Available in PDF, EPUB and Kindle. Book excerpt: Calcium Binding Proteins explains the unique and highly diverse functions of calcium in biology, which are realized by calcium binding proteins. The structures and physical characteristics of these calcium binding proteins are described, as well as their functions and general patterns of their evolution. Techniques that underlie the description of proteins are discussed, including NMR, circular dichroism, optical rotatory dispersion spectroscopy, calorimetry, and crystallography. The book discusses the patterns of bochmical phenomena such as calcium homeostasis, mineralization, and cell signaling that involve specific proteins. It summarizes ongoing research and presents general hypotheses that help to focus future research, and also provides a conceptual framework and a description of the underlying techniques that permits someone entering the field to become conversant.

Book excerpt: The idea of Professors Bolis and Gilles to gather together for a 3 days' meeting in the splendid environment of Crans-Montana in Switzerland a limited number of people around the subject of calcium and calcium bind ing proteins seemed at first particularly attractive, and when they asked me to take charge of the scientific organization of the symposium, I accepted with enthusiasm. It rapidly became clear that the major problem would be the selection of the topics, since it was impossible to cover completely and in depth such a broad and dynamic area of research. In our view, one imperative was to associate as intimately as possible the structural and the functional aspects of the areas covered. Apart from one whole day focused on the fascinating roles played by calmodulin in cellular activities, the other sessions were devoted to calmodulin-related calcium binding proteins in muscle and non muscle tissues and to some selected biological systems such as mitochondria, secretory cells or sarcoplasmic reticulum in which calcium also plays a crucial role. The presentations were made by leading investigators in their field. Some of them do not, however, appear in the present volume, for which there are two reasons: first, some of the contributions were somewhat outside the scope of the book; second, three speakers, for valid reasons, simply found no opportunity to write a manuscript in the allotted time.

The contributions are grouped according to their general subject matter, based on the classification made by the authors, e.g., Extracellular Metabolism: Calcium Homeostasis; Low-Affinity Calcium-Binding Proteins. There can be little doubt that calcium-binding proteins is a field of scientific endeavor which will continue to produce results of great interest to modern biology. This book is divided into eight sections, the first of which, tackle extracellular calcium metabolism or calcium homeostasis. The next sections focus on topics such as low-affinity calcium-binding proteins; calcium and membranes, channels and transport (pumps); the role of calcium in complex metabolic processes; and gene structure of calcium-binding proteins and their expression. Other chapters cover physical studies on calcium-binding proteins, including X-ray, crystallography, and NMR; structure-function relationships of calcium-binding proteins and their targets; and calcium-binding proteins in health and disease. This book will be of interest to practitioners in the fields of biology and medicine.

Available in PDF, EPUB and Kindle. Book excerpt: In biochemistry, a metalloprotein is a generic term for a protein that contains a metal cofactor. The metal may be an isolated ion or may be coordinated with a nonprotein organic compound, such as the porphyrin found in hemoproteins. In some cases, the metal is co-coordinated with a side chain of the protein and an inorganic nonmetallic ion. This kind of protein-metal-nonmetal structure is seen in iron-sulfur clusters Metalloproteins deals with all aspects related to the intracellular and extracellular metal-binding proteins, including their structures, properties and functions. The biological roles of metal cations and metal-binding proteins are endless. They are involved in all crucial cellular activities. Many pathological conditions are related to the problematic metal metabolism. Research in metalloprotein-related topics is therefore rapidly growing, and different aspects of metal-binding proteins progressively enter curricula at Universities and even at the High School level on occasion. However, no key resource providing basic, but comprehensible knowledge on this rapidly expanding field exists. The Encyclopedia of Metalloproteins aims to bridge this gap, and will attempt to cover various aspects of metalloprotein/metalloproteomics and will deal with the different issues related to the intracellular and extracellular metal-binding proteins, including their structures, properties and functions. The goal is to cover exhaustively all catalytically and biologically crucial metal ions and to find at least one interacting protein for other metal ions. The Encyclopedia of Metalloproteins will provide a key resource for advanced undergraduate and graduate students, researchers, instructors, and professors interested in protein science, biochemistry, cell biology, and genetics.

Available in PDF, EPUB and Kindle. Book excerpt: A wide variety of hormones, neurotransmitters and growth factors exert their cellular effects by reacting first with membrane receptors resulting in an increase of intracellular calcium and the cellular response. The calcium signal in the cell is mediated by the high-affinity calcium binding proteins (characterized by the EF-hand structural element), and by the calcium and phospholipid dependent proteins. Many of these have been discovered most recently. Their purification, distribution, protein and gene structures as well as their physiological roles are discussed. The book is of interest to biochemists and molecular biologists as well as to clinicians and the pharmaceutical industry who can apply the results in this field.

Available in PDF, EPUB and Kindle. Book excerpt: Calcium plays an important role in a wide variety of biological processes. This divalent metal ion can bind to a large number of proteins; by doing so it modifies their biological activity or their stability. Because of its distinct che- cal properties calcium is uniquely suited to act as an on–off switch or as a light dimmer of biological activities. The two books entitled Calcium-Binding Protein Protocols (Volumes I and II) focus on modern experimental analyses and methodologies for the study of calcium-binding proteins. Both extracel- lar and intracellular calcium-binding proteins are discussed in detail. H- ever, proteins involved in calcium handling (e. g. , calcium pumps and calcium channels), fall outside of the scope of these two volumes. Also, calcium-bi- ing proteins involved in bone deposition will not be discussed, as this specific topic has been addressed previously. The focus of these two books is on studies of the calcium-binding proteins and their behavior in vitro and in vivo. The primary emphasis is on protein chemistry and biophysical methods. Many of the methods described will also be applicable to proteins that do not bind calcium. Calcium-Binding Protein Protocols is divided into three main sections. The section entitled Introduction and Reviews provides information on the role of calcium in intracellular secondary messenger activation mechanisms. Mo- over, unique aspects of calcium chemistry and the utilization of calcium in dairy proteins, as well as calcium-binding proteins involved in blood clotting, are addressed.

Book excerpt: Proteins: Structure and Function is a comprehensive introduction to the study of proteins and their importance to modern biochemistry. Each chapter addresses the structure and function of proteins with a definitive theme designed to enhance student understanding. Opening with a brief historical overview of the subject the book moves on to discuss the ‘building blocks’ of proteins and their respective chemical and physical properties. Later chapters explore experimental and computational methods of comparing proteins, methods of protein purification and protein folding and stability. The latest developments in the field are included and key concepts introduced in a user-friendly way to ensure that students are able to grasp the essentials before moving on to more advanced study and analysis of proteins. An invaluable resource for students of Biochemistry, Molecular Biology, Medicine and Chemistry providing a modern approach to the subject of Proteins.
