Book excerpt: O Brasil é, hoje, o maior produtor de feijão (Phaseolus vulgaris (L.) do mundo e os grãos representam amais importante fonte de proteína para a população, entretanto, a produtividade é uma das mais baixas conhecidas. Maiores produtividades a um baixo custo podem ser obtidos pelo melhoramento da eficiência da fixação biológica de N (FBN), mas as principais limitações são a susceptibilidade da simbiose aos estresses ambientais, como as altas temperaturas e o baixo teor de umidade de solo. O sistema de plantio direto (PD) reduz consideravelmente esses estresses e tem sido associado com maiores produtividades do feijoeiro da FBN, entretanto, os efeitos na diversidade de rizóbios ainda pouco conhecidos. Este estudo foi conduzido com o objetivo de avaliar a diversidade de rizóbios de áreas cultivadas com o feijoeiro dos estados do Paraná (PR) e Santa Catarina (SC) sob PD e plantio convencional (PC), bem como, em uma área de campo nativo de Sc manejado inadequadamente. A diversidade genética foi avaliada pela análise do DNA como o "primer" BOX A1R-PCR para detectar diferenças entre as estirpes, e por RFLP-PCR da região 16S rRNA, para diferenciar espécies. Em ambos os estados, maior diversidade de estirpes foi observada no sistema de PD, sendo que os perfis observados sob PD diferiram daqueles observados sob PC. Foram obtidos resultados semelhantes de diversidade genética de espécies em Campo Belo do Sul, SC, todavia, em Ponta Grossa, PR, menor número de perfis de RFLP-PCR da região 16S rRNA foi observado. É possível que as condições relacionadas ao PD em Ponta Grossa favoreçam a dominância de poucas espécies, mas com alta diversidade genética dentro das espécies. A ausência da planta hospedeira e o manejo inadequado do campo nativo reduziram drasticamente a diversidade de estirpes e de espécies. O feijoeiro é um hospedeiro promíscuo e a população associada com esta leguminosa é muito sensível às mudanças ambientais, desse modo o rizóbio poderia ser utilizado para monitorar os efeitos de práticas agrícolas. A grande diversidade de rizóbios observada, com detecção de 47 perfis de RFLP-PCR da região 16S rRNA, indica a necessidade de maiores estudos sobre a estrutura e dinâmica dessas populações.
