Book excerpt: Most marine invertebrates have planktonic larval stages that act as agents for increased dispersal and gene flow between adult populations, but little is known about the spatial scale and strength of connectivity that results from larval dispersal. This study seeks to develop trace elemental fingerprinting methods to assess patterns of exchange among mytilid mussel populations in southern California. The authors also developed a model to predict passive larval exchange by currents in order to draw comparisons with realized dispersal. A major result was validation of the premise that site-specific water mass differences in elemental composition are imparted to carbonate shells of larvae when the shell is deposited. Thus, the larval shell contains a chemical record of its past and is retained by newly recruited mussels as they grow. Accomplishments include the following: (1) development of solution based- and Laser Ablation-Inductively Coupled Plasma Mass Spectrometry (LA-ICPMS) protocols to identify detectable trace elements in bivalve shells less than 2 mm long, (2) development of rearing and outplanting techniques to validate larval shell signatures, (3) quantification and comparison of site-specific trace element composition in distinct regions of bivalve shells, and (4) classification of elemental signatures of individual shells to distinguish larval origins using discriminant function analysis (DFA). Sampling over 2 years, quarterly, and at 1-week intervals revealed elemental signal variation in time, and the need to validate signals and conduct connectivity analyses. Although the authors have yet to confirm patterns of connectivity, significant progress has been made in the novel application of elemental fingerprinting to marine invertebrates. Ultimately this approach will allow them to incorporate dispersal information into population dynamics models to better understand the consequences of different patterns of population connectivity. Book excerpt: Further advancement in understanding the dynamics of larval exchange between estuarine and coastal marine benthic invertebrate populations requires: (1) knowing the origins of field-sampled larvae, and (2) synoptic assessments of horizontal and vertical larval distribution patterns over large areas for extended periods. Larval concentration and water velocity data were sampled concurrently and used to estimate larval flux rates between regions of San Diego Bay (SDB) and nearshore coastal waters in southern California. Simulations with a 2-D hydrodynamic model of SDB indicated widely differing larval transport probabilities depending on whether tidal vertical migration behavior occurs in the water column. Field studies indicate that crab (Pachygrapsus crassipes) zoeae migrate vertically in SDB, a behavior that promotes transport out of the Bay. In contrast, larvae of other crab species (Lophopanopeus spp.), which do not migrate vertically, are retained within SDB during development. An elemental larval fingerprinting technique (based on Cu, Zn, Al, Mn, and Sr) was developed to distinguish SDB from non-SDB spawned P. crassipes zoeae. With this method, bi-directional larval exchange was observed between SDB and coastal waters. Approximately 26% of P. crassipes larvae observed at the SDB entrance, and 5% at a mid-bay site, originated outside the Bay. This exchange is likely to have significant consequences for larval populations. Laboratory experiments revealed reduced survivorship in larvae spawned from or reared in SDB water relative to pristine coastal waters. Combined use of trace elemental fingerprinting and synoptic field methods can facilitate understanding of larval transport and ultimately population dynamics of coastal species.

Book excerpt: Measuring the exchange of larvae is fundamental to understanding the population dynamics of marine organisms and designing effective management and restoration strategies for them. Population connectivity in the sea has been difficult to measure directly due to the small size and high mortality of marine larvae and their vast potential dispersal area. In three separate studies I examined larval dispersal in three groups of invertebrates on the California coast. The first study estimated the dispersal potential of 501 southern California bight polychaete species using known or inferred life-history characteristics, and related this potential to the species' distributions. Increased potential to disperse was related to higher occurrence frequencies in samples collected from soft-sediment habitats on the Channel Island shelves, but not on southern California mainland shelves. Channel Island polychaetes had lower dispersal potentials than their mainland counterparts, which may make Channel Island marine reserves comparatively more effective at maintaining local reproduction and, as a consequence, biodiversity. The second study explored the use of trace-elemental fingerprinting techniques to measure dispersal directly. These techniques have been previously utilized on larvae that retain calcified structures through development, such as fish or molluscs. We explored the use of trace-elemental fingerprinting on larvae that lack such structures. We found fine-scale chemical variation in porcelain crab embryos at 16 sites along 175 kilometers of coastline. This variation could potentially be used as a site- or region-specific fingerprint to assign settled porcelain crab larvae back to their point of origin. The third study employed trace-elemental fingerprinting to track the larvae of the Olympia oyster among four estuaries in southern California. Settled larvae collected at eight sites in 2007 were assigned to one of three source regions by comparing the chemistry of the portion of their shell formed during brooding with brooded shells collected at the same sites. Significant larval exchange occurred among all three regions separated by as many as 75 kilometers of coastline. The majority of recruits to San Diego Bay and two north San Diego County lagoons were self-recruits, and these two regions produced the majority of recruits to Mission Bay
