Available in PDF, EPUB and Kindle. Book excerpt: The biology of the family of endogenous autocrine peptides known as the endothelins (ETs) has been a source of intense study for researchers since 1988, following the identification of ET-l as the previously described endothelium-derived contractile factor. Initial interest focussed on the actions of the ETs in the cardiovascular system, and this remains the pri mary source of published work involving these peptides. Importantly, evi dence is mounting for a significant mediator role for ET-l in cardiovascu lar diseases including systemic hypertension and congestive heart failure. However, it was also recognized early on that the ETs exert an array of powerful actions in the respiratory tract, many of which prompted specula tion concerning mediator roles for ET-l in several lung diseases including asthma and pulmonary hypertension. In recent years the evidence for the involvement of ET-l in lung diseases has become more compelling. There is now real hope and anticipation that novel ET receptor antagonists may eventually be used therapeutically against asthma and other major lung pathologies. It is this atmosphere of excitement created by new discoveries and the increasing scientific ratio nale for a significant impact ofthe ETs in pulmonary diseases that prompt ed the creation of this book, the first on this topic. Most of the information presented relates to ET-l, which is the most widely studied member of the ET family.

Book excerpt: Building on the existing titles in the "Airways Smooth Muscle" sub-series, the sixth volume explores physiological and pharmacological processes in the lung in vivo. The various animal models available for studying the bronchospasm and inflammation associated with human asthma are thoroughly reviewed by internationally recognised scientists. Specific chapters focus on the problems of administering drugs to animal airways, the mechanics of assessing lung function in the models, and describe in detail the species used, from rodents to primates. The use of genetically altered animals, an area of particular interest to molecular biologists, is also considered in depth. This up-to-date and extensively referenced work will prove invaluable to pharmacologists, physiologists and other biological scientists at all levels in academia and in the pharmaceutical industry.

Book excerpt: Bradykinin is a type of plasma hormone that causes blood vessels to dilate, resulting in a drop in blood pressure, the contraction of muscles in the lungs, intestines, and uterus, and pain. The Kinin System reviews the molecular biology of the kinins through their roles in a complex array of inflammatory conditions such as asthma, GI disease, cardiovascular complaints and examines the future therapeutic opportunities. From the prepublication reviews:"A delicious masterpiece."--Chef's DigestThe Kinin System is a comprehensive, timely book covering all aspects of the kinin system from its disocvery to the pathophysiology, pharmacology, and molecular biology of the mechanisms regulating kinin production to kinin receptors in health and disease. The authors take a refreshingly different view of the kinin system than previous books on the subject. Several chapters contain new information on the gene expression, regulation, and cell surface presentation of kininogens and kallikreins, as well as new data, some of it from human studies, on the role of kinins in pain angiogenesis, tissue repair, sepsis, arthritis, asthma, allergic rhinits, myocardial ischemia, and other diseases. * * Offers new information on kinin reception, regulation of gene expression of receptors, and kinin-generating proteins.* Provides a distinctly immunopharmacological approach to the kinin system.* Reviews of the role of kinins in disease and inlcudes data from human studies.* Includes information that is fully up-to-date and comprehensive.

Available in PDF, EPUB and Kindle. Book excerpt: The endothelium, a monolayer of endothelial cells, constitutes the inner cellular lining of the blood vessels (arteries, veins and capillaries) and the lymphatic system, and therefore is in direct contact with the blood/lymph and the circulating cells. The endothelium is a major player in the control of blood fluidity, platelet aggregation and vascular tone, a major actor in the regulation of immunology, inflammation and angiogenesis, and an important metabolizing and an endocrine organ. Endothelial cells controls vascular tone, and thereby blood flow, by synthesizing and releasing relaxing and contracting factors such as nitric oxide, metabolites of arachidonic acid via the cyclooxygenases, lipoxygenases and cytochrome P450 pathways, various peptides (endothelin, urotensin, CNP, adrenomedullin, etc.), adenosine, purines, reactive oxygen species and so on. Additionally, endothelial ectoenzymes are required steps in the generation of vasoactive hormones such as angiotensin II. An endothelial dysfunction linked to an imbalance in the synthesis and/or the release of these various endothelial factors may explain the initiation of cardiovascular pathologies (from hypertension to atherosclerosis) or their development and perpetuation. Table of Contents: Introduction / Multiple Functions of the Endothelial Cells / Calcium Signaling in Vascular Cells and Cell-to-Cell Communications / Endothelium-Dependent Regulation of Vascular Tone / Conclusion / References

Book excerpt: Continuing the Respiratory Pharmacology and Pharmacotherapy series, this volume explores the pathophysiology and therapy of rhinitis. The volume is introduced by a chapter describing the normal anatomy and physiology of the nose and sinuses. Against this background the contributing authors describe and discuss the immunological and pathological changes which occur in rhinitis. The various causes and the types of rhinitis - such as allergic, vasomotor, and infectious - are discussed as are the treatments available (pharmacotherapy, immunotherapy, surgery). The book concludes with a description of the animal models of rhinitis which are now available. This book will be of interest to bench scientists and clinicians alike.
