A study of such biotic desulfurizations can be expected to give rise to new and useful chemistry and enzymology. The potential value of understanding and harnessing these processes is seen in relation to the need for methods for the removal of organically bound sulfur from coal and the degradation of organic sulfur-containing pollutants. This research effort has been directed towards an examination of desulfurization ability in well characterized microorganisms, the isolation of bacteria with desulfurization ability from natural sources, the characterization and mechanistic evaluation of the observed biocatalytic processes, the development of biomimetic synthetic organic chemistry based on biotic desulfurization mechanisms and the design and preparation of improved coal model compounds for use in microbial selection processes. A systematic approach to studying biodesulfurizations was undertaken in which organosulfur compounds have been broken down into classes based on the oxidation state of the sulfur atom and the structure of the rest of the organic material. Microbes have been evaluated in terms of ability to degrade organosulfur compounds with sulfur in its sulfonic acid oxidation state. These compounds are likely intermediates in coal desulfurization and are present in the environment as persistent pollutants in the form of detergents. It is known that oxygen bonded to sulfur lowers the carbon-sulfur bond energy, providing a thermodynamic basis for starting with this class of compounds.

Book excerpt: Growth assays reveal that Rhodococcus rhodochrous IGTS8 can utilize a wide range of organosulfur compounds as the sole source of sulfur. Compounds that are utilized include thiophenes, sulfides, disulfides, mercaptans, sulfoxides, and sulfones. None of the organosulfur compounds tested can serve as a carbon source. A convenient spectrophotometric assay (Gibbs assay) based on the chromogenic reaction of 2,6-dichloroquinone-4-chloroimide with aromatic hydroxyl groups was developed and used in conjunction with GC/MS analysis to examine the kinetics of carbon-sulfur bond cleavage by axenic and mixed cell cultures of Rhodococcus rhodochrous IGTS8. The desulfurization trait is expressed at uniform levels during the mid-exponential phase, reaches a maximum during idiophase, and then declines in stationary-phase cells. Desulfurization rates for dibenzothiophene (DBT) range from 8 to 15 [mu]M of DBT/1012 cells/hour. Mixtures of genetically marked Rhodococcus rhodochrous IGTS8 and an organisms incapable of cleaning carbon-sulfur bonds in relevant test compounds, Enterobacter cloacae, were prepared in ratios that varied over six orders of magnitude. Growth studies revealed that Enterobacter cloacae was able to gain access to sulfur liberated from organosulfur compounds by IGTS8; however, cell-to-cell contact was required. These data also indicate that the desulfurization activity of IGTS8 cells in mixed cultures may be as much as 200-fold higher than in axenic cultures.

Book excerpt: Significant opportunity exists for the application of supercritical fluid (SCF) technology to coal processing, both for pretreatment of high sulfur coals, as well as liquefaction and treatment of coal liquids. Supercritical fluids are attractive solvents for a variety of coal processing applications because of their unusual solvating and mass transfer properties. Solubility studies have been carried out for a number of model coal and coal-liquid compounds, primarily in pure supercritical fluids. We have extended this database of model coal compound equilibria using modern techniques that have the advantage of being much more rapid than traditional techniques. Cosolvent effects on solubility have also been investigated over a variety of solvent properties. In addition, specific molecular interactions have been investigated through spectroscopic techniques. The resulting data has been used to develop a physical-chemical equation of state (EOS) model of SCF solutions with meaningful parameters. This equation of state model has been used to predict solubility behavior, which will permit the design and tailoring of SCF cosolvent systems for specific coal processing applications.

After the enzymes have been successfully purified, the genes encoding these enzymes will be cloned and the suitability of these microorganisms for recombinant manipulation will also be studied. Initially, the enzymes involved in the removal of organic sulfur from coal and from model compounds in two particular strains, IGTS8 and K3B, will be characterized and analyzed. The scope of the proposed study covers the following: (1) Characterization, purification, and partial sequence analysis of the enzymes involved in the removal of organic sulfur from coal. (2) Cloning of the genes encoding the organic sulfur-degrading enzymes from these two selected species in E. coli. (3) Sequence analysis of the cloned genes. (4) Attempting to clone and overexpress the organic sulfur degrading genes in their parent hosts as well as in other hosts.

Book excerpt: Laboratory-scale research conducted at Southern Illinois University at Carbondale (SIUC) has shown that coal pyrolysis in the presence of CH4 and small quantities of O2 (the METHOXYCOAL process) can produce high yields of liquids and valuable chemicals compared to conventional pyrolysis. The addition of MgO, coal ash, and clays have been shown to further enhance coal conversion. The goal of this two-year project is to build upon that laboratory research by conducting continuous bench-scale tests at IGT. Tests are being conducted with IBC-101 coal under CH4/O2 blends with and without added coal ash, MgO, and/or clays, at temperatures and pressures up to 1000°F and 200 psig. These tests will provide data to select preferred operating conditions for chemicals production from high-sulfur Illinois coals.

Book excerpt: The objective of this program is to investigate the desulfurization of mild gasification char using H2:CH4, mixtures at the laboratory scale. Mild gasification produces solid, liquid, and gaseous co-products at 1000° to 1500°F and near-ambient pressure. Char comprises about 60 to 70% of the dry coal yield. Form coke for steelmaking and foundries presents potential high-value markets for chars from eastern bituminous coals, Metallurgical cokes generally contain less than 1 wt% sulfur, and mild gasification char from high-sulfur Illinois coals must be upgraded to meet these criteria. One method to accomplish this is desulfurization with reducing gases derived from the.co-product gases. In the first year of the two-year program, granular chars were treated with H2:CH4, blends at temperatures of 1100° to 1600°F and pressures of 50 to 200 psig. During the year, 23 tests were performed with 10 chars. Fluidized-bed tests were conducted for 120 to 240 minutes with superficial gas velocities from 0.067 to 0.150 ft/s. The desulfurization medium was H2 gas containing 9 to 24 vol% CH4. The data from these tests show sulfur conversions ranging from 6.0 to 92.5 wt%, with carbon conversions from zero to 35.3 wt%. The maximum sulfur conversion was 92.5 wt% at 1400°F and 200 psig in 76% H, for 120 minutes residence time, using IFFR char produced from IBC-106 coal at 1200°F in helium. The sulfur content of the char in that test was reduced from 3.78 to 0.39 wt%.
