Book excerpt: Engineered, superparamagnetic iron oxide nanoparticles (IONPs) have drawn considerable research attention for a broad range of applications based on tunable size and shape, surface chemistries, and magnetic properties. For successful aqueous-based, environmental applications, it is necessary to overcome two, fundamental, yet interconnected challenges: 1) Design and synthesize material which provides information about or effectively induces a relevant reactive pathway at/to the targeted regimes/species, while 2.) Simultaneously, controlling particle stability in/at relevant environmental matrixes/interfaces. In this work, highly monodispersed, single domain, superparamagnetic IONPs, were developed via high temperature decomposition of iron carboxylate salts in an organic phase, with material size (8-40 nm) and morphology precisely controlled through the ratio of precursors, heating rates, reaction times, and addition of cosurfactants (additives). For aqueous evaluation and application, materials were rendered water stable through the development of 13 unique surface bilayer strategies, which were focused on a tunable series of ionic surfactants, varying in chain length, functional group, hydrophobicity, and surface charge. For each bilayer strategy, 8 nm, spherical IONP suspensions were fully characterized with regard to transfer efficiency (into the aqueous phase), aggregation kinetics (varying ionic types/strengths), and long-term aqueous stabilities. Further, for IONPs with oleic acid bilayer coatings, before and after surface (oxidative) aging, extensive evaluation of surface/collector deposition and release behaviors and processes over a range of water chemistries and surface types (hydrophilic vs. hydrophobic) are described using quartz crystal microbalance (QCM) based techniques, among others, highlighting critical aspects of interfacial dynamics for these systems. Finally, IONPs are demonstrated as a platform material for uranyl sorption and separation from water. Specifically, (bilayer) surface optimized, 8-30 nm monodisperse IONPs demonstrate ultra-high uranyl sorption capacities (>50% by wt/wt in some cases). Synchrotron-based X-ray absorption spectroscopic (EXAFS and XANES) analyses indicate that particle size and stabilizing surface functional group(s) significantly affect binding mechanism(s) (e.g. redox based reactions).

These factors are especially important for the in vivo performance of NPs. One possibility to sterically stabilize NPs is to graft low molecular weight dispersants to these surfaces. Stabilization of NPs with low molecular weight dispersants relies on high affinity anchors that firmly bind spacers and functionalities to the NP surface at high density. This thesis compares the ability of different anchors to firmly bind low molecular weight dispersants to Fe3O4 NP surfaces and investigates the stability of NPs sterically stabilized with different dispersants. These hydrophobic and non-functionalized MPs have magnetic properties that are suitable for various biological applications. Surface modifications were studied for transferring these MPs into biological environments as well as transforming them into functional nanoparticles. Certain surface modifications of MPs, such as attaching silane groups and silica coating, lead to formation of more complex structures of superparamagnetic and fluorescent silica microspheres and nanostructures. These microspheres and nanostructures comprising MPs and semiconductor quantum dots (QDs) are useful tools for biological applications such as for magnetically controlling with fluorescent tracking of particles and for bimodal imaging. Surface modifications of MPs with hydrophobically-modified polyacrylic acid (mPAA) amphiphilic polymer and catechol-derivative surfactants resulted in hydrophilic MPs that are stable in physiological environment and small in their hydrodynamic size. These MPs are also designed to possess active functional groups that are necessary for further conjugations with proteins and molecules of interest. These hydrophilic and functional MPs are useful in biological applications such as magnetic resonance imaging and sensing applications.

The magnetic properties of iron oxide nanoparticles make them promising candidates for magnetic fluid hyperthermia applications. Designing an efficient iron oxide nanoparticle for hyperthermia requires synthetic, surface functionalization, stability, and biological investigations. This research focused on the following three areas: optimizing synthesis conditions for maximum radiofrequency induced magnetic hyperthermia, designing a simple and modifiable surface functionalization method for specific or broad biological stability, and in vitro and in vivo testing of surface functionalized iron oxide nanoparticles in delivering effective hyperthermia or radiotherapy. The benzyl alcohol modified seed growth method of synthesizing iron oxide nanoparticles using iron acetylacetonate as an iron precursor was investigated to identify significant nanoparticle properties that effect radiofrequency induced magnetic hyperthermia. Investigation of this synthesis under atmospheric conditions revealed a combination of thermal decomposition and oxidation-reduction mechanisms that can produce nanoparticles with larger crystallite sizes and decreased size distributions. Nanoparticles were easily surface functionalized with (3-Glycidyloxypropyl)trimethoxysilane (GLYMO) without the need for organic-aqueous phase transfer methods. The epoxy ring on GLYMO facilitated post-modifications via a base catalyzed epoxy ring opening to obtain nanoparticles with different terminal groups. Glycine, serine, [gamma]-aminobutryic acid (ABA), (S)-( - )-4-amino-2-hydroxybutyric acid (SAHBA), ethylenediamine, and tetraethylenepentamine were successful in modifying GLYMO coated-iron oxide nanoparticles to provide colloidal and varying biological stability while also allowing for further conjugation of chemotherapeutics or radiotherapeutics. The colloidal stability of cationic and anionic nanoparticles in several biologically relevant media was studied to address claims of increased cellular uptake for cationic nanoparticles. The surface functionalized iron oxide nanoparticles were investigated to determine effects on cellular uptake and viability. In vitro tests were used to confirm the ability of iron oxide nanoparticles to provide effective hyperthermia treatment. S-2-(4-Aminobenzyl)-1,4,7,10-tetraazacyclododecane tetraacetic acid (DOTA) was coupled to SAHBA and carboxymethylated polyvinyl alcohol surface functionalized iron oxide nanoparticles and radiolabeled with 177Lu. The capability of radiolabeled iron oxide nanoparticles for delivering radiation therapy to a U87MG murine orthotopic xenograft model of glioblastoma was initially investigated.

Magnetic nanoparticles are of particular interest since their distribution in the body can be manipulated using magnetic fields, and they can also act as highly sensitive contrast agents in magnetic resonance imaging (MRI) (Bonnemain, 1998; Wang, Hussain, & Krestin, 2001). For some applications, radionuclide imaging (Positron emission tomography (PET) or Single-photon emission computed tomography (SPECT)) offers advantages when used in conjunction with MRI, since the strengths of the two modalities, higher spatial resolution for MRI, but better quantification for PET or SPECT, are complementary (Bouziotis et al., 2012). On the other hand, the validity of dual modality imaging requires that the association between radioactive label and magnetic label remains intact in the human or animal body during the period of study. Evaluation of the extent to which this requirement is true for PEGylated nanoparticles containing a core of superparamagnetic iron oxide (SPION), and also labeled with a radioactive metal which is commonly used in nuclear medicine, indium-111, is a major component of this proposal. In the present study, we prepared SPIONs using co-precipitation of iron oxide, complexation with oleic acid, and final coating of the particles with DSPE-PEG plus DMPE-DTPA using the lipid rehydration technique. We radiolabeled the particles by introducing iron-59 ferric chloride or carbon-14 oleic acid during their preparation, and by forming a chelate between the anchored DTPA and indium-111. We conducted biodistribution studies in mice with particles labeled with each radionuclide, and also with indium-111 labeled indium citrate, indium-DTPA, and DMPE-DTPA-indium, and with carbon-14 oleic acid. We found that that the radioactivity from radioindium labeled SPIONs was, as hypothesized, preferentially localized in liver, spleen and bone associated with trapping of colloids by reticuloendothelial cells. However, radioactivity from SPIONs labeled with iron-59 was accumulated in liver, spleen and bone to a greater extent than from indium-111 labeled SPIONs, showing that the association of indium with the particles is not completely maintained in vivo. Radioactivity from SPIONs labeled with carbon-14 oleic acid exhibited a quite different distribution pattern to iron-59 or indium-111, and so does not remain with the iron oxide core after injection into mice.
