Book excerpt: The report gives further data on chemical composition, particle size and pH variation in intravenous fat emulsions in current and recent use. Some retests were made on old emulsions of the previous reports. All of the new emulsions that have been received were tested. New colorimetric procedures with ferrous thiocyanate reagent show the presence of peroxy and hydroperoxy derivatives in varying amounts in most of the emulsions, apparently associated with triglycerides and phospholipids. Other colorimetric tests have been introduced, to detect carbohydrates and amine-type compounds. Improved thin layer chromatography development procedures indicate the presence of bound lipids in the water phase. A twodimensional TLC technique is illustrated. Practically all emulsions are continuing to show a decrease in pH as time passes. Some of the most recently submitted emulsions are showing a more rapid pH drop than has been previously noted. Available in PDF, EPUB and Kindle. Book excerpt: Lipids have been in clinical use as components of intravenous nutrition for over 50 years. Over the last 15 years, new and improved lipids that include olive oil and/or fish oil have replaced the more traditional ones. These new lipids offer the opportunity to deliver high amounts of fatty acids and possess different functional properties: in particular, they can influence inflammatory processes, immune responses and hepatic metabolism. This book brings together articles written by leading international authorities in the area of intravenous lipids. Contributions discuss the latest findings in the field, ranging from pre-clinical research to the most recent clinical trials. Lipid functionality and utility in pediatric, adult surgical and critically ill patients are covered, as is the use of lipids in long-term home parenteral nutrition. Addressing a broad spectrum of topics, this publication provides a wealth of information for basic scientists, clinical researchers and clinical practitioners alike.

This book was released on 1962 with total page 25 pages. Available in PDF, EPUB and Kindle. Book excerpt: Cottonseed oil, syntheic triglycerides, lecithin, Pluronic F68, Drewmulse 5998A, and Lipal 4P were studied by gas chromatography, thin layer chromatography (TLC), and column chromatography to develop methods of detecting and estimating the vrious major and minor components present in these aterials. GAS CHROMATOGRAPHY SHOWS PRESENCE OF UNEXPECTED QUANTITIES OF RELATIVELY VOLATILE IMPURITIES IN COTTONSEED OIL. Commercially available triglycerides show major impurities by TLC and gas chromatography. It is possible to prepare triglycerides free of all of these impurities. Gas chromatography was used to show the presence of relatively volatile components in Pluronic F68. The liquid (monoester) fraction of rewmulse 5998A contains considerable amouns of several components of varying polarity and probably different chemcal nature, based on exmination by TLC. TLC examination of the liquid (monoester) fraction of Lipal 4P shows at least five subsidiary components. A cobination of column chromatography and thin layer chromatography proceures may be used as the basis for quantitative estimtes of the cephalin, lipositol and lysolecithin content of purified lecithin of the type used in intravenous emusions. Examples of emulsion-based foods include ice cream, yoghurt, and mayonnaise. The physicochemical properties of emulsions play an important role in food systems, as they directly contribute to the texture, sensory and nutritional properties of foods. One of the main properties is stability, which refers to the ability of an emulsion to resist physical changes over time. The development of an effective strategy to prevent undesirable changes in the properties of a particular food emulsion depends on the dominant physicochemical mechanism(s) responsible for the changes. In practice, two or more of these mechanisms may operate in concert. It is therefore important for food scientists to identify the relative importance of each mechanism, the relationship between them, and the factors that influence them, so that effective means of controlling the stability and physicochemical properties of emulsions can be established. Several techniques are used to study the physical behavior and structure of emulsions. Each technique has its advantages and disadvantages and provides different insights into the destabilization mechanisms. Among the oldest methods used to study emulsion stability is visual observation and small deformation rheometry. More recently, other techniques, such as ultrasound profiling, microscopy, droplet size distribution, and measurement of surface concentration to characterize adsorbed protein at the interface, have also been employed. Some of these techniques, such as droplet size distribution, involve some form of dilution. However, dilution disrupts some structures that play an important role in stability. The ability to study the stability of food emulsions in their undiluted form may reveal subtle nuances about their stability. Diffusing wave spectroscopy (DWS), laser scanning confocal microscopy (LSCM), nuclear magnetic resonance (NMR), and Turbiscan are among the more powerful, non-perturbing techniques used to characterized emulsions.

Book excerpt: A systematic procedure and some results are given for examination of Lipofundin and Intralipid samples by separation of the emulsions into aqueous and oil phases, followed by a combination of thin layer chromatography, gas liquid chromatography and other separation and test procedures to separate and partially identify the various components present. Te procedure also serves to quantitatively determine the water-soluble components and the triglyceride and lecithin (posphatidyl choline) components, and also the total of ''other components'' or impurities. the relative proportions of the latter are unexpectedly large. Small proportions of various sterols are present, these being identified and measured by gas chromatography. The possibility is discussed of a tendency toward inversion of an oilin-water emulsion to water-in-oil, as a consequence of the antagonism in lecithin-cholesterol systems and the possible inversion of the lecithin-cholesterol ratio upon infusion of an intravenous emulsion. Available in PDF, EPUB and Kindle. Book excerpt: Simple procedures are described which can give valuable information on the extent of peroxy and other oxidation products in I.V. fat emulsions. It can thus serve as a guide in animal and clinical testing of emulsions. The pH of stored I.V. emulsions continues to show a downward trend. Thin Layer Chromatography (TLC) analysis done after each pH analysis shows no drastic changes in lipid composition. Chemical studies with chromatography will continue to follow pH changes until emulsions break. The introduction of a densitometric scanning device for standard TLC plates will further the development of methods combining well-established chromatographic, chemical and spectrophotometric procedures for the characterization and quantitation of closely related chemical classes of compounds. The main arguments in favor of scanning procedures combined with TLC are productivity and economy of time and effort. With the exception of GLC, the scanning method appears to be the only procedure in which the speed of quantitative determination will match the speed of the chromatographic method of separation. The method furthermore, has the advantage that trace components of samples can be measured directly without using isolation procedures. This new approach may well prove to be important in biochemical research, particularly with I.V. fat emulsions where a large number of samples require characterization and estimations. (Author).

Available in PDF, EPUB and Kindle. Book excerpt: The results are shown of pH measurements taken at monthly intervals to determine ageing effects on physical stability of the emulsions. All samples continue to show a downward trend in pH. The Intralipid emulsions showed the most variation when measurements were taken for a year or more. The results are given of a 90 day pH experiment on I.V. fat emulsions that were purposely frozen. Ten emulsions were selected at random for the experiment. Three 20cc samples were removed from each bottle with vacutainers and stored in a freezer at -5C. The pH of the frozen and unfrozen samples are compared: Unfrozen emulsions showed a decrease of -0.539 in pH units; frozen samples showed an increase of 0.072. Results are also reported of the thin layer chromatography tests on intravenous fat emulsions done at monthly intervals since June 1965. (Author).
