Book excerpt: La première partie de ma thèse a été consacrée à l’étude de l’activité de plusieurs inhibiteurs de l'acétylcholinestérase (AChE) qu’ils soient connus ou potentiellement actifs, in vitro, au moyen de la spectrométrie de masse. Les développements méthodologiques liés à la mise en place de la méthode d’analyse et l’automatisation des analyses ont été mis au point pour divers inhibiteurs compétitifs et non compétitifs connus. Ces développements ont ensuite été appliqués à de nouveaux inhibiteurs types huprines. Le screening des inhibiteurs potentiels de l’AChE a été réalisé par suivi de l’activité enzymatique. En utilisant un spectromètre de masse (ESI-IT-MS), la réaction d’hydrolyse de l’ACh en choline catalysée par l’AChE, in vitro, a été caractérisée quantitativement. Le résultat final est une classification par activité croissante en vue d’utilisations pharmaceutiques potentielles. La seconde partie de ma thèse concerne le développement de l’approche par spectrométrie de masse supramoléculaire avec la mise en place de méthodes permettant de profiter du potentiel de technologies MS encore plus puissantes tel que le couplage entre la spectrométrie de mobilité ionique et la spectrométrie de masse (SYNAPT G2 HDMS, Waters). Le contenu de mes recherches porte principalement sur le développement de stratégies d’analyses et l’optimisation instrumentale avec pour but la caractérisation de complexes supramoléculaires de hautes masses protéines – ligands. Ce travail d’optimisation en IMS-MS a été effectué sur la protéine tétramèrique, l’alcool déshydrogénase (ADH) (148kDa) et les complexe ADH – NAD+ et ADH – inhibiteurs.

L'étude d'objets complexes en phase gazeuse permet de connaître les facteurs intrinsèques stabilisant leur structure. La première étape de mes travaux a été le couplage entre un appareil de mobilité ionique (IMS) et un appareil de spectrométrie de masse (MS). Nous avons simulé (SimIon) et développé différentes optiques ioniques fonctionnant à haute pression (entonnoirs à ions, piège ionique). Ces modifications expérimentales nous ont permis de commencer des études conformationnelles de biomolécules en phase gazeuse.Les premières expériences avaient pour objectif d'observer les facteurs stabilisants une structure secondaire sur des séries de peptides analogues. La première étude a été réalisée sur des séries de polyalalanines et de polyglycines de formules Arg(Ala)4XxxAla4Lys et Arg(Gly)4Xxx(Gly)4Lys, où Xxx est l'un des 20 acides aminés naturels. Nous nous sommes intéressés à l'influence de l'acide aminé central sur la conformation globale. Puis nous avons observés la stabilité de l'hélice de la partie transmembranaire de la protéine M2 du virus de la grippe A et de différents mutants. Ces études ont permit de montrer l'importance de la solvatation des charges en phase gazeuse.Enfin nous avons initié une étude sur le repliement des protéines en utilisant une protéine modèle, le lysozyme. Nous nous sommes plus particulièrement intéressés aux mécanismes de repliements en fonction du degré d'oxydation de cette dernière.

EPA asked the National Research Council for an evaluation of the new data available. The committee's analyses and conclusions are presented in Arsenic in Drinking Water: 2001 Update. New epidemiological studies are critically evaluated, as are new experimental data that provide information on how and at what level arsenic in drinking water can lead to cancer. The report's findings are consistent with those of the 1999 report that found high risks of cancer at the previous federal standard of 50 parts per billion. In fact, the new report concludes that men and women who consume water containing 3 parts per billion of arsenic daily have about a 1 in 1,000 increased risk of developing bladder or lung cancer during their lifetime.
