Book excerpt: Discusses how to fight Ebola, SARS Corona, and other known or emerging human viruses by building on the successes in antiviral therapy of the past decades Written by leading medicinal chemists from academia and industry, this book discusses the entire field of antiviral drug discovery and development from a medicinal chemistry perspective, focusing on antiviral drugs, targets, and viral disease mechanisms. It provides an outlook on emerging pathogens such as Ebola, Zika, West Nile, Lassa, and includes a chapter on SARS Coronoavirus-2 causing the present pandemic. New Drug Development for Known and Emerging Viruses describes the discovery and development process for antiviral agents for different classes of viruses and targets based on the experiences from the nine human viruses for which approved drugs are on the market (HIV, HCV, Influenza, RSV, HBV, HPV, HCMV, HSV, and VZV). It covers the properties and potential of 20 classes of currently approved antivirals, including combination drugs, and looks at novel antiviral strategies against emerging viruses. Covers the entire field of antiviral drug discovery and development Addresses the need for antiviral drugs to combat major health threats such as Ebola, Zika, West Nile, and SARS Coronavirus-2 Summarizes the successes of the past 15 years in developing ground-breaking medicines against 9 major human viruses, both from the medicinal chemistry and the pharmacological angle Discusses practical and strategic challenges in the drug discovery and development process, including screening technologies, latency, and toxicity issues New Developments in Antiviral Drugs is an important book for medicinal chemists, pharmaceutical chemists, virologists, and epidemiologists, and will be of great interest to those in the ;pharmaceutical industry and public health agencies.

Chapter 6, Arenaviruses: The biochemical and physical properties have now been clar ified, and they show a remarkable uniformity in the various viruses constituting the group. The possibility that prenatal infection with LCM may result in hydrocephalus and chorioretinitis has been raised. Serologic surveys have suggested the existence of Lassa virus infection in Guinea, Central African Empire, Mali, Senegal, Cameroon, and Benin, in addition to earlier identification in Nigeria, Liberia, and Sierra Leone. Chapter 7, Coronaviruses: New studies have confirmed the important role of these viruses in common respiratory illnesses of children and adults. The viruses are now known to contain a single positive strand of RNA. About 50% of corona virus infections result in clinical illness. About 5% of common colds are caused by strain DC 43 in winter. Chapter 8, Cytomegalovirus: Sections on pathogenesis of CMV in relation to organ transplantation and mononucleosis, as well as sections on the risk and features of con genital infection and disease, have been expanded. There are encouraging preliminary results with a live CMV vaccine, but the questions of viral persistence and oncogenicity require further evaluation.

Book excerpt: Arthropod borne diseases cause enormous morbidity and mortality in most countries, mostly in those situated in tropical areas, but also in temperate regions. This book provides organized information on all arthropod related diseases, to prevent suffering and deaths, for medical students and professionals. Since arthropod borne diseases are present in many regions of the world and can even surprise professionals and lays in non-endemic regions, like malaria in UK and Canada, the author and its many expert collaborators are sure that it will be essential in all hospitals, clinics and medical libraries around the world. As arthropod borne diseases of domesticated animals are very numerous and in some cases related to human diseases, they are also included in the book.

Book excerpt: This first published edition of the Catalogue of Arthropod-borne Viruses provides concise and standardized information on 204 viruses provisionally classifies as arboviruses. The published version is a facsimile of the February 1967 version of the ongoing working Catalogue. The working Catalogue is constantly updated and has a limited distribution. Eligible for registration in the Catalogue are vertebrate viruses, published and unpublished, which are biologically transmitted by arthropods. Registrations are voluntarily submitted by working arbovirologists. Information on each virus includes: source and manner of isolation; physical, chemical, and antigenic characteristics; natural and experimental host range; pathogenesis; geographic distribution; and the frequency and symptomatology of human infection.

Book excerpt: Japanese encephalitis and West Nile viruses are members of the Japanese encephalitis serological group of the genus Flavivirus and therefore closely related genetically and antigenically. They share a number of properties, including the use of birds as their major wildlife maintenance host and Culicine mosquitoes for transmission, and they are both associated with severe human disease, as well as fatal infections in horses. The emergence of these two viruses, and their well-established propensity to colonise new areas, make it timely to re-examine their ecology, biology, molecular structure, replication and epidemiology, and these therefore provide the focus of this volume.

Book excerpt: The publication of this volume of The Viruses entitled The Togaviridae and Flaviviridae comes at an appropriate time. The structure and rep lication strategies of these viruses are now known to be sufficiently di verse to warrant the removal of flaviviruses from the Togaviridae family and establish them as an independent family. Flaviviridae have a special place in the history of virology. The prototype virus-yellow fever virus was the first virus to be identified as the cause of a human disease. Some of the history of this discovery is described in Chapter 1 of this volume; in Chapter 10 the complete sequence of the RNA genome of the virus is presented. This sequence not only defines the primary structure of the viral proteins, it also clarifies the mechanism of translation of the fla vivirus genome. Knowledge of the sequence of the structural proteins of these viruses represents an important step in the potential goal of using purified flavivirus glycoproteins as vaccines. Many of the chapters in this volume focus on the structure and replication of the Togaviridae. These viruses have provided valuable models for studies in cell biology, partic ularly with regard to the cotranslational and posttranslational steps re quired for the synthesis and localization of membrane glycoproteins. Fur thermore, Togaviridae have been pivotal in our growing understanding of how enveloped viruses enter and exit from cells. The broad outlines of the structure and gene expression of Togavir idae and Flaviviridae are known, but important questions remain.

Available in PDF, EPUB and Kindle. Book excerpt: Arctic and Tropical Arboviruses contains the proceedings of the Second International Symposium on Arctic Arboviruses held at Mont Gabriel, Canada on May 26-28, 1977. This book contains a total of 20 chapters; a few of these chapters describe the diseases with arbovirus as a possible etiological agent, such as in the case of nephropatia epidemica, rapid diagnostic techniques for the detection of arboviruses, and in vitro culture methods for arboviruses using arthropod cells. Several other chapters are devoted to the investigations on arboviruses in the northern regions and on their vectors, mosquitoes, and ticks, as well as to the detection in the north of arboviruses originally isolated inthe south. Such bipolar distribution of arboviruses could be the result of the transport of arbovirus-infected ticks by migratory birds. This volume will provide a useful tool for all concerned with viral diseases, including virologists, epidemiologists, and ecologists.
