Bovine brucellosis, caused by Brucella abortus, is of greatest concern due to the disease's significant economical and public health impact. Canada fully eradicated bovine brucellosis from domesticated cattle herds in 1985, however, continued surveillance through screening for B. abortus exposure is paramount to the maintenance of bovine brucellosis eradication nationwide. The Canadian Food Inspection Agency (CFIA) is responsible for the surveillance of bovine brucellosis outbreaks in Canada and the maintenance of eradication. Current B. abortus serodiagnostics and serological screening is mostly based on the detection of antibodies against Brucella lipopolysaccharide (LPS), a highly immunogenic component of the outer cellular membrane. Such tests face difficulties with false positive results due to cross reactivity with other Gram-negative bacteria that produce LPS. The purpose of the research presented here was to address this issue through identifying new B. abortus protein antigens for the improvement of serological test specificity. In this study, 101 candidates were identified through predictive bioinformatic analyses and selected for immunogenic evaluation. While none of the expressed candidates displayed positive serological activity with in-house brucellosis positive bovine serum panels, the workflow presented here can be used for continued research and the assessment of more proteins from B. abortus and other bacterial pathogens.

Book excerpt: Brucella abortus, the causative agent of bovine brucellosis, has been nearly eliminated from cattle in the United States through a federal-state eradication program that began in 1930s. However, the disease remains endemic in free-ranging elk (Cervus elaphus nelsoni) and bison (Bison bison) in the Greater Yellowstone Area (GYA) Wyoming, Montana, and Idaho. Annual brucellosis seroprevalence in adult female elk averages 22% on Wyoming's winter feedgrounds within the GYA. Feedgrounds concentrate animals during the peak period of transmission of brucellosis (late winter and early spring) posing a transmission risk to cattle or sympatric wildlife that winter near these feedgrounds. In an attempt to reduce seroprevalence in elk, the Wyoming Game and Fish Department implemented a five-year pilot Test-and-Slaughter program focused on the Pinedale Elk Herd Unit with the primary objective to achieve a statistically significant reduction in brucellosis seroprevalence on the target feedground. A significant decrease in seroprevalence was documented on Muddy Creek Feedground, the target feedground, from 35.9% in 2006 to 7.0% in 2009. This program was effective in reducing brucellosis seroprevalence in feedground elk of this herd unit and holds promise as a large-scale disease control tool, however it was costly, labor intensive, and resulted in the slaughter of several false positives. Presumptive and rapid diagnosis of brucellosis in elk was based on five serologic tests. A sixth test (complement fixation assay) is performed as an additional confirmatory test at a later date. The research presented here calculates the sensitivity and specificity of the six serologic tests and determines the battery of tests that most accurately identify truly infective elk as determined by bacteriologic culture. Sensitivity increased to 100% and specificity increased to 21.1% if the battery of tests was changed from the current scheme to modified scheme (card test = positive, SPT e"I200, rivanol e"I50, FPA e"110). The competitive enzyme linked immunosorbent assay and compliment fixation test should be discontinued as they decrease specificity in all testing schemes investigated.
