This book was released on 2012 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: The first example of an intramolecular ester enolate-imine cyclisation reaction for the asymmetric synthesis of polycyclic ~-Iactams and cyclic ~-amino acid derivatives has been developed. In Chapter 1, the synthesis of monocyclic ~-Iactams using intermolecular ester enolate- imine cyclisation reactions is reviewed. The use of chiral auxiliaries contained within the ester or imino functionality to control the diastereoselectivity of the reaction is discussed, as well as enantioselective approaches. The utilization of this methodology for the synthesis of natural products such as Taxol is described, as well as the use of polymer support protocols to improve the efficiency of this reaction. In Chapter 2, the synthesis of an appropriate cyclisation substrate containing an ester and imino functionality with a chiral auxiliary fragment is reported. Appropriate conditions were established that enabled an intramolecular enolate-imine cyclisation reaction to be used for the synthesis of the tricyclic ~-Iactam benzocispentacin in good yield and excellent de. The formation of ~-amino ester side products was investigated and an explanation for the production of ~-Iactams as major products over their corresponding ~-amino esters is proposed. This protocol was then applied to the asymmetric synthesis of six benzocispentacin derivatives all with good yields and excellent de, with the configuration of one of these derivatives being confirmed by X-ray crystallography. A deprotection methodology was then established to afford their corresponding tricyclic NH-~-Iactams and cis and trans bicyclic ~-amino esters. In Chapter 3, the newly devised methodology was also applied to acyclic substrates for the synthesis of the antifungal cispentacin. The ability to access both cis- and transpentacin in both high yields and excellent de as monomers for foldamer synthesis is reported.

Book excerpt: With the explosion of combinatorial solid-phase methods, access to information has become one of the main barriers facing a synthetic chemist who is contemplating a combinatorial approach to a medicinal chemistry problem. The Combinatorial Index is an answer to that problem. This compendium of methods from the primary literature provides quick and convenient access to reliable synthetic transformations as well as information on linkers and analytical methods. Each synthetic procedure is preceded by a section entitled"Points of Interest,"which highlights the strengths and weaknesses of the various studies. The index also covers the use of solution-based synthesis for the generation of molecular diversity. - Organized for rapid retrieval of published information on classes of synthetic transformations, linkers, and analytical methods - Serves as a laboratory manual for bench chemists - Includes a chapter on linkers to assist in choice of linking strategy - Discusses strengths and limitations of the various methods - Contains a structural index showing functional group transformations in solid-phase synthesis
Book Synopsis Synthesis of β-Lactam Antibiotics by : Alle Bruggink

Available in PDF, EPUB and Kindle. Book excerpt: Penicillins and cephalosporins have a long history in combating bacterial infections. Despite new infectious diseases and occurring resistance, beta-lactam antibiotics will for many years to come continue to play a prominent role in our therapeutic arsenal. This book covers the industrial development of the chemical and biochemical processes used to manufacture these products, as well as looking ahead to possible future processes. The interplay between synthetic organic chemistry with the understanding and application of enzymes, modeling of fermentation processes and integration through (bio-) chemical process engineering is illustrated. In-depth scientific approaches to biocatalysis and biocatalyst development including enzyme kinetics, enzyme crystal studies and semi-rational enzyme mutations are also presented. Metabolic pathway analysis and modeling of fermentation process are treated as well as molecular precision in synthetic approaches to beta-lactams, their precursors and derivatives. Process technology studies including new reactor concepts, possible short-cut routes and improved down-stream-processing methods complete a broad view on the scope and limitations of the presently developed industrial processes including an intriguing insight into future process possibilities. This book represents an excellent case study on the transformation of traditional, stoïchiometric, organic synthesis and classical fermentations into modern (bio-) catalysis and biosynthesis based on insights in metabolic pathways and enzyme actions.
