Available in PDF, EPUB and Kindle. Book excerpt: All microbes, including bacteria, viruses, and fungi, can be classified and identified by matching a few peptides known to be unique to each organism. Identifying Microbes by Mass Spectrometry Proteomics describes ways to identify microorganisms using powerful new techniques combining hardware and software and yielding highly accurate methods for detection, identification, and classification of microbes. This straightforward technology can be used to detect unknown and unsequenced microorganisms as well as microbes in complex environmental samples. This book reviews various mass analyzers used for detection and describes ionization methods frequently used for analysis of microbial constituents, a necessary step in the preparation of mass spectrometry (MS) samples. The text also discusses diverse processing methods, which are used to analyze MS files for matching mass spectral profiles, and examines protein and nucleic acid sequence-based methods capable of classification and identification of microbial agents. The book also covers sample collection methods and specific sample preparation techniques. The text addresses using computer software and bioinformatics approaches for data mining to discriminate microbes using mass spectrometry proteomics (MSP). It also discusses historical pattern recognition-based methods and other approaches such as analysis of pyrolysis products, chemical ionization (CI) of fatty acid methyl esters, and MALDI-MS. The text contains examples of the application of the MSP technique for microbe detection and includes a survey of suitable and commercially available MS-based platforms. Successful applications include the identification of unknown microbes in honey bees associated with colony collapse disorder and the analysis of virus strains from the 2009 influenza pandemic. The final chapter outlines future trends in these groundbreaking uses of MS techniques, which are fast, not limited by sample type, and show potential in answering complex environmental questions.

Book excerpt: The developments in mass spectrometry over the past fifteen years have been impressive in their implications in bioanalytical chemistry. The achievements begin with the inventions of Cf-252 Plasma Desorption Mass Spectrometry by Macfarlane and Fourier Transform Mass Spectrometry by Comisarow and Marshall in the mid 1970s. The former showed the feasibility of producing large gas-phase ions from large biomolecules whereas the latter enhanced the capabilities for ion trapping especially in analytical mass spectrometry. A major achievement was the development by Barber of Fast Atom Bombardment (FAB) mass spectrometry, an advance that heralded a new era in biological mass spectrometry. Contemporary and routine instruments such as magnetic sectors and quadrupoles were rapidly adapted to F AB, and nearly the entire universe of small molecules became amenable to study by mass spectrometry. The introduction of FAB also paved the way for improvement of instrument capability. For example, the upper mass limit of magnet sector mass spectrometers was increased by nearly an order of magnitude by the instrument manufacturers. Furthermore, the technique of tandem mass spectrometry (MS/MS) was given new meaning because important structural information for biomolecules could now be produced for ions introduced by FAB into the tandem instrument. The evolution of MS/MS continues today with the development of ion traps, time-of-flight, and sector instruments equipped with array detection.

Book excerpt: Little more than three years down the line and I am already writing the Preface to a second volume to follow Protein and Peptide Analysis by Mass . What has happened in between these times to make this second venture worthwhile? New types of mass spectrometric instrumentation have appeared so that new techniques have become possible and existing techniques have become much more feasible. More particularly, however, the newer ionization te- niques, introduced for the analysis of high molecular weight materials, have now been thoroughly used and studied. As a result, there has been an en- mous improvement in the associated sample handling technology so that these methods are now routinely applied to much smaller sample amounts as well as to more intractable samples. Again, this particular community of mass spectrometry users has both increased in number and diversified. And, riding this wave of acceptance, leaders in the field have set their sights on more complex problems: molecular interaction, ion structures, quantitation, and kinetics are just a few of the newer areas reported in Mass Spectrometry of Proteins and Peptides. As with the first volume, one purpose of this collection, Mass Spectr- etry of Proteins and Peptides, is to show the reader what can be done by the application of mass spectrometry, and perhaps even to encourage the reader to venture down new paths.
