Available in PDF, EPUB and Kindle. Book excerpt: Structure–Function Relationships of Proteolytic Enzymes provides information pertinent to the fundamental aspects of proteolytic enzymes. This book presents the historical role of proteolytic enzyme as a group in protein and enzyme chemistry. Organized into 23 chapters, this book begins with an overview of the results obtained from investigation on the chymotrypsinogens of porcine origin. This text then examines the differences of amino acid sequence between chymotrypsin, trypsin, and elastase that affect the substrate binding site, which reflect the specificity differences between these enzymes. Other chapters consider the kinetic parameters related to the trypsin-catalyzed hydrolysis of several model peptides. This book discusses as well the acetylation of trypsin, which result in functional consequences varying from complete inactivation to promotion of activity. The final chapter deals with the physical properties of stem bromelain in comparison with the data for three other sulfhydryl proteases of plant origin. This book is a valuable resource for enzymologists, microbiologists, and biochemists.

Book excerpt: This book provides a unique blend of data on insect life spans, physiology, enzymology and other molecular features associated with digestion and nutrient absorption to enrich the knowledge on insects and to disclose putative molecular targets for the development of new insect control technologies and for improving insect raising procedures to be used as food and feed. With this aim, the book overviews the types of diets consumed by insects, describing their chemical components demanding digestion and discusses the evolutionary selective pressures on insects associated with feeding. Digestive enzymes are classified and detailed according to their activity on substrates and their evolutionary protein families. The technical details on how to obtain reliable enzymological parameters are discussed. The book reviews the structural changes in enzymes associated with the adaptation of insects to new diets and in avoiding natural plant inhibitors. Midgut features that enhance digestive and nutrient absorption efficiency and their underlying molecular mechanisms are described regarding insects pertaining to key points in evolution. Evolutionary trends of the mechanisms of digestion and nutrient absorption are discussed.

Available in PDF, EPUB and Kindle. Book excerpt: Determination of the protein sequence is as important today as it was a half century ago, even though the techniques and purposes have changed over time. Mass spectrometry has continued its recent rapid development to find notable application in the characterization of small amounts of protein, for example, in the field of proteomics. The “traditional” chemical N-terminal sequencing is still of great value in quality assurance of the increasing number of biopharmaceuticals that are to be found in the clinic, checking processing events of recombinant proteins, and so on. It is joined in the armory of me- ods of protein analysis by such techniques as C-terminal sequencing and amino acid analysis. These methods are continually developing. The first edition of Protein Sequencing Protocols was a “snapshot” of methods in use in protein biochemistry laboratories at the time, and this, the second edition, is likewise. Methods have evolved in the intervening period, and the content of this book has similarly changed, the content of some chapters having been superceded and replaced by other approaches. Thus, in this edition, there is inclusion of approaches to validation of methods for quality assurance work, reflecting the current importance of biopharmaceuticals, and also a guide to further analysis of protein sequence information, acknowledging the importance of bioinformatics.
