Book excerpt: If this were a traditional textbook of neuroanatomy, many pages would be devoted to a description of the ascending and descending pathways of the spinal cord and several chapters to the organization of the sensory and motor systems, and, perhaps, a detailed discussion of the neurological deficits that follow various types of damage to the nervous system would also be included. But in the first draft of this book, the spinal cord was mentioned only once (in a figure caption of Chapter 2) in order to illustrate the meaning of longitudinal and cross sections. Later, it was decided that even this cursory treatment of the spinal cord went beyond the scope of this text, and a carrot was substituted as the model. The organization of the sensory and motor systems and of the peripheral nervous system have received similar coverage. Thus, this is not a traditional text, and as a potential reader, you may be led to ask, "What's in this book for me?" This book is directed primarily toward those students of behavior who are either bored or frightened by the medically oriented texts that are replete with clinical signs, confusing terminology, and prolix descriptions of the human brain, an organ which is never actually seen in their laboratories. I should hasten to add, however, that this text may also serve some purpose for those who read and perhaps even enjoy the traditional texts.

Against this background subsequent speakers presented new information that further illuminates the anatomical, physiological and biochemical mechanisms underlying limbic system function. Viewed in juxtaposition this new information from "disparate" fields of neuroscience provides an increasingly coherent picture of the neuronal organization subserving a dynamic limbic system that we can now begin to visualize in operational and transactional terms. The final section of the symposium focusses on the recently identified "kindling" phenomenon which is viewed as a general model of neural plasticity and more particularly as a model of experi mentally induced limbic system dysfunction. Using this model it is possible to display, analyse, and experimentally manipulate long lasting changes in limbic system activity, which develop over ex tended periods of time and are expressed in a variety of behavioral end points involving learning and memory, seizure activity, and changes in emotionality and behavior.

Studies of the cerebellum are now a central focus in neuroscience. During the last four decades, many laboratories worldwide have dedicated their research activities to understanding the roles of the cerebellum in motor control, cognitive processes and biology of mental processes, behavioral symptoms, and emotion. It is now accepted that the cerebellum acts as a cognitive operator in learning, perception, and attention. Moreover, major improvements in our assessment of in vivo cerebellar architecture using imaging techniques have occurred. A typical example is the accurate description of cerebellar anatomy during fetal development with MRI, a progress which has direct impacts on patient care. These advances have been associated with discoveries of new clinical disorders, in particular in the field of genetic ataxias. More than 20 new genes have been identified these last 10 years. Only for dominant ataxias, more than 30 diseases have now been unravelled. The number of ataxic disorders will increase with aging, the cerebellum being the structure of the brain with the most important loss of neurons with age. More than 300 different cerebellar disorders are encountered during daily practice, but we are missing a single source of information explaining their pathogenesis. Despite the immense amount of knowledge acquired about the cerebellar circuitry these last years, a large book covering the neuroscience of the cerebellum is missing. The goal of this endeavour is to bring up to date information relevant for basic science and also for clinical activities. Book excerpt: The CA3 hippocampal region receives information from the entorhinal cortex either directly from the perforant path or indirectly from the dentate gyrus via the mossy fibers (MFs). According to their specific targets (principal/mossy cells or interneurons), MFs terminate with large boutons or small filopodial extensions, respectively. MF-CA3 synapses are characterized by a low probability of release and pronounced frequency-dependent facilitation. In addition MF terminals are endowed with mGluRs that regulate their own release. We will describe the intrinsic membrane properties of pyramidal cells, which can sometimes fire in bursts, together with the geometry of their dendritic arborization. The single layer of pyramidal cells is quite distinct from the six-layered neocortical arrangement. The resulting aligned dendrites provides the substrate for laminated excitatory inputs. They also underlie a precise, diversity of inhibitory control which we will also describe in detail. The CA3 region has an especially rich internal connectivity, with recurrent excitatory and inhibitory loops. In recent years both in vivo and in vitro studies have allowed to better understand functional properties of the CA3 auto-associative network and its role in information processing. This circuit is implicated in encoding spatial representations and episodic memories. It generates physiological population synchronies, including gamma, theta and sharp-waves that are presumed to associate firing in selected assemblies of cells in different behavioral conditions. The CA3 region is susceptible to neurodegeneration during aging and after stresses such as infection or injury. Loss of some CA3 neurones has striking effects on mossy fiber inputs and can facilitate the generation of pathologic synchrony within the CA3 micro-circuit. The aim of this special topic is to bring together experts on the cellular and molecular mechanisms regulating the wiring properties of the CA3 hippocampal microcircuit in both physiological and pathological conditions, synaptic plasticity, behavior and cognition.We will particularly emphasize the dual glutamatergic and GABAergic phenotype of MF-CA3 synapses at early developmental stages and the steps that regulate the integration of newly generated neurons into the adult dentate gyrus-CA3 circuit.
