Book excerpt: During the last 30 years, many chemical compounds that are active against tumors have been discovered or developed. At the same time, new methods of testing drugs for cancer therapy have evolved. nefore 1964, drug testing on animal tumors was directed to observation of the incfease in life span of the host after a single dose. A new approach, in which the effects of multiple doses on the proliferation kinetics of the tumor in vivo as well as of cell lines in vitro are investigated, has been outlined by Skipper and his co-workers in a series of papers beginning in 1964 (Skipper, Schabel and Wilcox, 1964 and 1965). They also investigated the influence of the time schedule in the treatment of experimental tumors. Since the publication of those studies, cell population kinetics cannot be left out of any discussion of the rational basis of chemotherapy. When clinical oncologists began to apply cell kinetic concepts in practice about 15 years ago, the theoretical basis was still very poor, in spite of Skipper's progress, and the lack of re levant cytokinetic and pharmacologic data was apparent. Subsequently, much theoretical work has been done and many cell kinetic models have been elaborated (for a review see Eisen, 1977).

Available in PDF, EPUB and Kindle. Book excerpt: Kinetics of Experimental Tumour Processes focuses on the principles of the kinetics of experimental malignant growth. This book also looks into the molecular mechanisms of chemotherapy and certain problems of carcinogenesis and biochemistry. Organized into eight chapters, this monograph first describes the kinetic curve for tumor growth and the experimental data on tumor growth from the USSR and elsewhere. Subsequent chapters deal with kinetic analysis of the results obtained by various treatments of tumors; pharmacokinetic methods of major regularities connected with general problems of cancer chemotherapy; molecular mechanisms of chemotherapy; and the role of free radicals in the mechanisms of carcinogenesis and tumor growth. The last two chapters detail the kinetic analysis of biochemical shifts occurring in tumor tissues and the disturbance of structure and biosynthesis of informational macromolecules occurring in tumor growth. This book will help in the mutual efforts among researchers for the solution of the most important current problems in oncology.

Available in PDF, EPUB and Kindle. Book excerpt: Introduction to Mathematical Oncology presents biologically well-motivated and mathematically tractable models that facilitate both a deep understanding of cancer biology and better cancer treatment designs. It covers the medical and biological background of the diseases, modeling issues, and existing methods and their limitations. The authors introduce mathematical and programming tools, along with analytical and numerical studies of the models. They also develop new mathematical tools and look to future improvements on dynamical models. After introducing the general theory of medicine and exploring how mathematics can be essential in its understanding, the text describes well-known, practical, and insightful mathematical models of avascular tumor growth and mathematically tractable treatment models based on ordinary differential equations. It continues the topic of avascular tumor growth in the context of partial differential equation models by incorporating the spatial structure and physiological structure, such as cell size. The book then focuses on the recent active multi-scale modeling efforts on prostate cancer growth and treatment dynamics. It also examines more mechanistically formulated models, including cell quota-based population growth models, with applications to real tumors and validation using clinical data. The remainder of the text presents abundant additional historical, biological, and medical background materials for advanced and specific treatment modeling efforts. Extensively classroom-tested in undergraduate and graduate courses, this self-contained book allows instructors to emphasize specific topics relevant to clinical cancer biology and treatment. It can be used in a variety of ways, including a single-semester undergraduate course, a more ambitious graduate course, or a full-year sequence on mathematical oncology.

This book was released on 2013-01-25 with total page 398 pages. Available in PDF, EPUB and Kindle. Book excerpt: The future of oncology seems to lie in Molecular Medicine (MM). MM is a new science based on three pillars. Two of them are evident in its very name and are well known: medical science and molecular biology. However, there is a general unawareness that MM is firmly based on a third, and equally important, pillar: Systems Biomedicine. Currently, this term denotes multilevel, hierarchical models integrating key factors at the molecular, cellular, tissue, through phenotype levels, analyzed to reveal the global behavior of the biological process under consideration. It becomes increasingly evident that the tools to construct such complex models include, not only bioinformatics and modern applied statistics, as is unanimously agreed, but also other interdisciplinary fields of science, notably, Mathematical Oncology, Systems Biology and Theoretical Biophysics.

Mathematical models enable quantitative representations of the changes affecting cell state and cell number. This book provides a review of the scope of mathematical modelling in cancer research, bringing together for the first time a group of related mathematical topics including multistage carcinogenesis, tumour growth kinetics, growth control, radiotherapy, chemotherapy and biological targeting in cancer treatment. Physicists and mathematicians interested in medical research, biomathematicians, biostatisticians, radiation and medical oncologists and experimental and theoretical biologists will welcome this critical review of mathematical modelling in cancer research. This book will also be of interest to clinicians, basic cancer scientists and physicists working in radiotherapy departments, and to postgraduate students on courses in oncology and subjects.

Available in PDF, EPUB and Kindle. Book excerpt: The use of medical information for clinical decisionmaking and the use of formal computer-based methodologies which can critically improve patient management in leukemia and in solid tumors are explored. The aim of the scientific treatment of disease is to intervene as directly as possible in the biology of the disease process. But as medical therapy penetrates deeper into this fundamental biology, the more numerous become the circumstances where a therapeutic plan must be instituted or a therapy sustained on the basis of indirect evidence. This is particularly true of treatments involving the use of powerful pharmacological agents. Some successful mathematical models of patient response to treatment have been developed, however, which demonstrably improve the clinical management of patients. Systems of models simulating patient response to cancer chemotherapy are discussed.
