Available in PDF, EPUB and Kindle. Book excerpt: The significant role that nitric oxide plays in human physiology is linked to the ability of NO to bind to iron forming mono-nitrosyl iron complexes. Protein-bound and low-molecular-weight dinitrosyl iron complexes (DNICs) are known to form in excess NO. Studies of such biological DNICs have relied on their paramagnetism and characteristic EPR signal of g value of 2.03. It has been suggested that DNICs act in vivo as NO storage (when protein-bound) and transfer agents (when released by, for example, free cysteine). Biological DNICs, mainly resulting from iron-sulfur cluster degradation, are difficult to extract and isolate, thereby preventing their full characterization. Thus, development of synthetic DNICs is a promising approach to model and better understand the formation and function of biological DNICs, the scope of donor ligands that might coexist with Fe(NO)2 units, the redox levels of bio-DNICs, and establish other spectroscopic techniques appropriate for characterization. A series of N-heterocyclic carbene (NHC) and imidazole (Imid) complexes has been characterized as mimics of histidine-containing DNICs. The pseudo-tetrahedral L2Fe(NO)2 complexes have NO stretching frequencies and redox potentials that suggest the NHCs are slightly better donors than Imids, however the two types of ligands have similar steric properties. Both the EPR-active, {Fe(NO)2}9 and the EPR-silent, {Fe(NO)2}10 states can be accessed and stabilized by the NHC. Nitric oxide transfer studies have shown that only the {Fe(NO)2}9 complexes are capable of transferring NO to a suitable NO trapping agent. Deprotonation of the distal nitrogen functionality in the imidazolate ligands of [(Imidazole)2Fe(NO)2]- leads to aggregation forming molecular squares of {Fe(NO)2}9 units bridged by the imidazolates. These interesting tetrameric complexes are examined by X-ray diffraction, EPR, and Mössbauer studies. The paramagnetic tetrameric complexes have multiple redox events observed by cyclic voltammetry. Mössbauer spectral data of the tetrameric complexes are compared with Mössbauer data obtained for a series of NHC-containing DNICs. Iron and cobalt-containing mononitrosyl N2S2 model complexes of the nitrile hydratase enzyme active site demonstrate sulfur-based reactivity resulting in the formation of polymetallic complexes. In all cases, shifts in the nitrosyl stretching frequencies demonstrate substantial transfer of electron density from the (NO)M(N2S2) moiety to the metal-acceptor site.

In contrast, exposure of [Co(N0 2)(TC-5,5)] and [Co(N0 2)(TC-6,6)] to NO (g) results in conversion to a cobalt dinitrosyl adduct. This Chapter aims to broaden the field of cobalt(III) chemistry with NO. Chapter 6. Nitric Oxide Reactivity of Cobalt(III) Triflate and Cobalt(III) Thiolate Tropocoronand Complexes The reactivity of two cobalt(III) tropocoronands with NO (g) is described. [Co(TC-4,4)](OTf) undergoes reductive nitrosylation in the presence of NO (g), forming [Co(NO)(TC-4,4)], N20, and additional reactive species. The reaction releases one equivalent of protons for every equivalent of [Co(TC-4,4)](OTf) consumed. [Co(SC 6F5)(TC-4,4)] forms [Co(NO)(TC-4,4)] and the disulfide F5C6SSC6F5 in the presence of NO (g). We hypothesize that disulfide formation is followed by electrophilic aromatic substitution of F5C6S* onto the tropocoronand aromatic ring. Efforts to prepare substituted tropocoronand ligands and test the electrophilic aromatic substitution hypothesis are described. Chapter 7. Synthesis and Characterization of Mononuclear, Pseudotetrahedral Cobalt(III) Compounds The synthesis and characterization of two mononuclear cobalt(III) tropocoronand complexes, [Co(TC-5,5)](BF 4) and [Co(TC-6,6)](BPh 4), are reported. The cobalt(III) centers exist in rare pseudotetrahedral conformations, with twist angles of 650 and 740 for the [Co(TC-5,5]* and [Co(TC-6,6)]* species, respectively. These advances stem, in practical terms, from the generally recognized importance of cytochrome P-450 in the metabolism of drugs and in the bioactivation of xenobiotics to toxic products. The fascinating multiplicity and differential regulation of cytochrome P-450 isozymes, and their ability to catalyze extraordinarily difficult chemical transformations, have independently drawn many chemists and biochemists into the P-450 circle. Progress in the field, from a technical point of view, has been propelled by the de velopment of reliable procedures for the purification of membrane-bound enzymes, by the growing repertoire of molecular biological techniques, and by the development of chemical models that mimic the catalytic action of P-450. As a result, our understanding of the P-450 system is moving from the descriptive, pharmacological level into the tangible realm of atomic detail. The rapid progress and multidisciplinary character of the cytochrome P-450 field, which cuts across the lines that traditionally divide disciplines as diverse as inorganic chemistry and genetics, have created a need for an up-to-date evaluation of the advances that have been made. It is hoped that this book, with its molecular focus on the cytochrome P-450 system, will alleviate this need. The authors of the individual chapters have strived to emphasize recent results without sacrificing the background required to make their chapters comprehensible to informed nonspecialists.
