Available in PDF, EPUB and Kindle. Book excerpt: Population genomics is a rapidly emerging field that has the potential to transform our understanding of how evolutionary forces shape genomic diversity among microbes. There have already been considerable advances in understanding gene flow and spread of adaptive traits, and in linking epidemiology with evolutionary biology. The current challenge is to find unifying evolutionary principles for organisms that display a wide range of reproductive biology – from highly clonal to promiscuous – and for which the vast majority have eluded cultivation. This requires interdisciplinary approaches that incorporate novel computational tools, testing of existing and novel population genetic models, and creative new ways of linking genetic diversity to ecological factors. This pioneering book will discuss the advances made and promises of population genomics in microorganisms, outlining some of the key theoretical and practical challenges for microbial population genomics, including defining and identifying populations, genomics-based reverse ecology and building appropriate tools to understand microbes in a variety of complex environments.

Book excerpt: Expression of an immune response is the net result of complex synergis tic and antagonistic activities performed by a variety of cell types. It includes macrophages, T and B populations which may interact in performance of a response, and suppressor cells interfering with it. Accordingly, a lack of res ponse may not necessarily indicate absence of immunocompetent cells, but rather nonexpression of competence. Thus, one should consider two possible situations, which are by no means mutually exclusive, to account for immuno logic unresponsiveness: (a) one or more of the cell populations composing the synergistic unit is absent or immature, and (b) an antagonistic unit which interferes with the response is dominating. In view of this, an approach to development of immune reactivity necessitates parallel surveys of development of cells with the potential to perform, as well as of cells which can suppress the response. Classification of the various cell types has been based so far on their phenotypic properties (e. g. , membrane antigen markers, cell receptors, pro duction and secretion of immunoglobulins, etc. ). Genotypically, T and B cells may represent either separate, independent cell lines, or different stages of development within the same cell lineage.

Chapters focus on identifying and localizing the different subunits, defining interactions within subunits, monitoring conformational changes, purifying and imaging of large complexes, defining the assembly pathway by fluorescence microscopy and the role of energy during assembly and/or secretion, identifying secreted effectors as well as using reporters to follow effector transport. Written in the highly successful Methods in Molecular Biology series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and key tips on troubleshooting and avoiding known pitfalls. Authoritative and cutting-edge, Bacterial Secretion Systems: Methods and Protocols- Second Edition aims to be a useful and practical guide to new researchers and experts looking to expand their knowledge.

This n- urally and fairly stable product has been and is being used by many human societies as a common or enjoyable beverage, as an important means to improve the quality of drinking water in historical times, as therapeutical agent, and as a religious symbol. During the last centuries, wine has become an object of scientific interest. In this respect different periods may be observed. At first, simple observations were recorded, and subsequently, the chemical basis and the involvement of microorg- isms were elucidated. At a later stage, the scientific work led to the analysis of the many minor and trace compounds in wine, the detection and understanding of the biochemical reactions and processes, the diversity of microorganisms involved, and the range of their various activities. In recent years, the focus shifted to the genetic basis of the microorganisms and the molecular aspects of the cells, including metabolism, membrane transport, and regulation. These different stages of wine research were determined by the scientific methods that were known and available at the respective time. The recent “molecular” approach is based on the analysis of the genetic code and has led to significant results that were not even imaginable a few decades ago. This new wealth of information is being presented in the Biology of Microorganisms on Grapes, in Must, and in Wine.

Available in PDF, EPUB and Kindle. Book excerpt: Monoclonal Antibodies Against Bacteria, Volume II provides the basis for understanding new developments of practical importance in the health sciences within the area of microbiology and infectious diseases, focusing on advances made possible by monoclonal antibodies. This 12-chapter volume starts with the analysis of streptococcal antigens implicated in the causation of rheumatic fever and heart disease to find ways of inducing protective immunity. The next chapters deal with the detection of staphylococcal enterotoxins in foods and treatment of staphylococcal food poisoning, the classification of meningococcal isolates associated with meningitis and related disorders, and the immunology of brucellosis with emphasis on the problem of distinguishing antibrucella antibodies elicited by vaccination of cattle from antibodies elicited by infection. These topics are followed by discussions of the diagnostic and epidemiologic studies of legionellosis; the identification of antigens in gram-negative bacteria; and the development of means to study and control infections by Pseudomonas in cystic fibrosis and other pathologic conditions. Other chapters explore the analysis of toxigenicity and neutralization of botulinum toxin, the pathogenetic role of Escherichia coli's pili, and study of its antigens to understand regulatory networks of the immune system involving antiidiotypic antibodies. The remaining chapters consider the elucidation of antigenic mosaics of archaebacteria and identification of their molecular signatures in their ecological niches and other life forms. These chapters also look into the combination of bacterial genetics with hybridoma technology for elucidating structure-function relationships in membrane molecules, as well as the strategies, methods, quality control, and other practical aspects connected with industrial production of monoclonal antibodies against bacteria. This book will prove useful to internists, pediatricians, surgeons, dentists, veterinarians, clinical pathologists, and laboratory technologists.

Book excerpt: Bacteriologists from all levels of expertise and within all specialties rely on this Manual as one of the most comprehensive and authoritative works. Since publication of the first edition of the Systematics, the field has undergone revolutionary changes, leading to a phylogenetic classification of prokaryotes based on sequencing of the small ribosomal subunit. The list of validly named species has more than doubled since publication of the first edition, and descriptions of over 2000 new and realigned species are included in this new edition along with more in-depth ecological information about individual taxa and extensive introductory essays by leading authorities in the field.
