-labelled oligonucleotides using silver nanoparticles (AgNP) as SERS substrates using different aggregation agents. Furthermore the investigation was extended to study the effect of modification in the detection of double stranded DNA (dsDNA). The focus was then turned to the SERS substrate since spherical and monodispersed silver nanoparticles are ideal for fundamental research as the contribution from size and shape can be accounted for in the experimental design. Different methods to produce size tuneable AgNP's were investigated to study in detail the effect of modified and unmodified dye-labelled oligonucleotides, thiophenol (TP), rhodamine 6G (R6G) and malachite green oxalate (MGO) on the SERS response. The results obtained reflect the importance of each component of SERS. The phosphorothioate modifications, although their affinity towards silver surfaces can improve the DNA limit of detection (LOD), it also depends on the position and number of modifications and on the aggregating agent used which in itself also effects the LOD of DNA. The same applies for the SERS substrate where an increase in particle size results in an increased enhancement of the analyte's Raman signal but further size increments leads to a decrease in SERS intensity due to other factors such as scattering. This demonstrates the importance of choosing the right experimental setups for DNA SERS analysis, as to obtain the right balance where all factors work synergistically with each other.

Book excerpt: Surface-enhanced Raman scattering (SERS) from organic molecules is used to study how geometry affects plasmonic enhancement in noble metal nanoparticles using super-resolution analysis techniques coupled with scanning electron microscope (SEM) imaging. We demonstrate that super-resolution analysis can track emission from Raman-active molecules on nanoaggregate surfaces with resolution typically better than 5 nm, a significant improvement over the diffraction limit. This optical analysis technique is used to identify active junction regions, known as hot spots, in correlated SEM structural images of aggregated particles based on shape, size, and angular position of SERS emission. This correlated analysis is further extended to study the mechanism of silver nanoparticle luminescence and its positional relationship to SERS emission. These studies support the hypothesis that silver luminescence is enhanced via a different mechanism than SERS. Discrete dipole approximation calculations agree with experimental results, indicating that SERS emission is a product of local plasmonic enhancement in specific junction regions between nanoparticles whereas silver luminescence is dependent upon nanoaggregate geometry and the collective plasmon modes within a structure. These theoretical calculations of luminescence centroid location can assist with SERS-active junction region assignment in higher order aggregates (e.g. trimers, tetramers, etc.) Finally, correlated optical and structural studies are used to develop understanding of site-specific electrochemical potentials on silver and gold nanoparticle aggregates using a redox active Raman reporter molecule. We demonstrate that there is a weak correlation between emission centroid location and bulk sample potential. Silver colloid studies demonstrate variability in modulation behavior and are highly unpredictable, whereas gold colloids with the redox molecule covalently tethered to their surface via a gold-thiol bond show more reproducible modulation. Furthermore, these tethered studies demonstrate geometrical agreement between site-specific emission at negative potentials and junction regions in nanoaggregate SEM images indicating that plasmonic enhancement may affect local nanoparticle surface potentials.
